Abstract

The objectives of this study were to identify polymorphism of transforming growth factor β2 (TGF-β2) gene associated with Salmonella pullorum resistance in Indonesian chickens. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assays were used to genotype Indonesian chickens. Analysis of polymorphism was conducted by using PopGen 3.2 software. The effect of genotype on S. pullorum resistance was analyzed using the SAS General Linear Model (GLM) procedure. Genotyping was performed on 278 chickens from 7 Indonesian chicken populations (Sentul, Merawang, Pelung, Kampung, Parent Cobb broiler, The F1 crossbreed of Kampung x Parent Cobb broiler (KB) and F2 KB x KB. The product of amplification was 284 bp. The TGF-β2| RsaI locus was polymorphic in all populations, producing two alleles (T and C) and three genotypes (TT, CT, and CC). The result from the analysis of the allele and genotype frequency showed that the T allele had a higher frequency than the C allele in all populations. The χ2 analysis showed that the 6 chicken populations were deviated from Hardy-Weinberg equilibrium, with the exception of the Parent Cobb broiler chicken. The association result showed that TT genotype was significantly associated with S. pullorum resistance in Sentul chicken. Although the leukocyte concentration, leukocyte differentiation and H/L ratio in Sentul chicken with three of TGF-β2 genotypes (TT, TC, and CC) were not statistically different. In conclusion, polymorphism in the TGF-β2 chicken gene can be used as a candidate marker to increase S. pullorum immune response.

Abstract

Honey is one of the herbal supplements for increasing body health. The nutrient content in honey products can be reduced due to its processing, packing, storage, and industrial process. This study aimed to analyze the characteristic of honey and some of its nutrient contents of 5 honey samples that disperse among the society. This study was conducted from March to September 2019 in the Ministry of Industry Centre for Agro-based Industry Laboratory, Bogor, East Java. Honey samples were obtained from some wholesale supermarkets around the Jakarta area. Total carbohydrate and energy testings were using IK 5.4.5 method. Water content, diastase enzyme activity, hydroxymethylfurfural (HMF) content, and acidity tests were using the Indonesian National Standard  (SNI) procedure 3545 2013. HPLC method was used to test sucrose, glucose, and fructose, while AOAC 999.11 (9.1.09.2005 method was used to test Cu, Zn, and Fe content. Total carotenoid and flavonoid content were determined using spectrophotometry method.  The result revealed the characteristic of honey samples in this study, including the range of pH (3.56-4.16), water (17-22.9%), ashes (0,07-0,59), acidity (7.76-38.3 ml N NaOH 1/kg) and HMF (122-812 mg/kg) content as well as diastase enzyme activity (1.11-17.2 DN). Fructose, glucose, sucrose and reducing sugar content showed were, respectively, 21.5-39.3 gr/100 gr, 25.2-31.4 gr/100 gr, 0.5-1.75 gr/100 gr, and 58.4-71.2%. Honey samples contained 309-334 kal/100 gr of energy, 76.8-81.9% of carbohydrate, 0.03-0.39%, of protein and 0.10-0.65% of fat. Moreover, their mineral contents observed were 0.03-0.66 mg/kg of Cu, 0.09-3.13 mg/kg of Zn, and 1.59-20.1 mg/kg of Fe. Finally, total flavonoid and carotenoid of honey samples were, respectively, 0.02-0.07 %(b/b) and 0.05-0.64 mg/kg.

Abstract

Cinnamon is a type of plant that contains bioactive components and useful as a functional food used in the prevention of diabetes mellitus. The aim of this study was to identify the content of bioactive components of aquadest extract of cinnamon (Cinnamomum burmanii B.) using GC-MS. The results of the GC-MS analysis showed that there were eight different compounds contained in the aquadest cinnamon extract, namely cinnamaldehyde, coumarin, ethalic acid, phthalazinone, acetyl monoglyceride, itaconic anhydride, 3,5-dihydroxy-2-methyl-5,6-, and bis (2-ethylhexyl) platelets. The bioactive component of cinnamon identified in the Aceh region is coumarin at 37.01%. The bioactive component of cinnamon identified in the Jambi region was cinnamaldehyde of 33.50%. The bioactive component of cinnamon identified in West Sumatra is cinnamaldehyde of 17.49%. This study shows that there are different bioactive components from Sumatra, Indonesia. Chromatographic analysis using GC-MS showed that cinnamon from Sumatra, Indonesia contains bioactive components that have antihyperglycemic effects.Keywords: aquades extract, cinnamon, GC-MS, phytochemical

Abstract

This study aimed to evaluate the effect of liquid propolis of Trigona spp. on the immunological parameters in mice infected with Sthapylococcus aureus. The effects of propolis were analyzed using the macrophage activity as determined by the phagocytic activity and by the production of nitric oxide (NO) in Sprague Dawley rat peritoneal macrophages and the production of antibodies. The results showed that the liquid propolis produces (a) an increase in the phagocytic index, (b) a significant increase in NO production, and (c) an increase over the production of IgG antibodies. This study indicates that the ethanolic extract of propolis of Trigona spp. is able to activate macrophages, promote the production of No and antibodies. The combination of these results suggests that this extract has an immunomodulatory effect and can boost the immune responsse.

Abstract

Penelitian ini dilakukan untuk mempelajari status kesehatan anak sapi baru lahir yang diberi kolostrum berasal dari induk yang telah divaksinasi dangan vaksin E. coli in-aktif. Sebelas ekor anak sapi baru lahir digunakan dalam penelitian ini dibagi menjadi dua kelompok yaitu kelompok kolostrum (diberi kolostrum sapi) dan non-kolostrum (diberi susu sapi). Kolostrum atau susu sapi diberikan sebanyak 10% berat badan secara langsung setelah dilahirkan dan selanjutnya tiap 12 jam selama 3 hari. Uji tantang dilakukan dengan pemberian E. Coli K99 hidup peroral saat anak sapi berumur 14 jam. Pemeriksaan status kesehatan termasuk suhu tubuh, frekuensi pulsus dan napas serta kualitas defikasi pada 0,12, 24, 48, 72 dan 168 jam setelah uji tantang. Hasil penelitian ini memperlihatkan bahwa diarrhea timbul pada 12 sampai 26 jam setelah uji tantang pada semua anak sapi kelompok non-kolostrun dan beberapa anak sapi kelompok kolostrum. Kelompok non-kolostrum memperlihatkan tanda klinis dengan diarrhe parah sebagai diarrhe profus yang berwarna pucat kekuningan. Bahkan 1 ekor mati pada 3 hari setelah uji tantang. Sebaliknya pada kelompok kolostrum hanya memperlihatkan diarrhe sedang pada 5 dari 8 ekor. Kesimpulan dalam penelitian ini adalah konsumsi kolostrum yang berasal dari induk yang telah divaksinasi dengan E. Coli memperlihatkan sifat protektif terhadap infeksi E. coli K99.Kata kunci: Escherichia coli, kolostrum, anak sapi baru lahir. (The Health Performance of Neonatal Calves Received Colostrum from Cows Vaccinated by Escherichia coli)This experiment was conducted to study the health status of neonatal calves received colostrum from cows vaccinated by in-active E. coli. Eleven healthy newborn calves were used in this experiment and divided into two groups, i.e colostrum (received bovine colostrum) and non-colostrum group (received bovine milk). Colostrum or milk were given to the calves at 10% of body weight directly after birth and followed every 12 h, for three days. Challenges were done orally to all newborn calves when they were 14 hours of ages, used live E. coli K-99. Examination of health status included body temperature, pulses and respiration rates, and defecations quality at 0, 12, 24, 48, 72 and 168 hours after challenges. Results of the experiment showed that diarrhea appeared at 12-26 hours after challenges in all calves of non-colostrum group and part of calves in colostrum group. The non-colostrum group showed a severe clinical signs of diarrhea as watery profuse diarrhea with pale yellowish color. One calf even death at three days after challenges. In contrast, the colostrum group showed only a mild diarrhea in 5 out of 8 calves. In conclusion, the consumption of colostrum originated from cows vaccinated by E. coli showed protective properties against E. coli K-99 infection.Keywords: Escherichia coli, colostrum, neonatal calves

Abstract

The risk assessment of antimicrobial resistance is very important to determine the risk of decreasing antimicrobial efficacy can be used as a basis for policymakers in allowing or prohibiting the use of an antimicrobial. This study aims to assess the risk of using colistin against E. coli resistance in the broiler flock. Risk assessment is carried out qualitatively using primary data, interviews, and secondary data. To obtain primary data various studies were carried out including monitoring the prevalence of colistin-resistant E. coli and mcr-1 also mcr-2 genes in broiler flocks, mcr-1 gene transfer from E. coli to Salmonella Enteritidis, mcr-1 gene sequencing, mutant selection windows of colistin against E. coli, and also multiresistant of E. coli colistin-resistant. Assessment of the risk of E. coli colistin-resistant in the broiler flocks through direct contact with live broiler flock environment with the resulting assessment is a medium risk with low uncertainty. Since colistin sulfate is very critically important for humans, the reduced use of colistin sulfate in animal production is necessary to reduce the risk of resistance. Reducing the use of colistin sulfate requires the collaboration of various sectors such as the government, veterinary drugs industries, farmers, and consumers.

Abstract

This study aimed to investigate the phenotypic and genotypic of antibiotics resistance profile in Escherichia coli. The 30 samples come from cloacal swab of broilers in Cianjur, Indonesia. Isolation and identification of E. coli was performed by culture in McConkey agar, eosin methylene blue agar, Gram staining and five essential biochemical tests (IMViC). In this study, 10 isolates (33.3%) were confirmed E. coli positive. Phenotypic profile was performed by screening all isolates with 8 antibiotics of 6 antibiotic groups. The screening was carried by Kirby-Bauer disk diffusion method based on the standard of CLSI. For genotypic profile, each resistant isolate was detected antibiotic resistance-encoding gene. The result showed all isolates (100%) resistant against tetracyclin, oxytetracycline and erythromycin. Nine isolates (90%) detected nalidixic acid and enrofloxacin-resistant. The ciprofloxacin and gentamicin-resistant isolates were 70% and 40%, respectively. There was no resistant isolate for chloramphenicol. Multi drug-resistant was detected on 90% isolates. Only gyrA (100%) and tetA (80%) genes were detected. This study showed high rate of occurrence of antibiotic resistance in E. coli. Not all resistant isolates were detected in the antibiotic resistance-encoding gene in this study. Future research to detect resistance genes should use more varied target genes.

Abstract

Determination of surface charge of group B streptococci by ion exchange chromatography on DEAE-Sephacel revealed that bovine and human group B streptococcal isolates with protein surrface antigens alone, or bacteria with protein antigen in combination with polysacharide antigens, adhered strongly to the gel matrix. In contrary, cultures with polysacchaide antigens alone showed no comparable adherence properties. Removal of neuraminic acid from bacterial surface enhanced, but pronase treatment reduced the adherence values. The importance of type specific capsular sialylation for group B streptococcal surface charge could be confirmed with group B streptococci of serotype III and their transposon mutagenized asialocapsular mutants. In contrary to the encapsulated parent strains the asialo capsular mutants adhered strongly to the gel matrix. Comparable differences were observed with unencapsulated group B streptococcal variant strains and its isogenic encapsulated parent strains. The capsule material seemed to mask the surface proteins responsible for the adherence to the gel matrix. The determination of surface charge of group B streptococci by ion exchange chromatography might help to understand the importance of capsular sialylation for individual isolates of this bacterial species.
