Abstract

Ovarian cancer is one of cancer types that begins in the ovaries. Many kind of anticancer agents are developed for the treatment now. Camptothecin analogs are agent that inhibit the action of DNA topoisomerase I, a nuclear enzyme that relaxes the supercoiled DNA during the replication, recombination, transcription, and repair of DNA. FDA have been approved two camptothecin analogs for the treatment of ovarian and colorectal cancer, those are topotecan and irinotecan. Another analogs are in various stages of clinical development, including 9-aminocamptothecin, rubitecan, lurtotecan, and exatecan mesylate. The purpose of this study is to discover interaction pattern between topoisomerase I and camptothecin analogs through in silico docking. PyRx is used for DNA topoisomerase I and camptothecin analogs docking. The interactions between topoisomerase I and camptothecin analogs were visualized and analyzed using PyMol and LigPlus. Our results show that the binding pattern between another camptothecin analogs, including rubitecan, lurtotecan, and 9-aminocamptothecin, with DNA topoisomerase I is relative similar with irinotecan-DNA topoisomerase I complex, specifically in the binding of Arg364, Asn491, Lys493 and Lys532.6 Keywords: camptothecin analogs; in silico docking; interaction pattern; ovarian cancer; topoisomerase I

Abstract

In this study, I analyzed the transactional and interpersonal conversation texts found in grade VIII English textbook entitled “EOS English on Sky 2”. This textbook is still much used at SMP or MTs although it was published a couple years ago. I also analyzed the linguistic features of the transactional and interpersonal conversations in the English textbook. It is assumed by the writers of the English textbook that the transactional and interpersonal conversation texts found in the English textbook are compatible with the standard of content as mentioned in curriculum 2006, School-based Curriculum (KTSP or Kurikulum Tingkat Satuan Pendidikan). The standard of content in KTSP 2006 develops student’s linguistic competence in people’s experience, ideas, and feelings and understands various meanings. This study focuses on the issues of structural-functional approach which analyzes the speech function, structural approach which analyzes linguistic features,This is a qualitative study. In calculating the data and the final result of data percentage, quantification was used to support this study. The data are transactional and interpersonal conversation texts written in grade VIII English textbook. Units of analysis in this study are moves and clauses. The conversation texts are presented in 8 units. The moves were analyzed functionally and the clauses were analyzed structurally. The result shows that the speech functions of the transactional conversation texts are 54.5% matching the standard of content, the speech functions of the interpersonal conversation texts are 2.1% matching the standard of content, the linguistic feature applied in the transactional and interpersonal conversation texts uses the linguistic feature in functional literacy level, the speech functions of conversation texts introduced in EOS English on Sky 2 for MTs grade VIII are less compatible with the standard of content based on the compatibility levels.

Abstract

The need for tilapia fish too high to meet production targets. Several ways have been made to produce a male monosex fish, such as the use of hormone 17α-methyltestosterone, however, the more difficult and limited availability of these hormones, then needed another material that is easier to get and more efficient utilization. In addition, the necessary alternative methods of monosex more effective and efficient naturally to produce high quality tilapia fish. How to get more male fish to do with sex reversal method. Male monosex can be done through the forest bee honey supplementation in the feed which refers to the process of reproduction. Recombinant growth hormone (RGH) regulates body growth, reproduction, immune system and regulate the osmotic pressure on the fish Teleostei. The importance of other research about the use of honey in the feed masculinization by rGH in effort to obtain a superior seeds, rapid growth and generate monosex males. The purpose of this research is to know the effect of a dose of honey to feed containing rGH on growth and sex ratio on tilapia and honey and determine best dosage in feed containing rGH. Test fish used is larasati tilapia fish (Oreochromis niloticus) with an average weight of 0.4 g / head and an average length of 1.2 cm with a stocking density of 80 fishes / container. This research used experimental method to completely randomized design 4 treatments and 3 repetitions. The treatment in this study: treatment A (dose of honey 0 ml / kg of feed), B (dose of honey 25 ml / kg of feed), C (dose of honey 50 ml / kg of feed) and D (at a dose of honey 75 ml / kg of feed). Data observed specific growth rate (SGR), food convertion ratio (FCR), total feed consumption (TKP), survival rate (SR), the percentage of male and water quality. The results showed that the dose differences in feed containing honey RGH on growth and sex ratio in tilapia (O. niloticus) provides highly significant effect (P <0.01) on the SGR, FCR and percentage of male sex. Best dose treatment D is able to produce SGR, FCR respectively of 9.81%/day and 1.75gram presantase best male sex by 81.11%.
