Abstract

The Effect of Anorganic- Iron Supplementation on The Blood Cell of Humpback Grouper Fish (Cromileptes altivelis ) Infected by Kbrio parahaemolyticus This experiment was conducted to find the immunity response of humpback grouper (Chromileptes altivelis) correlated to the anorganic-iron supplement in feed. The blood cell was analyzed as the infection response of F5brioparahaemolyticus. The fish weight 6.9 5 0.22 g/ind. was cultured in 60x40~50cm aquarium and stocked at density 10 ind. / aquarium. After the fish fed by inorganic-iron supplementation at 0,25,50 and 100 mg/ kg for 68 days, then infected by J! parahaemolyticus. The blood cell showed the 50 and 100 mg Felkg inorganic-iron supplement enhanced erythrocytes, hematocrite, hemoglobin, lymphocytes and neutrophil after the 24hand the 72ndh ours infection. Total leucocytes backed to normal after the 7Pd infection and the index of phagocytic was higher with the 100 mg Fe/kg of iron supplementation. 1 Key words: iron (Fe), humpback grouper, feed, bacteria and blood cell.

Abstract

This study was conducted to find out the health status of humpback grouper fish(('hrtmtileptes altiverts) that are fed on inorganic-iron supplement. Both, thetreatment and the control was tested in anoxia condition. The fish (average weight4.18±0.13 g/ind) was cultured in 60x40x50 cm3 aquarium and stocked by 10indJaquarium. The fish was reared for 10 weeks: and they were fed threetimes/day at satiation. Results shown that the fish fed by non-iron supplement wasmore susceptible than the normal condition; base on the index of leucocytes,nionocytes. phagocytic and MCHC distribution. However, 25 mg Fe/kgsupplement in the diet was increased concentration of Fe by 35% - 100% in liver,intestine, body tissue and enhance fish vitality at anoxia condition. The survivalrate of fish increased by 1.3 times compare to the non-iron supplementation.

Abstract

Bawang hutan Eleutherine bulbosa (Mill.) Urb.merupakan tanaman obat yang memiliki kandungan oligosakarida, berpotensi sebagai prebiotik yang mampu meningkatkan kesehatan usus udang vaname (Litopenaeus vannamei). Tujuan dari penelitian ini adalah menguji efek dari ekstrak bawang hutan dengan konsentrasi berbeda terhadap perubahan pH media pada stimulasi pertumbuhan probiotik secara in vitro. Konsentrasi ekstrak bawang hutan terdiri dari : 10, 5, 2.5, 1.25, 0.625, 0.313, 0.156 mg/mL. Bakteri probiotik yang digunakan adalah Pseudoalteromonas piscicida 1Ub dan Bacillus sp. NP5,sedangkan bakteri enterik menggunakan Vibrio parahaemolyticus. Waktu inkubasi selama 24 jam dengan pengamatan warna dan pH media dilakukan pada jam ke-0, 14, 18, dan 24. Hasil penelitian menunjukkan bahwa setelah inkubasi selama 24 jam terjadi perubahan warna dan pH media. Nilai pH pada perlakuan probiotik lebih rendah dibandingkan dengan bakteri enterik. Nilai pH pada P. piscicida 1Ub lebih rendah dibandingkan dengan Bacillus sp. NP5. Perubahan pH yang lebih rendah menandakan probiotik mampu memanfaatkan oligosakarida yang ada pada ekstrak bawang hutan.Kata kunci : Eleutherine bulbosa, oligosakarida, pH, prebiotik, probiotik.

Abstract

Transgenic has been known as one of the applicable methods to improve growth performance of cultured fish. This study was performed to evaluate the growth performance, survival, and body composition of the 3rd generation of growth hormone (GH) transgenic common carp (TG). Juveniles (BW: 1.53 ± 0.03 g) were reared for 60 days in 250-L glass aquarium with stocking density of 25 fishes/aquarium. Fishes were fed with commercial feed (protein content 36%), three times a day to satiation. Growth and survival were measured every 20 days. Our results showed that TG fish has 1.49 times higher in average weight growth (p < 0.05) compared with the non-transgenic common carp (NT). Higher total feed consumption, survival, body protein content, protein and lipid retention, hepatosomatic index, and lower feed conversion ratio were also shown on TG fish compared with NT fish (p < 0.05). However, body lipid content and blood glucose level of TG fish were lower (p < 0.05) compared with the NT fish. Total ammonium nitrogen level in rearing media of TG fish was 51.78% lower (p < 0.05) than that of the NT fish. In conclusion, culturing of GH-TG common carp showed potential to achieve high productivity, efficient, and environmental-friendly aquaculture.

Abstract

Bacterial disease attack occurs at the hatchery stage, which is considered to be the most serious threat, and often results in mass mortality of shrimp larvae by vibrosis which is that caused by a luminous bacterium identified as Vibrio harveyi. This research was carried out to obtain local isolates of probiotic bacteria that were able to inhibit the growth of V. harveyi and effectively apply it as a biocontrol of vibriosis in shrimp cultures. The research was carried out as follows: (1) In vitro and in vivo selection of probiotic bacteria candidates, (2) Study of the action mechanism and characterization of the selected pro biotic bacteria, (3) Study on application of the selected probiotic bacteria as a biocontrol agent in shrimp cultures. Results of in vitro and in vivo selection provided the best three isolates, which were 1Ub, SKT-b and Ua. The survival rate of shrimp larvae which were not only inoculated by V. harveyi but also with 1Ub, SKT-b and Ua probiotic bacteria were 88.33, 83.33, and 81.67% respectively; where as the positive control treatment (merely inoculated with V. harveyi) gave a 41.67% survival rate and the negative control (without bacterial addition) was 68.33%. Studies using a rifampicin resistant marker (RfR) demonstrated that the number of V. harveyi MR5339 RfR cells in treatments without probiotic addition were higher than the treatment with the probiotic bacteria, in dead larvae, living larvae, as well as in the culture media. Partial sequencing of the I6S-rRNA gene showed that the I Ub isolate was similar to Pseudoalteromonas piscicida, whereas the SKT -b and Ua isolates were similar to Vibrio alginolyticus. Selected probiotic bacteria could be applied directly to shrimp larva culture media, or orally through enrichment of both natural and artificial food. Abstract

This research was carried out to evaluate the effect of cinnamon leaf (Cinnamomun burmanni) as a suplement inthe diet due to growth performance and nutriet composition of patin (Pangasius hypothalamus). A triplicateexperiment was conducted using fish with an initial body weight of 7.27 ± 0.28 g. Fish were culture in aquaria 50 x40 x 35 cm at a density of 10 fish/aquaria. Fish were fed on the diet contained either 0, 0.5, 1; or 1.5% respectively.Fish fed on the diet at satiation for 30 days. The result shows that the suplementation of cinnamon leaf powder inthe diet 0 to 1.5% was insignificantly affected feed consumption, specific growth rate, and survival rate of fish.However, the addition of Cinnamomumn burmanni up to 1% was significantly increased the level of total feeddigestibility twice, the protein digestibility to 87.39%, and protein retention 1.5 higher than the control (withoutcinnamon leaf powder). On the other hand, the diet of 1% Cinnamomumn burmanni were the optimal result of thisresearch with the feed efficient, decrease 30% of fat and have more compact flesh texture.

Abstract

This research was conducted to evaluate the digestibility and growth performance of catfish Pangasius hypopthalmus fed on Cinnamomum burmannii leaf powder and extract. Ten catfish with an average body weight of 0.02 ± 11.24 g were stocked into the 40 × 50 × 35 cm aquarium. Fish were fed on the diet contained of 0.1% cinnamon extract, 1% cinnamon powder and without cinnamon leaf. Fish were fed on those diets at satiation with times a day. Fish were cultured for 60 days. The results showed the bacterial total number in the gut of fish fed diet contained 0.1% cinnamon leaf extract was 7.8 x 107 cfu/g, greater than other treatments. Protein digestibility in 0.1% cinnamon extract increase 4.12% and body protein 58.72%. The whole body of fish fed on cinnamon leaf extract contained higher protein and lower fat. There was no different result in the growth of catfish

Abstract

This study aimed to evaluate the effect of extract and cinnamon (Cinnamomum burmannii) leaf flourin the diet in increasing quality of meat of catfish (Pangsianodon hypopthalmus). Catfish with a weight of319.64 ± 35.99 g/nets reared in 9 nets with dimension 2x1x1,5 m3 at a density of 15 fish/nets for 60 daysof maintenance. The fish were fed with leaves of cinnamon at a dose that was: 0% cinnamon leaf, 0.1%cinnamon leaf extract, and 1% cinnamon leaf flour. The fish were fed 2 times a day with the feeding rate of3% of the average weight of the body. The results showed the treatment of leaf extract and flour, cinnamoncapable of decreasing levels of body fat, fat meat by 10,31-12,27%, 37,26-50,23%, respectively, compared tocontrols (p<0,05), however, cinnamon leaf extract treatment is more effective in improving the quality ofmeat catfish that looks meat texture compact, white flesh color and a slightly sweet taste.Keywords: cinnamon leaf, meat quality, Pangsianodon hypopthalmus

Abstract

Catfish (Pangasianodon hypophthalmus) is a freshwater fish that has been produced in the form of a filet. One of the problems in producing good catfish fillet is compactness and brightness of catfish farmed meat. This research aimed to get feed formulation as enhancer  meat quality of striped catfish with added Cinnamon leaves flour (Cinnamomum burmannii)  and used shrimp head meal. A Fish with a weight of  208.98±25.76 g reared in 12 floating nets cage (2x1x1.5 m3) with density of 15 fish/nets for 60 days. As treatment, fish were fed with feed contains 1% cinnamon leaves,  45% shrimp head meal, and combined of cinnamon leaves and shrimp head meal, and as control used feed were formulated without cinnamon leaves and shrimp head meal. Fish were fed 2 times a daily with feeding rate 3.5% of average body weight offish. The test parameters observed were physical, chemical and organoleptic test of catfish meat. The results showed feed with contains cinnamon leaves and shrimp head meal could decrease level of body fat 14.7% compared than control (p<0.05). Feed with used cinnamon leaves and shrimp head meal gave a texture offillet fish more compact,  elastic and color of fillet fish white.
