Abstract

Kabupaten Kepulauan Meranti merupakan kabupaten hasil pemekaran dari Kabupaten Bengkalis. Dasar hukum berdirinya Kabupaten Kepulauan Meranti adalah Undang-undang Nomor 12 Tahun 2009, tanggal 16 Januari 2009. Mengingat usia kabupaten ini yang relatif muda, arahan pengembangan komoditas pertanian belum tersedia secara lengkap. Banyak aspek lahan yang harus diperhatikan agar pemanfaatan atau penggunaan lahan untuk pengembangan komoditas wilayah lebih efektif dan berdaya guna. Tujuan penelitian ini adalah: (1) Mengetahui komoditas unggulan di Kabupaten Kepulauan Meranti, (2) Menganalisis kesesuaian lahan untuk pengembangan berbagai komoditas pertanian, (3) Mengkaji dan menganalisis ketersediaan lahan yang berpotensi pengembangan untuk berbagai komoditas pertanian, dan (4) Menyusun arahan pengembangan komoditas pertanian. Hasil penelitian menunjukkan luas lahan sesuai dan tersedia di Kabupaten Kepulauan Meranti untuk pengembangan komoditas unggulan sebesar 108,277 ha. Komoditas yang dapat dikembangkan tiap kecamatan adalah sebagai berikut: Kecamatan Merbau adalah sagu (tanaman perkebunan), ketela pohon (tanaman pangan), jambu biji (buah-buahan) dan cabe besar, cabe rawit dan ketimun (sayur-sayuran); Pulau Merbau adalah karet dan pinang (tanaman perkebunan); Rangsang adalah sawo dan nenas (buah-buahan). Rangsang Barat adalah kopi dan pinang (tanaman perkebunan), pisang, pepaya dan manggis (buah-buahan); Tebing Tinggi adalah ketela rambat dan jagung (tanaman pangan) dan cabe besar, cabe rawit, kacang panjang dan ketimum (sayur-sayuran); Tebing Tinggi Barat adalah sagu (tanaman perkebunan), ketela rambat (tanaman pangan), pepaya, sukun dan nenas (buah-buahan); dan Kecamatan Tebing Tinggi Timur komoditas yang dapat dikembangkan adalah sagu untuk tanaman perkebunan.

Abstract

The objective of this study was to determine the carbohydrate content of salivary glands lingual,sublingual, and mandibular glands of water monitor (Varanus salvator). This studywas used the lingualglands, sublingual, and mandibularof an adult male water monitor. Salivary glands were fixed by soakingin a solution of 4% paraformaldehyde for four days. Then, the tissuewas processed into routine histologicalpreparations. Salivary gland tissue was stained with alcianblue staining (AB) pH 2.5 for detecting acidiccarbohydrates and was stained with periodic acid Schiff (PAS) for detecting neutral carbohydrates. Theresults showed that the lingual and sublingual glands showed a positive reaction to AB pH 2.5 stainingand PAS with the intensity varied from weak (+) to strong intensity (+++) scattered in the cytoplasm of thecell. In the cytoplasm of cells of the mandibular glands showed negative reaction (-) to the AB pH 2.5staining but reacted positively to PAS with the intensity varied from weak (+) to strong (+++). Based onthe result of the study it can be concluded that lingual and sublingual glands of Water monitor containacidic and neutral carbohydrates while mandibular glands only contain neutral carbohydrates.

Abstract

ABSTRAKPenelitian ini bertujuan mengetahui pengetahuan dan sikap pemilik rumah makan terhadap rantai pengolahan daging ayam di Banda Aceh. Penelitian ini dilaksanakan pada bulan Januari 2017 di beberapa rumah makan di Banda Aceh. Penelitian dilakukan dalam bentuk survei lapangan dengan mewawancarai responden menggunakan kuesioner terstruktur. Responden dipilih secara proporsional terhadap pemilik rumah makan di Banda Aceh. Kriteria responden meliputi berumur ≥ 20 tahun, pemilik rumah makan yang menjual olahan daging ayam, rumah makan dengan kebutuhan daging ayamnya ≥ 20 ekor/hari dan pemilik rumah makan di lokasi penelitian. Data hasil penelitian dianalisis secara deskriptif. Hasil penelitian terhadap 45 responden menunjukkan bahwa pengetahuan pemilik rumah makan terhadap rantai pengolahan daging ayam tergolong baik, namun sikap pemilik rumah makan terhadap rantai pengolahan daging ayam tergolong kurang baik, karena 22,22% responden menggunakan alat potong (pisau) yang sama dengan sayuran, serta 48,89% responden tidak mencuci tangan sebelum mengolah daging ayam.Kata kunci : pengetahuan, sikap, pemilik rumah makan, daging ayam, rantai pengolahan.ABSTRACT This study was aims to determine the knowledge and attitude of restaurant owners on chicken meat processing chain in Banda Aceh. This study was conducted in January 2017 in several restaurants in Banda Aceh. This research was conducted in the form of a field survey by interviewing respondents using a structured questionnaire. Respondents were selected proportionally which is restaurant owners in Banda Aceh. The criteria of respondents were aged ≥ 20 years, restaurant owners who sell chicken, restaurants that needs ≥ 20 chicken / day and restaurant owners in the study site. Data were analyzed descriptively. The results of 45 respondent shows that the knowledge of restaurant owners on chicken meat processing chain is quite good, but the majority of owners attitude towards chicken meat processing chain relatively poor because 22,22% of respondents using the same knife with vegetables and 48,89% of respondents do not wash their hands before treating chicken meat.Keyword : knowledge, attitude, restaurant owners, chicken meat, processing chain.

Abstract

Penelitian ini bertujuan menghitung jumlah koloni bakteri selulolitik pada sekum ayam kampung. Sampel yang digunakan yaitu isi sekum ayam kampung yang diambil di Tempat Pemotongan Unggas (TPU) Lambaro Aceh Besar sebanyak 5 ekor ayam kampung. Sampel dilakukan pengenceran 10-2 sampai 10-5, kemudian diinokulasi ke dalam media BHM-CMC agar dengan metode tuang (pour plate). Selanjutnya, diinkubasi dengan suhu 37˚C selama 96 jam. Koloni bakteri selulolitik yang tumbuh kemudian dilakukan pengamatan morfologi koloni (warna, posisi, pinggiran, dan diameter koloni), diameter zona bening, penghitungan jumlah total bakteri, dan pewarnaan Gram. Hasil penelitian menunjukkan bahwa koloni bakteri berwarna putih, berada di dalam dan di permukaan agar, memiliki pinggiran rata, diameter koloni 2-3 mm dan diameter zona bening 18 mm dan 29 mm. Bakteri berbentuk basil bereaksi negatif terhadap pewarnaan Gram, sedangkan bakteri berbentuk kokus bereaksi positif dan negatif terhadap pewarnaan Gram. Jumlah total bakteri pada isi sekum ayam kampung yaitu 5.3 x 105 cfu/g.Kata kunci: Ayam kampung, bakteri selulolitik, sekum, Carboxy Methyl Cellulase.                The aim of this study was to count the total count of cellulolytic bacteria colony in caecum of native chicken. The sample used was caecum contents of native chicken which taken from the poultry slaughterplace at Lambaro Aceh Besar. Sample were diluted from 10-2 to 10-5, then inoculated to agar BHM-CMC media with pour plate method. Then, incubated with temperature 37oC about 96 hours. A colony of cellulolytic bacteria which were grown then were observed by using colony bacteria criteria (colour, position, edge, and diameter of the colony), diameter of the bacterial zone, counting a total of bacteria which was grown, and Gram stain. The results showed that there is white bacterial colony, located inside and surface agar, has flat periphery, diameters of colony 2-3 mm and diameters of bacterial zone 18 and 29 mm. Bacill-shaped bacteria react negative to Gram staining, whereas coccus-shaped bacteria react positive and negative to Gram staining. Number of total bacteria in the native chicken’s caecum was 5.3 x 105 cfu/g Keywords: Native chicken, cellulolytic bacteria, caecum, Carboxy Methyl Cellulose.

Abstract

The objective of this study was to determine the anatomical morphology and histology of the intestineof water monitor (Varanussalvator). Intestine organ from an adult male water monito was used in thisstudy. Perfusion was performed to flow the 4% paraformaldehyde fixative solution through the heart.Intestine was collected, then divided into six sections, followed with dipping them into 4% paraformaldehydesolution for four days, continued histological preparations and stained with haematoxylin eosin (HE). Theanatomical morphology observation was performed before the intestine was processed into histologicalpreparations by observing the outer shape of the intestine and measuring it’slength. Morphologicalobservation was made †after processing theintestinal tissue by observing each portion of the intestinalwall structure. The results showed that transverse fold were found on the outer and inner surfaces of theintestine section I-IV, while in the V-VI part of the intestine, the folds were not observed anymore. Theintestine wall consisted of four layers, such as mucosa, submucosa, tunica muscularis, and serosa. Thevilli on intestine section I-IV were relatively longer and higher, while the intestine vili in section V-VI wererelatively short. The entire mucosa in water monitor was coated with absorptive columnar epitheliumwith goblet cells. Based on the result study, it can be concluded that the water monitor has more efficientdigestive process which can be seen morphologically from the relatively short intestinal, has transversefolds on the surface of the small intestine, and does not have cecum. The intestinal wall consists of layersi.e : mucosa, submucosa, tunica muscularis, and serosa.

Abstract

ABSTRAK Penelitian ini bertujuan mengidentifikasi karakteristik morfologi dan morfometri tulang-tulang pembentuk skeleton appendiculare antara itik (Anas platyrhynchos) dan entok (Cairina moschata). Sampel yang digunakan dalam penelitian ini berupa itik jantan (n=3) berumur 1 tahun dengan bobot badan 2-3 kg dan entok jantan (n=3) berumur 1 tahun dengan bobot badan 3-4 kg. Itik dan entok disembelih lalu dilakukan pemisahan (preparir) bulu, kulit, otot-otot dan organ-organ visceral. Setelah proses preparir, preparat direndam dalam larutan deterjen selama enam hari. Tulang-tulang penyusun skeleton appendiculare itik dan entok dipisahkan dari tulang tubuh lainnya. Proses pengawetan tulang dilakukan dengan merendam tulang-tulang dalam larutan formalin 5 % selama tiga hari kemudian dikeringkan pada suhu ruang. Setelah kering dilakukan pengamatan secara morfologi dan morfometri. Data morfologi dianalisis secara deskriptif sedangkan data morfometri dianalisis dengan uji t (P0,05). Berdasarkan pengamatan tulang-tulang pembentuk skeleton appendiculare terbagi atas dua kelompok tulang, yaitu ossa membri thoracici dan ossa membri pelvinae. Secara morfologi tulang-tulang pembentuk skeleton appendiculare itik dan entok memiliki bentuk yang sama, akan tetapi beberapa tulang entok lebih besar dibandingkan tulang-tulang itik. Secara morfometri ukuran beberapa tulang entok berbeda nyata dengan tulang-tulang itik (P0,05). Kesimpulan dari penelitian ini adalah tulang-tulang pembentuk skeleton appendiculare itik dan entok memiliki bentuk yang sama namun beberapa tulang berbeda ukurannya.  Kata kunci:  Itik, entok, skeleton appendiculare, morfologi dan morfometri. ABSTRACT This study aims to identify the morphological characteristics and morphometry of skeleton appendiculare ducks (Anas platyrhynchos) and muscovy ducks (Cairina moschata). The sample used in this study was drakers (n = 3) aged 1 year with a body weight of 2-3 kg and male muscovy (n = 3) aged 1 year with a body weight of 3-4 kg. Each of duck was slaughtered and then separated (for preparir section) to removed feathers, muscles, and visceral organs. After the preparation process the sample was soaked in detergent solution for six days. The skeletal bones that form ducks and muscovy ducks skeleton appendiculare were separated from other bones and subsquently. Soaked in 5% formalin solution for three days then drying it at room temperature. After drying, morphology and morphometry of bones were observed. Morphological data were analyzed descriptively while morphometric data were analyzed by student t test. Based on observations, the bones forming the appendicular skeleton were divided into two groups of bones, namely the thoracic bones and the pelvinae membrane. Morphologically the bones forming the appendiculare skeleton of ducks and Muscovy ducks have the same shape, but some Muscovy duck bones were larger than the bones of ducks. Morphometrically, the sizes of some Muscovy duck bones were significantly different from those of ducks (P0.05). The conclusion of this study is that the bones forming the appendiculare skeleton of ducks and Muscovy ducks have the same shape but several bones differ in size.Keyword:  Ducks, muscovy duck, skeleton appendiculare, morphology, and morphometry.

Abstract

The objective of this study was to identified the development of the immunoreactive follicle stimulating hormone (ir-FSH) cells in pituitary glands of rats (Rattus norvegicus)in different age using immunohistochemical (IHC) methods. The pituitary organ that used in this study was collected from eight white female rats aged 2, 4, 6, and 12 months. Pituitary glands were processed histologically and stained with IHC methods to detect the ir-FSH cells. The result showed that the ir-FSH cells were observed in pars distalis (PD), pars tuberalis (PT), and pars intermedia (PI) of adenohypophysis with different number of cells. Furthermore, ir-FSH cells were found abundantly (+++) in the PT and a few (+) in PD of rats aged two months. In rats aged 4 months those cells were distributed in slight number (+) in PT and abundant (+++) in PD. In addition, the cells were also found in a great quantities in rats whose ages were 6 months, (+++) in both PT and PD, but these cells decreased (++) in rats aged 12 months in PD and absent (-) in PT. In conclusion, the devalopment and distribution of ir-FSH cells were found in pars tuberalis and pars distalis adenohypophysis in the pituitary glands of rats aged 2 up to 12 months with different number and distribution patterns.

Abstract

This study aimed to determine the bone growth of broiler chickens fed with additional probiotic Aspergillusniger (AKBIS Prob). The organ taken was the femur bone. A total of 24 broiler chickens reared with additional feed on 4 treatments and 3 repetitions with a combination of additional feed namely P0 (commercial feed), P1 (commercial feed and 2% AKBIS Prob), P2 (commercial feed and 4% AKBIS Prob) and P3 ( commercial feed and 6% AKBIS Prob). The sampling of the femur bone was carried out on chicken 21 and 35 days old and observed histologically after stained with hematoxylin-eosin (HE). The results showed from the examination of macroscopic growth that the femur growth increased more at the age of 35 days compared to the age of 21 days in the treatment group P1, P2 and P3 who were given probiotic Apergillusniger (AKBIS Prob) where the amount of Ca and P required for growth can be fulfilled. On microscopic examination, the measurement parameters were the number of active osteoblasts and osteoblast that looked more passive on the 35th day of treatment, compared to the age of 21 days. While the number of osteoclasts was more at age 21 than at age of 35 days in the treatment group who were given probiotic Aspergillusniger (AKBIS Prob)as additional food.

Abstract

The purpose of this study is to determine the effect of the red dragon fruit peels (Hylocereus polyrhizus) extract on the growth of Salmonella pullorum. This research was conducted at the Laboratory of Microbiology, Veterinary Faculty, University of Syiah Kuala, Banda Aceh. Red dragon fruit peels that used in this study is obtained from  ripe fruit and Salmonella pullorum cultures is  obtained from  Microbiology Laboratory of Veterinary Faculty Unsyiah. Salmonella pullorum culture added evenly on the surface of Mueller Hinton Agar, the number of the bacteria have been standarized using Mc Farland'1 method. Discs soaked in the red dragon fruit peels extracts at different concentrations (20 mg/ml, 40 mg/mL, and 60 mg/ml). Ampicillin is used as positive controls and DMSO (dimethyl sulfoxide) was placed on the surface of Mueller Hinton Agar. The parameters in this study is the inhibition zone diameter. Data were analyzed descriptively. The results showed that the red dragon fruit peel extract is able to inhibit the growth of Salmonella  pullorum and the average of  inhibition diameter zone at a concentration 60 mg / ml is 9.6 mm, 40 mg / ml is 9.4 mm and 20 mg / ml is 9.3 mm

Abstract

This study aimed to determine the effect of fermented kelor leaves (Moringa oleifera) flour as additive in animal feed to increase the weight of boiler carcass. The samples used were 28 DOC strain Cobb CP-707 aged 1 day. This study was conducted using one way pattern complete randomized design with 4 treatments and 7 replications. Control group (P0) was fed with commercial feed; P1 was given 5% fermented kelor leaves flour;  P2 was given 7,5% fermented kelor leaves flour; and P3 was given 10% concentration of fermented kelor leaves flour. Feed replacement was conducted on 08.00 am and 5.00 pm everyday, and water was given ad libitum. Chemical content of flour before and after fermented was analyzed at Research Centre of Biotechnology and Biological Resources, IPB. Body weight measurement was carried out at Veterinary Public Health Laboratorium. Data were analyzed using ANOVA with SPSS 16.0. Mean value (±SD) of carcass weight for P0, P1, P2, and P3 were 938,71±180,83, 1011,86±101,64, 1010,71±126,69, and 907,43±190,35, respectively. This study showed that adddition of fermented kelor leaves flour for 21 days did not affect (P0,05) the weight of broilers carcass.
