Abstract

Basil leaf (O. basilicum L) could be developed as a herbal supplement  due to its phenolic compounds which have antioxidant activity. Extract of this plant would be more effective as a natural antioxidant if it is coated with nanochitosan. Nanochitosan was prepared from the parrot fish (scarus sp) scale and chitosan of crab’s shell by gelatin ionic method. The purpose of this study was to determine  the effects of the nanochitosan coating on phenolic content of basil leaves. The parameters tested were phenolic content of  the basil leaf extracts and nanochitosan coated basil leaf extracts. The results show that the  size’s average of nanochitosan  from fish scales were in 657,4 nm. Phenolic compound was not detected in both the fishscale and crab’s shell derived chitosan. In the other hand, phenolic compound of the basil leaf extract  showed the presence of phenol content of 34.38 mg/kg. In basil leaf extracts coated with nanochitosan from fish scales and nanochitosan of crab’s shell showed a phenolic content of 39.28 mg / kg and 57.44 mg/kg respectively. This suggests that synthesis of phenolic compound was induced when the extract of basil leaves was coated by nanochitosan.Keywords: basil leaves, chitosan, nanoparticles, phenolic compounds

Abstract

An increasing production of avocado in Indonesia  has resulted in abundant seeds as wastes. Avocado’s seeds contain potential antioxidant compound, such as phenolic compound, which could be exploited for human consumption. Utilization of seeds as functional food could be more efficient by coating with a certain substance. Chitosan is a bio-compatible and non-toxic compound, and could be used as coating agent in the form of nanochitosan. The purpose of this study was to evaluate the content of  phenolic-compound in extract of avocado’s seed with and without coated by nanochitosan. In this study, the extract of avocado’s seed was coated with  Parrot fish scale (Scarus sp) derived nanochitosan, compared with nanochitosan from crab’s shell. Nanochitosan was prepared from chitosan by gelation ionic method. The results show that avocado’s seed extract has 44.89 mg/Kg of phenolic content,  much lower than the content in the extract coated with nanochitosan of fish and crab shell, which were 84.08  and 113,67 mg/Kg, respectively.   Therefore, coating with nanochitosan was proven increasing the phenolic compound in the extract of avocado’s seed.  Keywords: avocado, nanochitosan, phenol

Abstract

Konsumsi ikan laut sangat diminati oleh masyarakat Indonesia, termasuk di Sulawesi Utara. Berlimpahnya restoran seafood yang menyediakan ikan laut sebagai menu utamanya dan penjualan ikan laut di pasar turut meningkatkan produk samping dari ikan seperti sisik yang kurang dimanfaatkan secara optimal. Untuk meningkatkan nilai guna dari sisik ikan laut sebagai suatu produk sampingan perikanan, maka perlu adanya riset untuk mengetahui komposisi kimia yang terkandung dalam sisik ikan laut sebagai upaya menyelidiki potensi sisik untuk dapat dijadikan sumber biomaterial yang dapat berguna bagi manusia. Penelitian ini menggunakan sisik ikan yang berasal dari perairan laut yaitu ikan kakatua, kakap merah, napoleon, salem, dan sahamia. Sejumlah 50 gram sisik dari setiap jenis ikan laut yang dikumpul dianalisis proksimat untuk mengetahui kadar-kadar kimia di dalamnya seperti air, abu, lemak, protein, dan karbohidrat. Kandungan sisik ikan laut yang sudah dikeringkan secara umum adalah air 11%, abu 39%, lemak 5%, protein 30%, dan karbohidrat 15%.

Abstract

Background: One of the important minerals for growth is calcium to build new bone. Moringa oleifera has been known as high nutritional content and bioactive compound tree that could act as antioxidant and improved several diseases like diabetes, arthritis, cardiovascular. Phytochemical analyses have shown that Moringa oleifera contain of high macro minerals like calcium, potasium, magnesium, and phosphor. Several previous studies have shown that mineral nanoparticle have higher bioavailability than natural compounds.. Objectives: The purpose of this study was to analyze the effect of moringa oleifera nanoparticle leaf on mineral serum and bone at growth stage of Sprague Dawley male rats. Methods: In this study, the pre and post controlled group design and complete random design was used. The subjects were 27 growing male Sprague dawley rats aged 2 month were devided into three group which received standard feed contain CaCO3 (control group), intervention feed contained 450nm and 750nm nano Moringa oleifera for 60 days. The data of the experiment was analyzed by paired T-test and Duncan test at 95% confidence level.Results: The result showed that there was no significance different for both of intervention group for mineral serum and fermur bone. While for the tibia bone, the result showed that the calcium, phosphor and magnesium were significantly increase (p<0.05). The intervention group received 450nm showed more effective improvement of magnesium level (p<0.05) than intervention group received 750nm.Conclusion: The intervention of moringa oleifera nanoparticle leaf had a positive effect in decreasing serum and bone mineral levels in male rats. There were significant increased at magnesium level measured in tibia bones after intervention, with higher increasing at group received 450nm nanoparticle.

Abstract

Utilization of fish Lemuru ( Sardinella lemuru ) into fish meal has a byproduct in the form of fish oil . It is known that the content of omega-3 fatty acids in fish oil lemuru highly enough . So that the existing oil content in fish processing waste of lemuru needs to be recovered by purification , so that fish oil obtained has better quality . In this study, fish oil is purified by the alcaline refining methode . The result of the process of purifying fish oils have a better quality than that of unrefined fish oil . But the content of saturated fatty acids to decrease. The quality of fish oil purified with alcaline methode and purification done by the plant were not significantly different

Abstract

Agel ropes have lower mechanical properties (breaking strength and elongation) than synthetic fiber ropes. Chitosan coating is expected to improve the mechanical properties of agel ropes. The aims of this study are to investigate the effect of immersion in chitosan on the physical and mechanical properties of agel ropes, and to examine the breaking strength and elongation of coated agel ropes after being soaked in both freshwater and seawater. This experimental study was conducted at the Laboratory of Fishing Gear Technology, Department of Fisheries Resources Utilization IPB University, and Laboratory of Forest Product Engineering, Department of Forest Products IPB University. The experiment involved 3 mm in diameter of two agel ropes incorporating control rope and agel rope soaked in 1% chitosan for 24 hours. The control rope was light brown, stiff, and rough, while the treated rope was darker, less stiff, and smoother. The breaking strength of control and treated rope was significantly different (P<0.05), which in respect constitute 15.06 kgf and 24.54 kgf. The elongation of both ropes were also significantly different (P<0.05), making 10,56% for control and 18,04% for treated rope. Breaking strength of the agel rope decreased by 50% after being immersed for 28 days. Keywords: agel rope, chitosan, breaking strength, elongation

Abstract

Highly organic substances contained within fishery industrial wastewater will pollute the environment when it flows into public water body without any previous treatment. Chitosan is polymersubstance extracted from shrimp shell which is able to wrap solid particles suspended in liquids and bring them together and agglomerate makes it suitable as a coagulant aid. The objective of this research was to study the effectiveness of chitosan in order to decrease pollutant parameter in the fishery industrial wastewater. The quality of chitosan used during this research confirmed to Protan Laboratory standard. Results showed that chitosan as a coagulant has good performance in pH range of 6.6-6.8 and at concentration of 175-225 ppm. Within these conditions, chitosan could decrease turbidity value 94.33%-95.17%, TSS (Total Suspended Solids) 95.10%-95.82%, BOD (Biochemical Oxygen Demand) 72.09%-79%, COD (Chemical Oxygen Demand) 76.72%-79.40%, TKN (Total Kjeldahl Nitrogen) 12.50%-27.82%, NH3 80.84%-82.56%, NO2-N 47.69%-54.74%, NO3-N 19.39%-40.14%. The best chitosan concentration to use was at 200 ppm. Abstract

Upaya penghambatan kemunduran mutu ikan dapat dilakukan dengan menggunakan bahan pengawet. Bahan pengawet yang sebaiknya digunakan adalah yang bersumber dari bahan alami, untuk meminimalkan pengaruh bahan kimia yang berbahaya apabila menggunakan bahan sintetik. Salah satu bahan pengawet alami dari hasil perairan yang aman adalah kitosan. Ikan patin segar dengan berat 500-600 gram dipreparasi menjadi bentuk fillet skin on, fillet kemudian direndam selama 3 menit dalam larutan kitosan dengan konsentrasi 0 %, 1,5 %, dan 3 %, dibiarkan terbuka tanpa kemasan pada suhu ruang selama 18 jam dengan selang waktu pengamatan setiap 6 jam. Pengamatan dilakukan terhadap nilai organoleptik fillet, pengukuran nilai pH, nilai TVB, dan nilai TPC fillet ikan patin. Rancangan percobaan yang digunakan adalah rancangan acak lengkap (RAL) pola faktorial. Faktor percobaannya adalah perlakuan larutan kitosan konsentrasi 0 %, 1,5 %, dan 3 %, dan lama penyimpanan 0 jam, 6 jam, 12 jam, dan 18 jam. Nilai pH tertinggi terdapat pada fillet ikan tanpa larutan kitosan penyimpanan jam ke-0, yaitu sebesar 6,94; nilai pH terendah pada fillet ikan patin dengan larutan kitosan 3 % penyimpanan jam ke-18, yaitu sebesar 5,41. Nilai TVB fillet ikan tertinggi adalah pada fillet tanpa kitosan penyimpanan jam ke-18, yaitu 28,84 mg N/100 g sampel, sedangkan nilai terendah pada fillet dengan larutan kitosan 1,5 % pada jam ke-0, yaitu 10,36 mg N/100 g sampel. Nilai TPC terendah adalah 1,27x10^4 koloni/g terdapat pada fillet ikan dengan perlakuan larutan kitosan 3 %penyimpanan jam ke-0, sedangkan nilai TPC tertinggi adalah 7,15x10^7 koloni/g pada fillet tanpa perlakuan kitosan penyimpanan jam ke-18. Penggunaan larutan kitosan 1,5 % memberikan hasil yang terbaik berdasarkan parameter penampakan daging, tekstur, bau, nilai pH dan nilai TVB fillet. Sedangkan penggunaan larutan kitosan 3% memberikan hasil terbaik untuk parameter lendir dan nilai TPC fillet. Uji regresi linear menunjukkan bahwa penggunaan larutan kitosan mampu mempertahankan kesegaran fillet ikan patin 2 jam  lebih lama dibandingkan dengan fillet ikan patin tanpa perlakuan larutan kitosan.Kata kunci: antibakteri, enzimatis, fillet, kitosan,

Abstract

Agar adalah bentuk koloid dari suatu polisakarida kompleks yang diekstrak dari beberapa kelompok alga merah (Rhodophyceae). Salah satu pemanfaatan dari agar adalah sebagai media untuk pertumbuhan mikroba. Agar bakto adalah salah satu medium kultur yang digunakan untuk membantu menggelifikasi berbagai jenis larutan medium atau larutan kaldu (broth). Penambahan kitosan sebagai absorben pada proses pemurnian agar diharapkan dapat menghasilkan agar yang murni (refine agar) yang dapat digunakan untuk agar bakto sebagai media pertumbuhan mikroba. Tujuan dari penelitian ini adalah memodifikasi berbagai konsentrasi kitosan dan waktu proses absorbsi dalam pembuatan agar bakto sebagai media pertumbuhan mikroba. Penelitian dibagi menjadi dua tahap, yang pertama adalah tahapan proses isolasi kitin dan deasetilasi kitin menjadi kitosan dan penelitian utama yang dilakukan yaitu pembuatan agar bakto dengan menambahkan berbagai konsentrasi kitosan (0,5%, 1%, 1,5%) sebagai absorben terhadap agar yang dihasilkan dari Gracilaria sp. dan agar batang. Perlakuan kedua yang diberikan yaitu waktu proses absorbsi setelah dilakukan penambahan kitosan. Proses absorbsi yang dipilih 0 menit, 15 menit, 30 menit, dan 45 menit. Kombinasi perlakuan paling optimum untuk agar bakto berbahan baku Gracilaria sp. adalah pada konsentrasi kitosan 1% dengan lama pemanasan lanjutan ekstraksi selama 45 menit. Kombinasi perlakuan paling optimum untuk agar bakto berbahan baku agar batang adalah pada konsentrasi kitosan 0,5% dengan tanpa pemanasan lanjutan.Kata kunci: agar bakto, Gracilaria sp. , kitosan

Abstract

Air merupakan sumber daya alam yang sangat penting dalam kehidupan manusia dan digunakan untuk berbagai kegiatan, termasuk kegiatan pertanian, perikanan, peternakan, industri, pertambangan, rekreasi, olah raga dan sebagainya. Masalah utama sumber daya air meliputi kuantitas air yang sudah tidak mampu memenuhi kebutuhan manusia yang terus meningkat dan kualitas air untuk keperluan domestik terus menurun khususnya untuk air minum. Tujuan dari penelitian ini adalah mengetahui kemampuan kitosan dalam menurunkan jumlah bakteri koliform dan konsentrasi besi di dalam air sumur dan untuk mengetahui kemampuan kitosan didalam proses pemurnian air sumur. Penelitian ini dilakukan dalam dua tahap, yaitu koagulasi dan filtrasi. Pada proses penjernihan air sumur, didapatkan hasil bahwa penurunan nilai TPC (Total Plate Count) dan konsentrasi besi yang signifikan bila dibandingkan dengan kontrol (tanpa kitosan) adalah larutan kitosan 1 ppm, dimana nilai TPC air sumur sebesar 5,6 x 103 cfu/ml dan nilai konsentrasi besi yaitu 0,08 mg/l. Pada proses filtrasi (tahap akhir), didapatkan hasil bahwa filter dengan kitosan 5 gram sudah dapat menghasilkan air dengan kualitas memenuhi syarat kesehatan, dilihat dari parameter pH, kekeruhan, kadar besi, dan bakteri koliform. Dengan nilai pH sebesar 6,7+0,01, nilai kekeruhan sebesar 4,5+0,71 NTU, kadar besi < 0,016 mg/l, dan nilai bakteri koliform 0 MPN/ml. Parameter TSS hasil pemurnian belum termasuk ke dalam air dengan kualitas memenuhi syarat kesehatan (Raini 2004). Berarti parameter TSS masih perlu diturunkan.Kata kunci: air murni, absorbsi, kitosan, koagulasi, filtrasi.
