Abstract

Many microbial populations are found in the rhizosphere, including in the soil layer that follows the plant root system. Rhizosphere soils contain more bacteria, fungi and actinomycetes than non-rhizosphere soils. One of the antibiotic producing bacteria has been isolated from the rhizosphere. The purpose of this study was to isolate and characterize the biochemistry of antibiotic producing bacteria and to evaluate their superiority in inhibiting other types of bacteria. Three microbial isolates were isolated from the rhizosphere of winged bean (K) and banana (P1 and P2) plants. All isolated isolates were tested for antagonism with E. coli, Bacillus, and Pseudomonas bacteria on Nutrient Agar (NA) media to determine antibiotic-producing bacteria. Furthermore, identification and characterization of colony morphology and a series of antagonism and pathogenicity tests were carried out. The three isolates had irregular colony forms (isolates K and P1) as well as oval shapes (isolate P2), flat surface, glossy texture, milky white or cream color, and irregular edges. The antagonism test showed that isolates P1, P2, and K could inhibit the growth of bacterial colonies E. coli, Bacillus, and Pseudomonas, respectively. In the pathogenicity test, the three microbial isolates did not show symptoms of necrosis in tobacco leaves after 2 days of observation.

Abstract

ACC deaminase is a microbial cytoplasmic enzyme that cleaves ACC, a precursor of ethylene, in the stressed plant. The aims of this study were to isolate, identify, and detect the presence of ACC deaminase gene-encoding rhizobacteria from the rhizospheric soil of pineapple plants that have been exposed to abiotic and biotic stress, specifically herbicide, flooding, and Phytophthora spp. stress. A total of 49 rhizobacterial isolates were obtained, seven of which were observed for their growth on DF medium containing 3 mM L-1 ACC. The four best-growing isolates were selected for genomic DNA extraction. They were molecularly identified as Stenotrophomonas maltophilia (3), Burkholderia territorii (2A), Pseudomonas oryzihabitans (5B), and Bacillus tropicus (1E). A set of primers, 105F-acdS 5’-TGCCAAGCGTGAAGACTGC-3’ and 244R-acdS 5’-GGGTCTGGTTCGACTGGAT-3’, were constructed to amplify the ACC deaminase gene (acdS). Based on melt peak curve analysis, four products appeared to show a specific single peak at 86, 89, 87, and 89.5°C, indicating a single product was produced. In addition, a Blast search showed that these four products met the ACC deaminase feature and their acdS sequences were clustered into an ancestral group compared with the bacterial strains deposited in GenBank. These results suggest that ACC deaminase gene-encoding rhizobacteria from a pineapple plantation of tropical origin may affect the acdS sequences and may contribute to the host plant’s stress tolerance.

Abstract

Applying PGPB (Plant Growth-Promoting Bacteria) as bioinoculant under the excessive and continuous use of chemical fertilizer, pesticide, and herbicide need to be highlighted nowadays. One of the most beneficial PGPB is potassium-solubilizing rhizobacteria. Potassium is one of the elements that plant absorbs in a large amounts along with nitrogen. Soil in various regions of Indonesia have low potassium solubility. Therefore, finding the best novel isolates contributing to potassium solubilization need to be carried out continually. The aim of this study is to find potassium-solubilizing bacteria from two different rhizospheres (banana and chili plant) in Leuwikopo Experimental Garden and a cow manure in Cattle Pen, IPB University. To obtain the potassium-solubilizing isolates, we observed and characterized the best growing isolates and measured the solubilizing zone on Alexandrov agar medium. Three out of 10 isolates were selected and tested their pathogenicity on tobacco leave. Solubilization index of three isolates from the highest to the lowest were MPK P (3.07), MPK KT (3.03), and MPK C (2.63), respectively. Necrotic on tobacco leave after isolates treatment showed negative result which means that the three isolates were not pathogenic to plant host. The isolates are recommended to be used as bioinoculant both in greenhouse and field plants which lack of potassium availability in soil.

Abstract

Digital-based laboratory inventory management has begun to be implemented in various universities today. The Laboratory of Soil Science, University of Mataram needs to apply this method to improve the quality of practicum and research services. The borrowing and use of laboratory equipment by conventional methods make it difficult for laboratory staff to store and secure data properly. This community service aims to optimize the conventional method with a method based on the Quick Response Code (QR Code). The design of the QR Code was carried out online which was then attached to each tool and then tested and socialized to users. This program was carried out for 2 months from August to September 2022. The output of this service is the design of a simple QR Code-based information system that can simplify the process of borrowing tools and make them easier to use. The results of implementing the QR Code show that storing data is more well-organized and safe and also makes it easier for beginner users to use the laboratory equipment with the operational procedure and video tutorials provided.

Abstract

Siswa-siswi peserta Kompetisi Sains Nasional (KSN) Biologi SMA Negeri 4 Praya memiliki masalah dalam hal penguasan materi yang tidak pernah diajarkan di kelas dan kurangnya motivasi belajar. Hal tersebut akan menghambat proses pembelajaran efektif yang melibatkan daya analisis, nalar, dan berpikir kritis siswa dalam menjawab soal-soal olimpiade berkarakter High Order Thinking Skills (HOTS). Selain itu, dorongan sekolah agar siswa dapat meraih prestasi olimpiade menjadi tantangan tersendiri. Oleh karena itu, pengabdian kepada masyarakat ini bertujuan untuk menguatkan kompetensi akademik siswa dalam memperisapkan diri menuju KSN Biologi tingkat kabupaten. Metode yang digunakan adalah pemberian motivasi dan latihan soal melalui metode drill. Pelaksanaan kegiatan dilakukan melalui tahapan seleksi peserta tingkat sekolah, pembinaan akademik, evaluasi, dan survey tingkat pemahaman dan kepuasan siswa terhadap metode yang diterapkan. Kegiatan pembinaan siswa berjalan dengan efekif dan lancar selama 19 kali pertemuan tatap muka di kelas. Hasil survey tingkat pemahaman dan kepuasan siswa setelah mengikuti pembinaan menunjukkan bahwa 2 dari 5 peserta merasa paham/puas (42,60%) dan 2 dari 5 peserta merasa sangat paham/puas (36,00%) terhadap pembinaan yang diberikan. Selain itu, 2 dari 4 peserta yang mengikuti lomba tahun 2020 berhasil meraih juara I dan II. Hasil ini menyimpulkan bahwa metode yang diterapkan mampu meningkatkan pemahaman dan kepuasan siswa terhadap materi Biologi, motivasi diri, metode pembinaan, pendampingan akademik, dan keterampilan dirinya yang berujung pada keberhasilan akademik siswa. Harapannya metode ini dapat diterapkan secara intensif dan komprehensif di sekolah lain tanpa mengesampingkan faktor-faktor lain guna menunjang keberhasilan akademik siswa peserta olimpiade. Strengthening the Competencies of Students of SMA Negeri 4 Praya towards Biology Olympiad at District Level through Delivering the Motivation and Drill Method Students of SMA Negeri 4 Praya who participated in the National Science Competition (KSN) of the Biology Olympiad had a problem mastering academic content that had never been taught in class and lack of motivation. This issue will impede the learning process that involves students' analytical, reasoning, and critical thinking skills in answering High Order Thinking Skills (HOTS)-type questions. In addition, the school's encouragement so the students can succeed in the competition is another challenge. Therefore, this community service aims to strengthen the student's academic competency in preparing for the competition. The method used is delivering motivation and the drill method. The stages were carried out by selecting participants at the school level, academic coaching and learning, evaluating, and surveying regarding understanding and satisfaction of students. Student coaching activities run effectively and smoothly during 19 face-to-face learning in class. The survey showed that 2 out of 5 students felt understood/satisfied (42.60%) and 2 out of 5 felt very understanding/satisfied (36.00%) with the coaching provided. In addition, 2 out of 4 participants who took part in the 2020 competition succeeded in winning 1st and 2nd place. These results conclude that the methods applied were able to increase students' understanding and satisfaction of Biology material, self-motivation, coaching method, academic coaching, and students' self-skills, and leads to the success in academic context. This method hopefully can be applied intensively and comprehensively in other schools without putting aside other factors to support the academic success of the Olympiad participating students.

Abstract

Pengabdian kepada masyarakat ini bertujuan untuk mendampingi dan memberikan pengalaman kepada masyarakat tani di daerah Montong Tekot, Desa Sukadana dalam membudidayakan tanaman anggur di lahan kering yang tak produktif. Rangkaian kegiatan pendampingan ini meliputi koordinasi dengan mitra, persiapan dan pengolahan media tanam, persiapan bibit, penanaman, pemupukan, perawatan, dan evaluasi kegiatan. Metode yang digunakan adalah Participatory Rural Appraisal (PRA) yang melibatkan 10 anggota kelompok tani “Patuh Bersama”. Sebanyak 9 dari 10 sangat antusias dan berperan aktif dalam mengikuti kegiatan pendampingan dan menerima kunjungan dari tim pengabdian masyarakat Universitas Mataram. Melalui observasi dan evaluasi, sekitar 90% peserta tergolong puas dengan pengalaman pendampingan budidaya tanaman anggur yang diberikan. Sangat penting pula diketahui bahwa pertumbuhan vegetatif tanaman anggur tumbuh dengan baik yang ditandai dengan bertambah tingginya tanaman dengan rata-rata lebih dari 50 cm dan berkembangnya sulur tanaman dalam waktu sekitar 1 bulan. Diharapkan program budidaya tanaman anggur ini menjadi salah satu program strategis jangka panjang berkelanjutan karena sangat potensial dikembangkan di daerah beriklim kering yang mendukung kebutuhan kawasan pariwisata KEK Mandalika. Coaching of Grape Cultivation in Semi-Arid Areas to the Farmer Group “Patuh Bersama” in Desa Penyangga Sukadana, Mandalika Special Economic Zone, Central Lombok  This community service aims to assist and provide experience to the farmer community in the Montong Tekot area, Sukadana Village in cultivating grapes on dry, unproductive land. This series of coaching activities includes coordination with partners, preparation and processing of planting media, seed preparation, planting, fertilizing, maintenance and evaluation of activities. The method used was Participatory Rural Appraisal (PRA) which involved 10 members of the "Patuh Bersama" farmer group. As many as 9 out of 10 were very enthusiastic and played an active role in participating in coaching activities and receiving visits from the Mataram University community service team. Through observation and evaluation, around 90% of participants were satisfied with the grape cultivation assistance experience provided. It is also very important to know that the vegetative growth of grape plants is growing well, which is indicated by the increase in plant height by an average of more than 50 cm and the development of plant tendrils in about 1 month. It is hoped that this grape cultivation program will become one of the long-term sustainable strategic programs because it has great potential to be developed in dry climate areas that support the needs of the Mandalika SEZ tourism area.

Abstract

The unfavorable rhizosphere environment for the growth of rhizobacteria and plants such as flooding and herbicide is relatively common for these rhizobacteria to express the acdS gene which encodes ACC deaminase. This study aims to observe the expression of the acdS gene in the rhizobacterial isolates from pineapple plants affected by the environmental stressors. The expression of the acdS gene was observed by growing the bacterial isolate on a Dworkin-Foster medium containing 3 mM.L-1 ACC and observing the RNA abundance of the isolate based on the Cq value in the RT-qPCR reaction. The selected bacterial isolates were then identified molecularly using the 16S rDNA gene. This study showed that 2 out of 7 bacterial isolates growing on a DF+ACC medium were selected for the RT-qPCR process. The two isolates came from flooding and herbicide stressors with Cq values ​​of 17.86 and 20.87, respectively. The two isolates were 1F and 2H which were identified molecularly as Burkholderia sp. and Pseudomonas psychotolerans in which both genera of bacteria commonly have the acdS gene. The two isolates can be used as inoculant candidates in the production of biological organic fertilizers as well as in their empowerment through land use management practices to help plants tolerate environmental stressors and promote plant growth.

Abstract

The development of cavendish banana cultivation has higher potential compared to other types of bananas. This is because Cavendish bananas have a higher market selling value, especially as they are a type of banana that sells well on the international market. One of the villages that has the potential to develop cavendish banana tree cultivation is Sukadana Village. This village is one of the buffer villages for the Mandalika Special Economic Zone which is expected to provide basic agricultural needs in the Mandalika area. Service activities will be carried out in December 2022 until may 2023 with the target farmer group Patuh Bersama. This service activity aims to increase community income through assistance in the cultivation of Cavendish bananas. The results of the service activities carried out are providing insight to the community that cavedish bananas are a type of banana that is commonly consumed by the international community. Apart from that, the community can also develop the cultivation of cavendish bananas not only in their yards, but can also cultivate them on unused agricultural land

Abstract

Di desa Sukadana, dusun Montong Tekot, kecamatan Pujut, Lombok Tengah, masalah utama adalah jumlah limbah kotoran sapi yang sangat banyak, dengan populasi sapi mencapai 36.466 ekor yang menyumbang sekitar 55% dari total limbah di kabupaten Lombok Tengah, Nusa Tenggara Barat. Kondisi minimnya pengetahuan dan pengalaman masyarakat mitra dalam mengelola limbah kotoran sapi menjadi kompos pupuk kandang menjadi alasan utama dilakukannya pengabdian ini. Kegiatan ini bertujuan memberikan pengetahuan kepada kelompok tani mitra tentang pengolahan limbah kotoran sapi menjadi kompos pupuk kandang dengan metode pengomposan cepat dan memberikan pengetahuan kepada kelompok tani mitra tentang cara memanfaatkan dan menerapkan kompos pupuk kandang pada tanaman pakchoy. Kegiatan pengabdian kepada masyarakat ini melibatkan kelompok tani dan mahasiswa MBKM. Rangkaian kegiatan telah dilaksanakan sesuai rencana dan tujuan yang telah ditetapkan. Melalui kegiatan penyuluhan, petani meningkatkan pengetahuan mereka tentang pengolahan limbah ternak menjadi produk yang bermanfaat. Melalui workshop pengomposan cepat, petani memahami pentingnya penggunaan bioaktivator seperti M-11 dan konsorsium BPF untuk mempercepat proses pengomposan. Dari hasil menunjukkan bahwa bioaktivator M-11 lebih efektif dalam menguraikan limbah organik dibandingkan konsorsium BPF, hal ini terlihat dari perubahan nilai pH yang lebih rendah pada bioaktivator M-11. Melalui pendampingan dalam budidaya pakchoy, petani menyadari manfaat kompos limbah ternak sebagai pupuk penyubur tanah dan tanaman. Petani tetap memerlukan kegiatan semacam ini, terutama berkaitan dengan transfer teknologi untuk menjaga kesuburan tanah dan meningkatkan pertumbuhan tanaman dengan biaya produksi yang lebih terjangkau. Transfer of Fast Composting Technology and Utilization of Compost Products in Pakchoy Plants Abstract Sukadana village, Montong Tekot hamlet, Pujut subdistrict, Lombok Tengah, faces a significant challenge in managing the vast amount of cattle manure generated by its 36,466 cattle, accounting for 55% of the total waste in Lombok Tengah Regency, West Nusa Tenggara. Limited knowledge and experience among local farmers in composting cattle manure into organic fertilizer necessitates this community service initiative. Empower local farmer groups with the knowledge and skills to convert cattle manure into organic fertilizer using a rapid composting method. Educate farmer groups on the effective utilization of cattle manure compost in pakchoy cultivation. Collaboration between farmer groups and MBKM students in community service activities. Successful execution of planned activities and goal achievement. Enhanced knowledge among farmers regarding livestock waste management into valuable products. Understanding of bioactivator usage (M-11 and PSB consortium) to expedite the composting process. Superior effectiveness of bioactivator M-11 in decomposing organic waste compared to PSB consortium, evident in lower pH values. Recognition of cattle manure compost's benefits as a soil and plant fertilizer through pakchoy cultivation guidance. Continuous demand for technology transfer initiatives to maintain soil fertility and promote cost-effective crop growth.
