Abstract

This research was conducted to determine the immunomodulatory effect of ethanol extract of neem leaves (Azadirachta indica) to the number of peritoneal macrophages in mice wich induced by BCG vaccine. Bacillus Calmette-Guerin (BCG) vaccine contained an attenuated Mycobacterium bovis. Mycobacterium bovis belongs to the Mycobacterium Tuberculosis Complex (MTC) group that has a similar phenotype characteristic with Mycobacterium tuberculosis and similar clinical manifestations of tuberculosis.The type of the research that used in this study is laboratory experimental research with Post Test Design Design Only Control Group Design. The research was conducted at the Faculty of Veterinary Medicine of Airlangga University in July 2017 using 25 male mice divided into five groups. The dosage of ethanol extract of the neem leaves given was 200 mg / Kg BW with variation for two days, four days, and six days are given.In the result of statistical data analysis using Kruskal-walis test, it is known that the significance value p = 0,03 (p <0,05), that means immunomodulatory of ethanol extract of neem leaves (Azadirachta indica) give an effect to peritoneal macrophage cell number in mice wich induced by BCG vaccine, so that neem leaves ethanol extract can be applied to tuberculosis patients.

Abstract

Blood Agar Plate (BAP) with sheep blood is the medium used for the identification and isolation of the bacterium Staphylococcus aureus. Because to produce is difficult, but is needed in health education institutions, BAP then used media as a human blood substitute. This study was to observe the pattern of growth of Staphylococcus aureus in media BAP human blood group O, AB and the blood of the sheep as a controle. With the aim to test the feasibility of using human blood group O and AB as essential compounds in the manufacture of a blood substitute sheep BAP media. Staphylococcus aureus growth patterns observed macroscopically to colony morphology, the number of colonies, and the diameter of the haemolysis zone. The suspension of bacteria used were pure cultures of Staphylococcus aureus (ATCC 25923) which is synchronized with Mc Farland 0.5 Standart solution, then do thinning 10-2. The suspension of bacteria that are already experiencing thinning planted in the media BAP human blood group O, AB and sheep blood each at 9 petri dish.From the observation of colony morphology, the number of colonies, and the diameter of the zone of haemolysis performed statistical tests using Kruskal Wallis test values obtained Asymp. Sig of 0.352 for the number of colonies and haemolysis zone diameters greater than α (0.05), which means there is no significant difference between the growth pattern of Staphylococcus aureus in media BAP using human blood group O, AB and sheep blood as a medium BAP controle.

Abstract

Toxoplasmosis is zoonotic diseases caused Toxoplasma gondii and can infected women. Risk factor Toxoplasmosis infection in women are habits of washing hand after contact wih animal. Thepurpose of the study was to research seroprevalence antibody IgG Toxoplasma gondii among women in Rangkah 6 Surabaya. This research is observational purposive sampling study.Total sample amount 45 sample from women and analyzed antibody IgG Toxoplasma gondii using ELISA technique. The result showed 57,5% (26/40) seropositive antibody IgG Toxoplasma gondii whereas42,5% (14/40) seronegative antibody IgG Toxoplasma gondii. Seropositive 6 of 26 women (23,1%) is pregnant women. Seronegative IgG T.gondii 5 from 14 (35,7%) is pregnant women.These research with Chi-Square test was level value p=0.976that meansno significantrelations between antibody IgG Toxoplasma gondii with habits of washing hand.The prevalence of Toxoplasma gondii antibody among women is high in Rangkah 6 Surabaya. these research will be neededAppropriated to screen women before and during pregnancy, should be dealt in order to minimize of this infection.

Abstract

At the beginning of the Corona Virus Disease 2019 (COVID-19) pandemic, rapid test examinations were widely used as a screening for Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) infection. The purpose of this examination was to detect SARS-CoV-2 IgM/IgG antibodies in the patient's body. One of these tests uses the immunochromatographic method. This study aims to determine the validity of immunochromatography. The study was conducted from August to September 2020. The sample used in this study was 100 patients. The research was conducted at Husada Utama Hospital Surabaya, Indonesia. According to the study's findings, the Zybio brand reagent kit has an accuracy of 85%, a sensitivity of 82%, a specificity of 88%, a positive predictive value of 87%, and a negative predictive value of 83%. In the group of patients who experienced clinical symptoms, < 7 had a sensitivity of 50%, specificity of 88%, positive predictive value of 60%, negative predictive value of 83%, and accuracy of 77.94% while the group of patients experiencing clinical symptoms > 7 days, had a sensitivity value of 100 %, specificity of 88%, positive predictive value of 84%, negative predictive value of 100%, and accuracy of 92.68%. Based on these results, the conclusion is that the Zybio brand reagent kit has a relatively high sensitivity, specificity, positive predictive value, negative value, and sample accuracy. In the group with clinical sensitivity < 7 days, the positive predictive value and accuracy are lower than the sample group with clinical symptoms > 7 days but have the same specificity.

Abstract

Gastritis is an inflammatory disease that can last for a long time. Helicobacter pylori infection is known as the main cause of peptic ulcer and the most common cause of gastritis. Helicobacter Pylori infection will cause mononuclear (MN) and polymorphonuclear (PMN) cell infiltration and will increase the production of proinflammatory cytokines in the gastric mucosa compared to non-Helicobacter Pylori gastritis. Helicobacter Pylori with histopathological examination diagnosis. This type of research is descriptive observational with purposive sampling data collection technique at the Anatomical Pathology Laboratory Installation, RSPAL dr. Ramelan Surabaya. The data used are the results of a biopsy of 60 samples that were determined as gastritis with histopathological examination of the identification of Helicobacter Pylori bacteria in gastric tissue classified by the level of bacterial density using The Updated Sydney System, as well as haemogram data for complete blood examination to further calculate the neutrophil lymphocyte ratio (NLR). The data were processed using descriptive statistical tests to determine the average neutrophil lymphocyte ratio (NLR). Based on the statistics of the r-Spearman test, the density of Helicobacter Pylori, (Pν = 0.000) below < 0.005 indicates that there is a strong relationship between the neutrophil lymphocyte ratio (NLR) and the density of Helicobacter Pylori bacteria.

Abstract

Background Smoking has become one of the leading causes of death worldwide. The negative impact of smoking usually occurs after a few years when a person starts actively smoking can harm the body's organs and become a major risk factor for diseases with inflammatory components. Systemic inflammatory reactions are characterized by an increase in inflammatory cytokines, blood cell counts and blood viscosity. Examination CRP and ESR are the two most frequently measured laboratory tests in assessing the response of a systemic inflammatory process. This study aims to determine and analyze the correlation between ESR values ​​and hs-CRP in active smokers at coffee shops, East Surabaya. Methods: The type of research used is correlation research with cross sectional. The sample used is 30 active smokers who meet the inclusion criteria. This research was carried out on February 21 – April 11, 2022. The examination of ESR values ​​and hs-CRP levels was carried out at Balai Besar Laboratorium Kesehatan (BBLK) Surabaya. Results: It was found that 30 respondents had been actively smoking >5 years with a percentage of 100%. as many as 8 respondents consumed 1-10 cigarettes/day with a percentage of 27%, 19 respondents consumed 10-20 cigarettes per/with a percentage of 63%, and 3 respondents consumed >20 cigarettes/day with a percentage of 10%. The average value of ESR is 8.37 mm/Hr and The The average value of the of hs-CRP 1.11 mg/L. Conclusion: Based on statistical tests, There is no correlation between the value of ESR and hs-CRP in active smokers in a coffee shop in the East Surabaya area (p value = 0.099) with a sufficient relationship and a positive direction of relationship (r = 0.307).

Abstract

Jombang Regency was the 10th most prevalent smoker in East Java in 2018 and experienced an increase in the prevalence of smokers by 0.1% from 2013. The high number of smokers raises a concern. Smoking may lead to an increase of hs-CRP and LDL cholesterol levels, where the increase in these two parameters is closely related to the formation of atherosclerotic plaques, as a cause of cardiovascular disease. Physical activity of each individual is something that needs to be known because it affects the levels of hs-CRP and LDL cholesterol. This study aims to analyze the correlation between hs-CRP and LDL cholesterol in active smokers with physical activity. This study used an observational analytic study with a cross-sectional design. This research was conducted in March-April 2022 with a total sample of 32 active smokers who work as pedicab drivers at Pondok Pesantren 'DU', Jombang Regency. The hs-CRP examination was carried out using the Latex Turbidimetric Assay method, while the LDL cholesterol examination was using the Homogeneous Enzymatic Colorimetric Assay method. Based on the statistical analysis of the Spearman correlation, the results obtained p>0.05 (p = 0.687, r = 0.074), which means that there is no correlation between levels of hs-CRP and LDL cholesterol in active smokers with physical activity.

Abstract

Diabetes mellitus type 2 is a metabolic disease that occurs due to abnormalities in insulin secretion, insulin action or both. One of the most common disorders in the body system is the red blood cell index consisting of MCV, MCH and MCHC. Spirulina platensis is a blue-green microalgae that contains phycocyanin so that it can increase hemoglobin levels, erythrocytes and blood profile in the bone marrow. The purpose of this study was to determine the difference before and after giving Spirulina platensis supplementation for 10 days on the erythrocyte index in patients with type 2 diabetes mellitus. This type of research was one group pretest-posttest. This research was conducted at the Waru Community Health Center, Sidoarjo Regency from December 2020 to May 2021. The results showed that the mean value of MCV decreased before: 80.19 fl and after: 80.1 fl. Meanwhile, the average value of MCH and MCHC increased, namely MCH before: 27.57 pg and after: 27.68 pg, then MCHC before: 34.33 g / dL and after: 34.51 g / dL. In the third statistical test, the significance value (p-value), namely the MCV (0.863), MCH (0.773) and MCHC (0.627) shows that (p> 0.05), it can be concluded that there is no significant difference before and after supplementation. Spirulina platensis on the erythrocyte index (MCV, MCH and MCHC) in people with type 2 diabetes mellitus.

Abstract

The consumption rate of Monosodium Glutamate (MSG) in Indonesia has increased every year. Uncontrolled use of MSG in Indonesia for a long period of time can cause toxic effects on the body. The free glutamate content produced by MSG can affect the work of the immune system, especially in the innate immune system and cause oxidative stress. To determine the effect of exposure to Monosodium Glutamate (MSG) on the viability of monocyte cells. This study is a laboratory experimental in vitro with a post test only control group design. A total of 10cc of peripheral venous blood was isolated using the ficoll gradient centrifugation method. The results of monocyte cell isolates were exposed to Monosodium Glutamate (MSG) according to groups. Group I: negative control, group II: monocyte cells + MSG 3%, group III: monocyte cells + MSG 6%, group IV: monocyte cells + MSG 9%. Subsequently incubated for 24 hours at 37 °C in 5% CO2. Then the viability test was carried out using trypan blue staining. Monocyte cell viability calculations were carried out under an inverted microscope with a magnification of 400x per 100 cells. The data obtained were analyzed statistically using the one-way Anova test followed by the LSD test. The average viability in each group was obtained as follows, monocyte cell viability in the control group was 63%, group II was 47%, group III was 45% and group IV was 35%. There is an effect of exposure to Monosodium Glutamate (MSG) on the viability of monocyte cells with the most significant effect being the 9% MSG concentration with an average viability of 35%.

Abstract

Coronavirus Disease 2019 (COVID-19) is an infectious disease caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS CoV-2). So far, the incidence of direct blood-borne transmission is not widely known. This is because the viremia of SARS-CoV-2 virus infection is very small, between 2-3 days after infection. Immunoglobulin M (IgM) is an antibody that is formed in the early days of a person being infected with the virus, which is around the third day and can persist in the blood for up to 3-4 months after infection. The purpose of this study was to perform serodiagnosis of SARS-CoV-2 antibodies in blood donors in Surabaya. The type of research used is an experimental laboratory with an exploratory research design. The research sample consisted of 100 donor blood samples from UTD PMI Surabaya. Rapid diagnostic test examination at the Immunology Laboratory, Medical Laboratory Technology Department, and the Immunoserology Laboratory of BBLK Surabaya, Indonesia for antibody titer examination using the ELISA method in May 2022. Results of antibody analysis against SARS-CoV- 2 in blood donors in Surabaya by 58% (Cut Off Index = 0.1569), so further research is needed on factors that can affect the formation of antibodies. Keywords: SARS-CoV-2 IgG; SARS-CoV-2 IgM; blood donor
