Abstract

Tomatoes are well-known as the source of antioxidants due to the content of lycopene, i.e., a potential free radical inhibitor. Gel peel-off masks are among the chemical products containing antioxidants. The objective of this study is to formulate and examine the antioxidant activity of the gel peel-off mask made of tomato (Solanum Lycopersicum L.) extract using the DPPH method. The first step of the study was extracting tomato fruits by macerating the fruits using alcohol solvents 96%. The obtained thick extract was used as the material of the gel mask; the percentage of the extract comprised 3% (F1), 5% (F2), and 7% (F3), and it used PVA basis 11% and viscolam 3%. According to the result, the gel peel-off masks meet the requirements of physical and storage stability evaluation. The result of the antioxidant test reveals that the IC50 value of the three masks is F1 189.22 µg/mL, F2 89.34 µg/mL, and F3 36.77 µg/mL. Further, the result of T-Test reveals that P-value = 0.019 (α = 0.05), indicating a difference in the result of the antioxidant test of the three formulae in day one (t0) and day 28 (t28).

Abstract

Flavonoids are chemical compounds derived from plants which are a group of secondary metabolites. Flavonoids Flavonoid compounds are polyphenolic compounds that have 15 carbon atoms arranged in a C 6 -C 3 -C 6 configuration, namely two aromatic rings connected by 3 carbon atoms that can or cannot form a third ring. Flavonoids are widely distributed in angiosperms, gymnosperms, and pteridopites. These flavonoids provide physiological and pharmacological effects on living things. The purpose of this study was to identify the flavonoid compounds present in the leaf extract of the blood sambang plant (Excoecaria cochinchinensis L). In this study, a multilevel maceration extraction was used using n-hexane, ethyl acetate and methanol solvents and then the filtrate was evaporated using an evaporator. Positive phytochemical screening results indicated the presence of flavonoids in methanol extract and negative in n-hexane and ethyl acetate extracts. From the methanol extract, TLC was carried out with an eluent ratio of methanol ethyl acetate (8: 2), it was obtained the separation of two stains where one of the stains had the same Rf value as the Rf value of flavonoids, then performed a UV-Vis spectrophotometric analysis resulting in three absorption wavelengths, namely band I (350 nm) and band II (255 nm and 267 nm). the spectrum obtained is a class of flavone compounds.

Abstract

Endophytic fungi live in mutualistic symbiosis and can produce secondary metabolites similar to their host. This research aims to isolate, characterize, and test the antioxidant activity of secondary metabolites from turmeric stems. This research is experimental research where the method used for endophytic fungi isolation is the direct planting method, and antioxidant activity is tested using qualitative and quantitative methods with DPPH (1.1-diphenyl-2-picylhydrazyl). One fungal isolate was obtained with the fungal isolate code (BK). Based on macroscopic and microscopic characterization, the BK fungal isolate indicates the Deuteromycota class's characteristics. The qualitative antioxidant test results indicate that the fraction of BK endophytic fungal isolate indicates antioxidant activity, indicated by the yellow color against a purple background on the TLC plate observed under visible light. The respective ICso values for the ethyl acetate and n-hexane fractions were 52.989 µg/ml and 89.725 µg/ml, categorized as strong antioxidants. The methanol fraction had an ICso value of 119.02 µg/ml, categorized as moderate, while the pure supernatant fraction had an ICso value of 156.86 µg/ml, categorized as weak antioxidant activity.

Abstract

Candida albicans (C. albicans) is a fungus that causes candidiasis. C. albicans has the enzyme Lanosterol 14- α demethylase. Lanosterol 14-α-demethylase has bioactivity in converting lanosterol to ergosterol, a special sterol found in fungal membranes, which mediates membrane permeability and fluidity. One way to treat candidiasis is to use traditional medicinal plants. Sungkai (Peronema canescens) can be used as an anti-fungal medicine. Drug development efforts can be done using the In silico method. Objectives of this study was to determine the interaction between terpenoid, flavonoid, and phenol compounds in Sungkai leaves against Lanosterol 14-α demethylase in Candida albicans. The research design used was a pre-experimental one-shot case study. Toxtree software was used to test the toxicity of compounds. Test for compound potency using the Pass Online webserver. Docking molecular using PyRx software. Visualization of docking results using Discovery Studio 2019 Software. Physicochemical test of compounds using Lipinski Test. The results showed that the compounds that had a low risk of toxicity were butanoic, catechol, guaiacol, hydroquinone, isopropanol, methanoic acid, palmitic acid, and phytol. Compounds that have anti-fungal activity based on Pharma-expert analysis are butanoic acid, genkwanin, guaiacol, hydroquinone, isopropanol, palmitic acid, and phytol. Compounds that have hydrogen bonds and binding affinity value 10 and RMSD value 2 are butanoic acid, catechol, genkwanin, guaiacol, hydroquinone, isopropanol, methanoic acid, palmitic acid, and quinic acid compounds. These compounds are thought to inhibit the Lanosterol 14-alpha demethylase enzyme in C. albicans. Compounds that comply with Lipinski's rules are anthocyanin and genkwanin compounds. The groups of compounds found in Sungkai leaves that have antifungal activity are the terpenoids, flavonoids, and phenols.
