Abstract

The evaluation of cytotoxic effect on ethanolic extract from the fruit rinds of asam kandis (Garcinia cowa Roxb.) on T47D human breast cancer cells in vitro had been evaluated. The cytotoxic effect was carried out by using MTT assay. Extraction of dried fruit rinds of G. cowa Roxb. was macerated with ethanol 70%. The extract was prepared into 4 concentration groups, 100, 10, 1, and 0,1 μg/ml. Results showed that the IC50 of G. cowa Roxb. ethanolic extract was 19,33 μg/ml ± 1,15 on breast cancer T47D cells. The percentage of cells viability and concentration were analyzed analyzed by the two-way analysis of variance (ANOVA) followed by Duncan test. Result showed that the ethanolic extract of G. cowa Roxb gave a significant effect on T47D breast cancer cells viability (P<0.05).

Abstract

The cytotoxic effect of ethanolic extract of Garcinia cowa Roxb. roots had been investigated on female mice using Micronucleus Assay method. Sixty mice were grouped into five: negative control, positive control, dosage I (extract 30 mg/kgBW), dosage II (extract 100 mg/kgBW), and dosage III (300 mg/kgBW) and then each group were grouped into three groups based on the duration of treatment. The group of mice were induced by cyclophosphamide 50 mg/kgBW intraperitoneally. After 30 hours, the treatment groups were administered the extract orally during 3, 7, and 15 days. After that, mice were sacrificed and the femur bone marrow was taken. Some of cytotoxic parameters were observed i.e micronuclei cells and hematocrit level. The percentage of micronuclei (MN) cells was calculated from the quantity of micronuclei cells of the slides of femur bone marrow by using microscope. Results confirmed that ethanolic extract of Garcinia cowa Roxb. roots decreased percentage of micronuclei cells not significantly (P > 0.05). The most lowering of micronuclei cells was showed by the extract in the dosage of 100 mg/KgBW for 15 days.

Abstract

A cytotoxic compound was isolated from n-hexane the stem bark of Garcinia griffithii T. Anders. (Guttiferae) as colourless needles from which melted at 242-243oC. Based on spectroscopies data of this compound was identified as isoxanthocymol. Cytotoxic assay using Microculture Tetrazolium (MTT) assay showed that isoxanthocymol was active against breast cancer cell line MCF-7, with IC50 value 35,46 μg/ml.   ABSTRAK Telah diisolasi senyawa sitotoksik dari kulit batang Garcinia griffithii T. Anders. (Guttiferae). Senyawa diisolasi dari fraksi n-heksana berupa amorf tidak berwarna dengan jarak leleh 242-2430C. Dari data spektroskopi massa, 1H-RMI, 13C-RMI, diketahui bahwa senyawa tersebut adalah isoxanthocymol. Pengujian aktifitas sitotoksik isoxanthocymol dilakukan dengan metode Microculture Tetrazolium (MTT) terhadap sel kanker payudara MCF-7, dan memperlihatkan aktifitas dengan nilai IC50 sebesar 35,46 μg/ml.

Abstract

In this study a High Performance Liquid Chromatography (HPLC) has been used for determination of rubraxanton on bark extracts Garcinia spp (Garcinia mangostana, Garcinia Cowa, Garcinia griffitii, Garcinia dioica and Garcinia forbesii). HPLC system consisted of C-18 reversed phase column with a length of 250 mm, diameter 4.6 mm, 20 mL injection volume, mobile phase methanol: water (gradient system with polarity) and flow rate of 1ml/min. Rubraxanton levels obtained in this study; 9.161% for G.mangostana, 6.942% for G.cowa, 6.762% for G.dioica, 0.499% for G.forbesii and 0.229% for G.griffitii. The method has been validated for specificity, linearity, accuracy, precision, limits of detection (LOD) and limits of quantitation (LOQ). The linearity of the method can be seen from the regression coefficient r=0.9996 with a linearity range from 1.72 to 55 ug/ml. Recovery of rubraxanton in the extract of G.mangostana was between 99.61 to 101.08%. Intra and inter day precision showed relatively small level of standard deviation (lower than 2%). Limit Of Detection (LOD) and Limit Of Quantitation (LOQ) are 0.55 ug/ml and 1.82 ug/ml respectively.Keywords : rubraxanton, bark extracts, Garcinia spp.

Abstract

The cytotoxic effect of ethanolic extract of the leaf of Garcinia cowa Roxb had been studied b bone marrow micronucleus test. Fourty five female mice were clasified into five groups: negative control, positive control, 30 mg/kgBW extract, 100 mg/kgBW extract, and 300 mg/kgBW extract. Each group was divided into three groups based on the duration of treatment. The mice were induced with 50 mg/kgBW cyclophosphamide intraperitoneally. After 30 hours, the extract were administered orally into the treatment groups for the duration of 3,7, and 15 days. After treatment, the mice were sacrified to obtain their femur marrow. The parameter investigated was the number of micronucleus. It was shown that the extract at does of 30, 100 and 300 mg/kgBW decreased the percentage of micronucleus cells significantly compared with positive control (p<0,05). The highest decrease of micronuclei cells was showed by the dose of 100 mg/kgBW.Keywords : cytotoxic effect, Garcinia cowa Roxb, bone marrow micronucleus test.AbstrakTelah dilakukan penelitian tentang efek sitotoksik dari ekstrak etanol daun asam kandis (Garcinia cowa Roxb) dengan Metoda Penetapan Mikrinukleus. Sebanyak 45 ekor mencit putih betina, dibagi menjadi lima kelompok yaitu kontrol negatif, kontrol positif, 30 mg/kgBB), 100mg/kgBB, dan 300 mg/kgBB. Tiap-tiap kelompok dibagi lagi menjadi tiga kelompok berdasarkan lama pemberian sediaan uji. Mencit diinduksi dengan siklofosfamida 50 mg/kgBB secara intraperitoneal. Setelah 30 jam, kelompok uji diberikan ekstrak uji secara oral selama 3,7 dan 15 hari. Setalah perlakuan, mencit dikorbankan dan diambil sumsum tulang femurnya. Parameter yang diamati adalah persentase jumlah sel mikronuklei. Hasil penelitian menunjukkan sediaan uji pada dosis 30, 100 dan 300 mg/kkBB mampu menurunkan persentase jumlah sel mikronuklei secara signifikan dibandingkan kontrol positif (P<0,05). Penurunan persentase sel mikronuklei paling besar diberikan oleh sediaan uji dosis 100 mg/kgBB.Kata kunci : Efek sitotoksik, Garcinia cowa Roxb, metode penetapan mikronukleus.

Abstract

Lantana camara Linn is a family of Verbenaceae which grows wild and is widespread in various both tropical and sub-tropical countries. Isolation essential oil of the L. camara leaves extracted by hydrodistillation and were analyzed by Gas Chromatography-Mass Spectrometry (GC-MS). The results of GC-MS analysis show isocaryophyllene (14.39%), ρ-cymene (8.17 %), β-cubebene (7.8 %), α-pinene (7.64 %), and β-elemene (5.51 %) as the main compound. The cytotoxic activity of the isolated essential oil was highly toxic both to Artemia salina shrimp larvae (LC50 15.92 µg/mL) and to breast cancer cells T-47D (IC50 10.67 µM).

Abstract

Several of Calophyllums genus have been searched and proven as medicinal plants and Calophyllum macrophyllum is one of its genus. We have been isolated a compound from ethyl acetate fraction of the stem-bark. The compound was flavan-3-ol (5,7,2’,5’-tetrahydroxy flavan-3-ol). This research aimed to determine inhibition antidiabetic activity and affinity of its compound on α-glycosidase enzyme. In vitro antidiabetic effect shown by IC50 9.10 µg/ml and docking by Arguslab 4.01 shown by ΔG -10,24 kkal/mol as in silico test. The inhibition activity of flavan-3-ol against the α-glucosidase enzyme that is very strong shows its promising potential as a candidate for antidiabetic drugs.

Abstract

Professional nursing service delivery system (SP2KP) is a component of the application of professional values in nursing practice, management and nursing care provision in hospital ward units and self-professional development. The purpose of this study was to determine the relationship between education and training with the implementation of SP2KP by implementing nurses in the inpatient room of Raden Mattaher Jambi Hospital. The type of research carried out in this study is a quantitative study using an analytic research design with cross sectional approach (n = 132 nurses). The test used in this study is the Chy-Square test. Data is collected by using a questionnaire that has been tested for validity and reliability. The results of the study of the relationship between education and training with the implementation of a professional nursing service delivery system by implementing nurses in the Inpatient Room of Raden Mattaher Jambi Hospital. Education that has a significant relationship with the implementation of SP2KP (p value = 0.010). Researcher's suggestion to implementing nurses to improve education will affect the skills and knowledge of implementing nurses in the implementation of SP2KP in the inpatient room of Raden Mattaher Jambi Hospital.
