Abstract

Penelitian ini bertujuan untuk mempelajari morfologi kelenjar aksesori muncak jantan secara makroanatomi dan mikroanatomi. Seekor muncak jantan dewasa berumur 4-5 tahun dengan bobot badan 19 kg digunakan pada penelitian ini. Muncak terlebih dahulu di-exanguinasi untuk dikoleksi kelenjar aksesori kelaminnya. Untuk memperoleh gambaran mikroanatomi, sampel kelenjar aksesori diproses dengan teknik histologi dan diwarnai dengan pewarnaan hematoksilin-eosin (HE). Hasil pengamatan makroskopis menunjukkan bahwa kelenjar aksesori muncak jantan terdiri atas ampula, duktus deferens, kelenjar prostat, kelenjar vesikularis, dan kelenjar bulbouretralis. Karakteristik histologi kelenjar aksesori muncak adalah ditemukannya kelenjar prostat yang berbentuk pars diseminata dengan kelenjar-kelenjar sekretori tersebar di sekeliling lumen uretra pars pelvina dimana secara makroskopis kelenjar tersebut tidak dapat diamati. Tipe kelenjar sekresi pada ampula, kelenjar vesikularis, dan pars diseminata prostat adalah tubuloalveolar, sedangkan pada kelenjar bulbouretralis tipe tubular. Dapat disimpulkan bahwa morfologi kelenjar aksesori muncak jantan memperlihatkan kemiripan dengan kelenjar aksesori pada ruminansia kecil lainnya seperti kambing, domba, reeves muntjak, dan pampas deer.

Abstract

Penelitian ini bertujuan mengetahui sebaran karbohidrat pada usus biawak air dengan metode histokimia. Organ usus dari satu ekor biawak jantan dewasa dikoleksi melalui prosedur perfusi dan selanjutnya difiksasi dengan larutan paraformaldehid 4%. Usus biawak dibagi menjadi enam bagian, yaitu bagian I sampai VI dan selanjutnya diproses menjadi preparat histologi. Deteksi sebaran karbohidrat pada lapisan mukosa usus dilakukan dengan pewarnaan alcian blue (AB) pH 2,5 untuk karbohidrat asam dan periodic acid schiff (PAS) untuk karbohidrat netral. Hasil penelitian menunjukan bahwa sebaran karbohidrat asam ditemukan pada sel goblet pada keenam bagian usus dengan intensitas reaksi sedang (++) sampai kuat (+++), dan tidak ditemukan pada struktur usus lainnya. Sebaran karbohidrat netral ditemukan di seluruh permukaan jaringan usus dengan intensitas reaksi lemah (+) sedangkan intensitas reaksi sedang (++) sampai kuat (+++) ditemukan pada sel goblet. Jumlah sel goblet penghasil karbohidrat asam dan netral pada usus bagian I-IV lebih sedikit (+~++) dibandingkan usus bagian V-VI (+++). Berdasarkan hasil penelitian dapat disimpulkan bahwa sebaran karbohidrat asam dan netral ditemukan diseluruh bagian usus dengan intensitas pewarnaan yang bervariasi. Jumlah sel goblet yang terdeteksi menghasilkan kedua jenis karbohidrat tersebut lebih banyak ditemukan pada usus bagian kaudal.Kata kunci: jaringan usus, sel goblet, karbohidrat asam dan netral, Varanus salvator. (Histochemical Study of Intestinal Carbohydrates Distribution in the Water Monitor (Varanus salvator))The objective of this study was to elaborate the distribution of carbohydrate in intestine tissue of water monitor using histochemical method. Intestinal organ from an adult male water monitor was collected after perfused and subsequently fixed in paraformaldehyde 4%. Intestinal organ was divided to six regions e.g. I to VI and then processed to histological slides. The carbohydrate distribution on the mucosal surface of intestinal tissue was stained with alcian blue (AB) pH 2.5 for detect the acid carbohydrates and periodic acid Schiff (PAS) for the neutral carbohydrates. The results showed that the distribution of acid carbohydrates found in the goblet cells at the I to VI regions with vary intensity of staining reaction with good staining (++) to intense staining (+++), whereas other intestinal structures did not contain the acid carbohydrates. Furthermore, the distribution of neutral carbohydrates was found in the whole intestinal tissue with weak reaction (+), while good staining (++) to intense staining (+++) was appeared in the goblet cells. Additionally, the number of goblet cells containing acid and neutral carbohydrates at the I-IV region was fewer (+~++) than at V-VI region (+++). Conclusion of this study is the distribution of acid and neutral carbohydrates appeared in all regions of the intestinal tissue. The large number of goblet cells that secreting both of carbohydrate type was found at the caudal of intestinal tissue.Keywords: intestinal tissue, goblet cells, acid and neutral carbohydrates, Varanus salvator.

Abstract

The lesser mouse-deer (Tragulus javanicus) is one of the Indonesia biodiversity. This animal distributes only in South-East Asia and Hindia. The lesser mouse-deer is the smallest ungulate in the world and regarded as an ideal model to biomedic and ruminant research. As their population becoming extincted, conservation efSort is very important. One of the eflorts in supporting the lesser mouse-deer conservation is by improving our knowledge reproductive physiology of the female lesser mouse-deer. The aim of this study was to investigate the histological characteristic of ovarian follicles development of the female lesser mouse-deer. Experiment was done on the ovary (n=4). The ovary was fixed in Bouin solution andprocessed according to standard of histology. The slides were stained in HE. At luteal phase, the developing follicles can be classified into 10 stages. The number of developing follicles in the left ovary were higher then in the right.

Abstract

The present research aimed to study the characteristics of frozen-thawed semen in beef bulls ex-import in Ungaran, Indonesia. 5 heads Simmental and 5 heads Limousin of 5.5 years old were used in this research. The research was done in Ungaran Artificial Insemination Center, Central Java, Indonesia and Laboratory of Reproduction and Obstetry, Faculty of Veterinary Medicine, Gadjah Mada University, Yogyakarta, Indonesia. Frozen semen was obtained from this AI center. Results of the research indicated that the average individual motility of Simmental and Limousin is 39.00±5.48% and 36.00±2.24%. The average percentage of live spermatozoa is 54.00±10.75% for Simmental and 48.20±9.78% for Limousin while  the average proportion of abnormal spermatozoa is 9.60±3.36% and 6.80±4.15% for both Simmental and Limousin, respectively. On average, Simmental frozen-thawed semen have the higher mean proportion of sperm motility, live sperm, and abnormal sperm than that of Limousin frozen-thawed semen

Abstract

Muntjak (Muntiacus muntjak muntjak) belongs to Cervidae family which distributed in Java Island and Southern part of Sumatera. This cervid has been protected by Indonesian Government since 1999. In order to support breeding program of the species and to avoid them from extinction, itsreproductive biology such as spermatogenesis and the correlation to semen quality is important to be investigated. Therefore, the objective of this study was to examine spermatogenesis and semen quality of two adult male muntjaks during antler growth periods that consist of hard antler (H), casting (C), andvelvet antler (V). Testicular tissues and semen (ejaculates) were obtained by core needle biopsy and electroejaculation methods respectively. Testicular tissues were processed histologically and stained with periodic acid Schiff (PAS) to observe spermatogenesis whereas semen was evaluated to obtaine itsquality. The results showed that spermatogenic activities were detected in all antler periods which marked by PAS positive staining (magenta colour) of round and elongated spermatid acrosomes. In H period, spermatogenic activity was higher than those C and V periods. According to semen evaluation,motile spermatozoa were found with different concentration in all antler periods. The highest sperm concentration (x 106 spermatozoa/ml) in both of muntjaks was found in H period (506.25 ± 61.87), and slightly decreased in C (288.75 ± 37.12), and V periods (362.60 ± 17.68). These finding showed thatspermatogenesis to produce spermatozoa is taken place while muntjaks are in C and V periods with differ activities that provable with the existence of motile spermatozoa from ejaculates in both of male muntjaks. Therefore, muntjaks could provide reproductive function throughout the year of reproductiveaseasonality which is similar to the reeves and formosan muntjaks

Abstract

The objective of this study was to determine the carbohydrate content of salivary glands lingual,sublingual, and mandibular glands of water monitor (Varanus salvator). This studywas used the lingualglands, sublingual, and mandibularof an adult male water monitor. Salivary glands were fixed by soakingin a solution of 4% paraformaldehyde for four days. Then, the tissuewas processed into routine histologicalpreparations. Salivary gland tissue was stained with alcianblue staining (AB) pH 2.5 for detecting acidiccarbohydrates and was stained with periodic acid Schiff (PAS) for detecting neutral carbohydrates. Theresults showed that the lingual and sublingual glands showed a positive reaction to AB pH 2.5 stainingand PAS with the intensity varied from weak (+) to strong intensity (+++) scattered in the cytoplasm of thecell. In the cytoplasm of cells of the mandibular glands showed negative reaction (-) to the AB pH 2.5staining but reacted positively to PAS with the intensity varied from weak (+) to strong (+++). Based onthe result of the study it can be concluded that lingual and sublingual glands of Water monitor containacidic and neutral carbohydrates while mandibular glands only contain neutral carbohydrates.

Abstract

The data available on the female reproductive organ of mouse deer (Tragulus javanicus) is still very limited. A study was therefore conducted to investigate the distribution and the concentration of carbohydrate residues during the development of ovary follicles. An ovary at luteal phase was used in this study. Thin sections of the ovary were prepared occording to the standard methods and they were then histochemically stained with flourecnece-labelled lectins such as peanut agglutinin (PNA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), Winged bean agglutinin (WGA) and Ulex europaeus agglutinin (UEA). The result showed that changes in the distribution and the concentration of carbohydrate occured during the development of the follicle. During the preantral stage, the cytoplasm of oosit contained carbohydrate with the residues of glucosa dan mannosa. Zona pelusida contained carbohydrates with residues of glucosa, mannosa, galactosa dan N-asetylgalactosamine, whereas extracellular matrix contained carbohydrate with the residues of glucosa dan mannosa. In the antral follicle, the cyitoplasm of oocytes contained carbohydarte with the residues of galactosa dan N-asetylgalactosamine, whereas its zona pelusida, extracellular matrix and follicular fluid contained carbohydarte with the residues of fucosa, N-asetylglucosamin and cyalic acid. Diffrences in the types and the distribution pattern of carbohydrates were observed in this study, both in preantral and antral follicles.
