Abstract

The improper and uncontrolled uses of antibiotics against pathogenic bacteria have resulted in the occurrence of MultiDrugs Resistant (MDR) strains. There is now an urgency to find alternative antibiotics to combat the MDR strains.Sponge associated microorganisms are among the most interesting and promising marine natural product sources whichproduce polyketide and non ribosomal peptide products with various biological activities. In this study, marine bacteriawere isolated from sponge Aaptos sp. collected from North Java Sea, and were screened for antibacterial activityagainst MDR strains. Three out of 64 bacterial isolates were successfully screened and were found to be active againstMDR strains, in which two isolates (SPA1 and SPA5) were active against resistant strain Escherichia coli and one isolate(SPA21) against resistant strain Proteus sp., respectively. These active isolates were also capable of amplifying NRPS(Non Ribosomal Peptide Synthetases) gene fragments necessary for the biosynthesis of non ribosomal peptides. Themolecular identification based on partial 16S DNA nukleotide sequences indicates that the active isolates areHalomonas aquamarina, Alpha proteobacterium, and Pseudoalteromonas luteviolacea.

Abstract

The metabolites from microorganisms are a rapidly growing field, due to the suspicion that a number of metabolites obtained from reefs invertebrates are produced by associated microorganisms. Less than 2% of microbial flora has been successfully isolated from marine environment. Coral reefs are the most diverse marine ecosystems, however, little is known about the microbial diversity in these ecosystems. It is expected that still quite a few parts of unexplored culturable invertebrate-associated microorganisms exists in the reef environments. The present study aimed at estimating the biodiversity of secondary metabolite-producing microbes associated with reefs invertebrates such as coral, soft coral and sponge collected from geographically different areas.

Abstract

Human activities in the Karimunjawa National Park increase water pollution, including heavy metals such as lead (Pb) and copper (Cu). The existence of heavy metals in sea waters can not be degraded, but it can be absorbed and accumulated in the body of organisms, including coral, Pocillopora damicornis. Given the existence of coral P. damicornis as sessile organism which are able to absorb lead (Pb) and copper (Cu) from the water, therefore, it is necessary to conduct research on heavy metals content in the coral. The purpose of this study was to determine the content of heavy metals lead (Pb) and copper (Cu) on the coral P. damicornis taken from several locations around the Karimunjawa National Park. The results of the study showed that heavy metals content in the water was not detected. While the highest Pb and Cu were found in the coral collected from Burung Island that was equal to 0.00405 ppm and 0.01121 ppm, respectively. The lowest Pb substance were found in the coral collected from Kemojan canal i.e. 0.00143 ppm. Sample collected from Indonoor, Kemojan canal and Karimunjawa port did not contain Cu substance. Based on Pb and Cu data contained in the coral and water samples Minister of Environment Decree No.51st, 2004, it could be concluded that the conditions of Karimunjawa National Park was still in the stage of no contamination.

Abstract

Penyakit karang telah diidentifikasi sebagai salah satu faktor utama yang memperburuk kondisi terumbu karang global. Adanya penyakit karang menimbulkan kekhawatiran akan kelestarian dan keanekaragaman terumbu karang. Ulcerative White Spots (UWS) adalah salah satu penyakit karang yang menginfeksi karang Favites sp. Penelitian ini bertujuan untuk mendapatkan isolat bakteri karang yang aktif menghambat isolat bakteri karang terserang penyakit UWS serta mengkarakterisasi secara biokimia. Penelitian dilaksanakan pada bulan Oktober 2013 - Maret 2014. Sampel diambil di perairan Pulau Panjang, Jepara. Studi mikrobiologi di Laboratorium Terpadu UNDIP Semarang, uji biokimia di Laboratorium Manajemen Kesehatan Hewan Akuatik, BBPBAP Jepara. Survey lapangan dilakukan dengan menggunakan metode belt transect. Bakteri diisolasi dari karang sehat dan karang yang terserang penyakit UWS. Purifikasi bakteri menggunakan metode streak. Uji antibakteri menggunakan metode overlay dan difusi agar. Uji biokimia menggunakan metoda Cowan and Steels. Hasil penelitian menunjukkan terumbu karang di Perairan Pulau Panjang memiliki tingkat prevalensi penyakit yang tinggi yaitu sebesar 66,037%. Hasil uji aktivitas antibakteri diperoleh isolat FSB 2.5 dan GSB 2.2.1 yang mampu menghambat pertumbuhan isolat bakteri karang terserang penyakit UWS. Karakterisasi bakteri secara biokimia dari isolat FSB 2.5 dan GSB 2.2.1 menyatakan bahwa bakteri masuk dalam genus Pseudomonas sp

Abstract

Munculnya sifat resistensi dan infeksi patogenitas bakteri membuat para ilmuwan berupaya untuk menemukan obat baru. Salah satu upaya yang dilakukan adalah pemanfaatan organisme laut sebagai agen antibakteri alami. Beberapa penelitian melaporkan aktivitas biologis Sargassum sp. dalam bidang farmakologi. Tujuan penelitian ini adalah mengetahui aktivitas antibakteri ekstrak Sargassum cinereum terhadap bakteri E.coli dan S. epidermidis serta mengetahui golongan senyawa dan toksisitas ekstrak S.cinereum. Pengambilan sampel dilakukan secara purposive sampling methods. Pengeringan sampel dilakukan secara kering angin (air drying). Ekstraksi dilakukan secara bertingkat dengan menggunakan tiga pelarut yang berbeda kepolarannya. Uji aktivitas antibakteri dilakukan menggunakan metode difusi agar (Kirby-bauer) konsentrasi 100, 50, 25, 15, 5, 1, 0,5, dan 0,1 µg/disc dengan 3 kali pengulangan. Analisis fitokimia dilakukan secara deskriptif kualitatif berdasarkan perubahan warna serta karakteristik fisika kimia suatu golongan. Uji toksisitas dilakukan menggunakan larva A. salina umur 48 jam (instar III/IV). Nilai toksisitas akut (LC50) dilakukan dengan menggunakan persamaan regresi linier. Hasil penelitian menunjukkan bahwa ekstrak S. cinereum pelarut etil asetat memiliki aktivitas antibakteri terbaik dengan zona hambat 5,08 ± 0,56 (bakteri E. coli) dan 6,69 mm ± 0,14 (bakteri S. epidermidis) serta memiliki aktivitas bakteriosidal. Hasil analisis fitokimia menunjukkan ekstrak Sargassum cinereum mengandung senyawa alkaloid (pelarut etil asetat), steroid (ketiga pelarut), saponin dan tanin (pelarut metanol). Hasil uji toksisitas menunjukkan ekstrak S.cinereum pelarut etil asetat memiliki toksisitas yang sangat toksik dengan nilai LC50-24 jam sebesar 24,25 ppm (sangat toksik kategori kronik).

Abstract

Waste is one of the major problems in various countries in the world, one of which is the waste of marine products, for example clam shells. This waste can be processed into chitin and chitosan. Chitosan alone has a lot of benefits that have relatively high economic value, such as chitosan as a natural source antibacterial that can be effectively produced. Deacetylation degree of chitosan determined by using data from FTIR spectra and the value was 69.11349%. The results showed that there were soluble chitosan with 1% acetic acid. In the antibacterial test of chitosan on E. coli and S. aureus showed inhibition zone on agar medium. Chitosan provided large inhibition zones on 0.01 μg/disk and 0.02 μg/disk concentrations , however conditions factor of the media effected the growth of those bacteria. Bacteria E. coli bacterium was more sensitive to salinity than thah of S. aureus, while the S. aureus bacterium is more sensitive to pH than that of E. coli bacterium.

Abstract

Along with the increasing resistance of bacteria in the health world, it is necessary discover a new drug. Sources of new antibacterial bioactive compounds can be obtained from seaweeds. The purposes of the research were to determine the antibacterial activity of crude extract seaweed againts the skin disease bacteria, Pseudomonas aeruginosa, Staphylococcus epidermidis and Micrococcus luteus, and to identify classes of compounds contained in crude extract of seaweed. This research was done from January to Mei 2012. Seaweeds sampling (Caulerpa sp, Eucheuma sp, Gracilaria sp dan Sargassum sp) was conducted in territorial water of Jepara. Extraction process and analysis of bioactive compound of crude extracts was conducted at Laboratory, Marine Science Department, Diponegoro University, Semarang. Test antibacterial activity was conducted at Medicine Laboratory, Ungaran. The method used in this research was experimental laboratories. Extraction process was conducted with solid-liquid extraction method. Extract were tested for antibacterial activity by agar diffusion method according to Kirby-baurer. Based on the antibacterial activity’s assay, 12 extracts which have antibacterial activity against test bacteria P. aeruginosa, S. epidermidis and M. luteus were obtained the 12 seaweed extracts the highest consentration of inhibition zone was 200 µg/disk in ethyl acetate extract of the Sargassum sp was the most active extracts against bacteria P. aeruginosa and M. luteus, while the methanol extract of Sargassum sp was the most active extracts against the bacteria S. epidermidis. Steroids was the most dominant compounds on all of 12 seaweed extracts.

Abstract

ABSTRAK : Marine biofouling merupakan tumbuhnya organisme yang tidak diinginkan pada suatu permukaan yang terendam air laut. Penempelan ini dapat menyebabkan gangguan teknis dan kerusakan. Penelitian ini bertujuan untuk mencari senyawa aktif antifouling yang ramah lingkungan sebagai pengganti TBT. Materi yang digunakan adalah ekstrak rumput laut Sargassum echinocarpum dan S. cinereum dengan berbagai macam pelarut (n-heksan, etil asetat, dan metanol). Sampel diambil dari perairan Gunungkidul, Yogyakarta. Bakteri biofilm diisolasi dari substrat plat kayu mahoni yang direndam selama 2 minggu. Metode dalam penelitian ini adalah Uji aktivitas antifouling menggunakan metode disk-diffusion, Uji Fitokimia, Uji Biokimia, dan Uji Brine Shrimp Lethality Test. Hasil penelitian menunjukan aktivitas antimikrofouling terbaik ditunjukkan oleh ekstrak kasar S. cinereum dengan pelarut etil asetat dan S. echinocarpum dengan pelarut metanol. Berdasarkan analisa One-Way ANOVA dengan signifikansi <0,05menunjukkan ekstrak dengan pelarut metanol dan etil asetat tidak berbeda nyata, sedangkan ekstrak dengan pelarut n-heksan tidak membentuk zona hambat. Uji Biokimia menunjukkan jenis bakteri yang dapat dihambat adalah Bacillus sp., dan Nacordiasp. Hasil fitokimia menunjukkan ekstrak S. cinereum dengan pelarut etil asetat mengandung senyawa alkaloid, quinon, steroid, dan flavonoid. Sedangkan ekstrak S. echinocarpum dengan pelarut metanol mengandung senyawa quinon, triterpenoid, dan flavonoid. Uji Brine Shrimp Lethality Test menunjukkan bahwa ekstrak S. echinorcarpum dan S. cinereum dengan pelarut metanol dan n-heksan memiliki nilai LC50 568,4347 bpj; 663,613 bpj; 639,711 bpj; dan 855,3114 bpj dengan kategori toksik. Dapat disimpulkan dari penelitian ini bahwa ekstrak S. cinereum dan S. echinocarpum dapat digunakan sebagai sumber antifouling.  ABSTRACT : Marine biofouling is the growth of unwanted organism on the structure, which is submerged in seawater. The attachment of those organisms could lead to technical because it attaches and destroys the structures.The aim of this research to find an active antifouling compound that is environmentally friendly as a replacement for TBT. This research used a sample of S. echinocarpum and S. cinereum, which was extracted in various solvents (n-hexane, ethyl acetate, and methanol). The seaweed samples were taken from Gunungkidul, Yogyakarta. The biofilm bacteria was isolated from the mahogany plate substrate that has been submerged in the sea for two weeks. Antifouling activity test used the disk diffusion method, Phytochemicals test used the Harborne test methods, Biochemical test used the Cowan and Steel methods, and Brine Shrimp Lethality Test used the Meyer methods. The results showed that the antimicrofouling activity is most well demonstrated by the extract of S. cinereum with ethyl acetate solvent and S. echinocarpum with methanol solvent. Based on the analysis of One-Way ANOVA with significance <0.05 is known that the different types of solvent does not affects the results of inhibitory zone where methanol and ethyl acetate  are not significantly different, which the extract with n-hexane solvent does not form an inhibition zone.Biochemistry test shows that the type of bacteria that can be inhibited is Bacillus sp., and Nacordia sp.. The results showed that the phytochemical extracts of the S. cinereum crude extract with ethyl acetate solvent contains alkaloid, quinone, steroid and flavonoid compounds. While the crude extract of S. echinocarpum with methanol solvent contains quinone, triterpenoid and flavonoid compounds.Brine Shrimp Lethality Test showed that the crude extract of S. echinorcarpum and S. cinereum with methanol and n-hexane occupy the toxic category with LC50 values 568.4347 bpj; 663.613 bpj; 639.711 bpj; and 855.3114 bpj. It was concluded that the crude extract of S. cinereum and S. echinocarpum making it possible to be used as a source of antifouling.

Abstract

Limbah cangkang kerang simping dan kerang darah di Semarang merupakan jenis limbah hasil perikanan yang pengolahannya belum maksimal sehingga masih menjadi masalah lingkungan. Penelitian ini bertujuan untuk mengetahui kualitas kitosan dua bagian cangkang kerang simping dengan warna berbeda dan cangkang kerang darah yang dapat dilihat dari derajat deasetilasinya serta mengetahui aktivitas antioksidan dari kitosan ketiga jenis cangkang tersebut.Pembuatan kitosan dilakukan dengan proses demineralisasi, deproteinasi, dan deasetilasi. Uji derajat deasetilasi dilakukan dengan metode spektroskopi FTIR dan uji aktivitas antioksidan dilakukan dengan metode DPPH. Uji proksimat kitosan setiap cangkang dilakukan untuk memperoleh data pendukung. Penelitian ini dilaksanakan pada bulan April - Mei 2014. Sampel cangkang diambil dariTambak Lorok, Semarang. Hasil penelitian menunjukkan bahwa derajat deasetilasi kitosan cangkang simping merah sebesar 66,27%, kitosan cangkang simping putih sebesar 71,37%, sedangkan pada kitosan cangkang kerang darah sebesar 69,72%. Hasil uji DPPH menunjukkan persentase negatif pada masing-masing ketiga sampel kitosan cangkang yang ditandai dengan tidak adanya perubahan warna pada DPPH. Berdasarkan hasil penelitian tersebut dapat disimpulkan bahwa ketiga sampel kitosan cangkang tidak memiliki aktivitas antioksidan sebagai senyawa penangkal radikal bebas.
