Abstract

Background: Aqueous extract of purple sweet potato tuber shown to decrease the blood glucose levels and has antioxidant properties in rats with streptozotocin-induced diabetes. The purpose of this study is to prove that an increase in anthocyanin levels in the blood of rats after administration of aqueous extract of purple sweet potato tubers in healthy and diabetic rats. Methods: This is an experimental study with randomized post-test only control group design. The subject of experiments were 72 rats, divided into two major groups, namely  diabetic rats (treatment group 36 rats) and control group (36 healthy rats). Each major group was divided into 6 small groups each 6 rats. The control group was given aqueous extract of purple sweet potato tuber with a dose of 4 ml single dose. The diabetic group were given streptozotocin and followed by aqueous extract of purple sweet potato tuber 4 ml single dose.  Observed variables include: blood glucose levels to define diabetes. To determine the levels of anthocyanin, the blood were taken and the level of anthocyanin were examination by HPLC method, in healthy  and diabetic rats after 30 min, 1 hours, 2 hours, 4 hours, 8 hours and 16 hours of giving single dose of aqueous extract of purple sweet potato tubers. Results: The results showed an increase in blood sugar levels were significant (p < 0.05) in group given streptozotocin compared to control. Anthocyanin levels in the blood of two groups were significantly different from 30 min after administration of the extract to 8 hours after administration of the extract (p < 0.05). Higher anthocyanin levels (p < 0.05) was observed in control group (healthy rats) compared to rats with streptozotocin-induced diabetes. Peak levels of anthocyanin in blood were achieved after 2 hours of feeding. Conclusion: The conclusion of this study is the absorption of anthocyanin in aqueous extract of purple sweet potato tuber better in healthy rats compared to diabetic rats. The peak levels of anthocyanin was reached after 2 hours of aqueous extract of purple sweet potato tubers single dose consumption either in healthy rats and in diabetic rats.

Abstract

Background: Hypercholesterolemia can cause oxidative stress, endothelial dysfunction and atherosclerosis. Anthocyanin can prevent oxidative stress, it will decrease the inflammation by decreasing the pro inflammatory cytokine in animal models. Purple sweet potato in Bali has been proven of having high anthocyanins content. The aim of these study was to prove the ability of aqueous extract of Balinese purple sweet potato (Ipomoea batatas L.) in decreasing the blood interleukin-1 level in hypercholesterolemic rabbits. Methods: Subjects of this study were 18 male adult rabbits divided into 3 groups with randomized post test only control group design. Group 1 rabbits were given standard diet as a control group. Group 2 rabbits were given high cholesterol diet. Group 3 rabbits were given aqueous extract of Balinese purple sweet potato 4 ml/Kg.BW/day and high cholesterol diet. After 60 days of treatment, the blood level of total cholesterol, MDA and interleukin- 1 were measured as post test examination. The data were analysed by using anova. Abstract

Background: Purple sweet potato tuber extract in liquid form, shown to lower blood pressure of hypertensive rats and hypertensive patients. Liquid dosage form, increasingly unstable when stored for long periods, thus it is necessary to find a more stable dosage form. The objective of this research was to prove that dry extract of purple sweet potato tuber has the same effects with liquid extract in hypertensive rats. Method: The is a randomized pre-test and post-test control group design study. Thirty hypertensive model rats were divided into 5 groups (6 animals for each group). Group 1 was treated as a control group. Group 2 to group 5 were treated as treatment groups. Treatment groups were given liquid extract at a dose of 3 ml, dry extract with the dose of 200 mg, dry extract that stored in a certain time with a dose of 200 mg and given a mixture of dry extract with a carrier substance (capsule formula) with a dose of 200 mg, respectively, for 4 weeks. Variables observed were blood pressure, SOD and MDA level of the blood. Results: The results showed that the group given the purple sweet potato tuber extract lowers blood pressure were significantly in liquid extract or dry extract (p <0.01). The group that was treated with capsule formula showed more decrease in blood pressure than dry extract group (p <0.01). The similar results happened on MDA and SOD level in blood. Conclusions: Thus, it can be concluded that dried extract of purple sweet potato tubers has the same effectiveness with liquid extract, both as an antihypertensive and also antioxidant. Capsules formula is more effective than liquid extract and dry extract.

Abstract

Compliance of hypertensive patients to take medication is one of many determinant factors to achieve successful treatment. Side effects and the expensive price of drugs are the causes of the incompliance of patients taking the medication. Utilization of herbal medicine is a new hope to resolve the issue. Purple sweet potato tuber is a plant part that expected has beneficial effect in lowering blood pressure because it contains anthocyanins which are antioxidants and can preserve endothelial function. To prove these hypothesis, a study was conducted with randomized control group pre and post-test design. The  study was done on 20 adult male Wistar rats that were divided into two groups of 10 rats.  Both groups of rats were made hypertensive by administering high doses of NaCl. Control group of rats given only NaCl alone for 14 days. Treatment group were given NaCl and water extract of purple sweet potato tuber with a dose of 4 cc per day for 14 days. Before treatment and during treatment, blood pressure were taken everyday with special sphygmomanometer. The results indicate a significant difference in blood pressure between the control group with treatment (P = 0.0001). In the treatment group, it was observed that there was a significant decrease in blood pressure compared to the control group (P=0.0001). From the results of this study, it can be concluded that administration of purple sweet potato tuber water extract may lower high blood pressure of rats induced by NaCl.

Abstract

Food stuffs with high flavonoids content  are believed to prevent various diseases caused by oxidative stress because of its antioxidants effect.  Purple sweet potato leaves have been proved containing high flavonoids, and can be developed very easily. To prove antioxidant and hypolipidemic properties of the water extract of purple sweet potato leaves, one research was conducted in the Department of Pharmacology, Faculty of Medicine, Udayana University. The study design was randomized control group pre- and post-test.  Twenty adult male wistar rats were divided into two groups of 10 rats. Both groups of rats were given high-cholesterol diet for three months to induce dyslipidemia. Control group of 10 rats were given only high-cholesterol diet alone, whereas the treatment group also treated with purple sweet potato leaf water extract with a dose of 6 cc per day divided into two doses. Before treatment and after treatment, lipid profile and blood SOD levels were measured. The results showed a decrease in total cholesterol, triglycerides and LDL cholesterol significantly in the treated group (P = 0.0001). In the treatment group there was also an increase in HDL cholesterol and blood SOD which was significantly differ than the control group (P = 0.0001). From the results of this study, it can be concluded that administration of water extract of purple sweet potato leaves can improve the lipid profile and increase blood SOD of rat given high-cholesterol diet.

Abstract

Pada keadaan normal platelet tidak dapat beragregasi atau adhesi, namun bila terjadi gangguan fungsi platelet atau terjadi kerusakan pada endotel pembuluh darah dapat terjadi adesi dan aggregasi platelet. Agregasi platelet dapat menyebabkan kelainan yang fatal seperti acut myocardial infarction, ischaemic heart disease, stroke,dan lain-lain. Semua kelainan tersebut dapat diatasi atau dicegah dengan obat-obat antiplatelet. Hingga saat ini telah beredar puluhan jenis obat antiplatelet dengan kebaikan dan kekurangan masing-masing. Salah satu dasar pertimbangan dalam pemilihan obat yang dipakai adalah farmakodinamik dari obat-obat antiplatelet, sehingga pengetahuan tentang farmakodinamik dari obat-obat amat diperlukan. Adanya berbagai reseptor pada permukaan platelet merupakan salah satu faktor penentu farmakodinamik dari obat-obat anti platelet. Reseptor ADP( Adenosin diphosphat) terutama P2Y12 merupakan peran sentral dalam aktivasi dan agregasi platelet sehingga obat yang dapat menghambat/antagonis reseptor ADP merupakan harapan baru dari obat-obat antiplatelet. Clopidogrel merupakan antagonis reseptor ADP yang baru selain ticlopidin. Clopidogrel dikatakan lebih efektif dibandingkan aspirin dan obat lain pada kasus myocardial infark, ischaemic stroke, dan vascular desease, oleh karena itu nampaknya pemahaman menganai mekanisme kerja clopidogrel perlu deketahui.

Abstract

Cancer is still one of the leading death causes worldwide. Consuming antioxidants can reduce the cancer incidence. Anthocyanin that contained in purple sweet potatoes can inhibit DNA damages. In order to prove the activity of purple sweet potatoes on cancer cell, it is necessary to have some direct research on cancer cell line.This study is a simple experimental research method. The activities of the anticancer are evaluated from cytotoxic and antiproliferative effects on cell line breast cancer T47D. Cytotoxic effects is examined with cultured cell stained with tryphan blue exlusion. Each well are contain of 36,000 cells, which is given ethanol extract purple sweet potatoes with 10 dose variation starting from 500  g/mL up to 10,000  g/mL replicated three times, then the inhibitory percentage is calculated. Antiproliferative activity is evaluated by incubating cancer cells that have been given ethanol extract purple sweet potatoes in 3 doses variation; 500, 1,000 and 2,000  g/ml in 24, 48 and 72 hours.Result of this study showed that ethanol extract purple sweet potatoes had cytotoxic activities on T47D breast cancer cell line in these dose variations, 500; 1,000; 2,000; 3,000; 4,000; 5,000; 6,000; 7,000; 8,000; 9,000 and 10,000  g/ml. The cytotoxic activity on each of dose variations above are 27.56; 42.67; 57.78; 66.67; 72.44; 79.56; 85.33; 87.56; 92.44;100; and 100 %. Ethanol extract purple sweet potatoes showed antiproliferative activities on T47D breast cancer cells in 24, 48, 72 hours incubation in concentrations 500; 1,000; and 2,000  g/ml; those are 35,700; 29,800; 25,500 cells (24 hours); 72,500; 60,300; 52,600 cells  (48 hours); 149,500;122,600;107,300 cells  (72 hours). In conclusion, ethanol extract purple sweet potatoes had cytotoxic and antiproliferative activities on cell line breast cancer T47D.

Abstract

Pendahuluan: Ubi jalar ungu merupakan varian ketela rambat dan memiliki pigmen antosianin yang bermanfaat sebagai antioksidan. Tujuan penelitian adalah untuk melihat efek perlindungan oleh ubi jalar ungu terhadap kerusakan akibat radikal bebas yang timbul setelah aktifitas fisik berat. Metode: Penelitian eksperimental dilakukan pada 24 ekor mencit jantan jenis Balp/C yang dibagi dalam 4 kelompok dengan rancangan penelitian randomized control group post-test only design. Pemeriksaan histologis sel hati dilakukan untuk melihat adanya gambaran sel nekrosis. Analisis statistik uji anova dilakukan untuk mengetahui perbedaan jumlah sel yang mengalami nekrosis antar kelompok. Hasil: Jumlah rerata sel nekrosis pada kelompok aktifitas fisik berat (0,667) lebih besar dibandingkan kelompok kontrol (0,00). Rerata sel nekrosis pada kelompok aktivitas fisik maksimal dengan ekstrak ubi jalar ungu (0,167) lebih rendah dibandingkan kelompok tanpa ubi jalar ungu (0,667), p&lt;0,05. Kesimpulan: Ubi jalar ungu mampu mengurangi angka kejadian nekrosis pada sel hati akibat radikal bebas setelah beraktivitas fisik maksimal.

Abstract

Pain is an unpleasant sensory and emotional experience that can affect the quality of life and leads to decreased productivity in patients. Low back pain (LBP) is one of the significant causes of disability worldwide with lifelong incidence. The purpose of this literature review describes the potential of anthocyanin-based Poly (Methyl Methacrylate) (PMMA)   nanoparticles as the management of inflammatory pain in the Hernia Nucleus Pulposus (HNP ). The method used is a literature study by entering the keyword. Of the 77 journals reviewed, 47 journals were found by the topic and used as a reference for this work. The anthocyanin-based PMMA nanoparticles act as anti-nociceptors by inhibiting microglia that produce inflammatory mediators in HNP. Poly (Methyl Methacrylate) nanoparticles have specific targets in microglia. Anthocyanins have the effect of inhibiting inflammatory pain through many destinations. Anthocyanin inhibits the synthesis of nitric oxide (NO ) and prostaglandin E2 (PGE 2) and inhibits the activation of p38 MAPK and NF-kB pathways that express TNF-α and IL-1β genes as anti-nociceptive. The anthocyanin-based PMMA nanoparticles have potential as a novel therapy for inflammatory pain in HNP. There has been no research between these modalities. Therefore, further research is needed to find out the exact potential of anthocyanin-based PMMA nanoparticles.

Abstract

Pendahuluan: Pakaian compression base layer dinyatakan mampu mempercepat sirkulasi darah untuk membantu penguraian asam laktat sehingga mempercepat pemulihan denyut nadi, meningkatkan proses transfer panas dari tubuh ke lingkungan yang dapat menjaga suhu ideal tubuh dan meningkatkan kenyaman ketika berolahraga. Tujuan: Tujuan penelitian ini adalah untuk membuktikan penggunaan pakaian compression base layer mempercepat pemulihan fisiologis tubuh ditinjau dari percepatan waktu pemulihan denyut nadi dan suhu tubuh serta meningkatkan kenyamanan tubuh mahasiswa ketika berolahraga. Metode: Penelitian dilakukan dengan desain eksperimental control group post-test only yang dikembangkan dalam bentuk rancangan silang (two-period cross over design) dengan jumlah sampel 30 mahasiswa yang dibagi menjadi dua kelompok. Waktu pemulihan denyut nadi dan waktu pemulihan suhu tubuh diukur berdasarkan waktu yang dibutuhkan oleh masing-masing sampel (dalam satuan detik) untuk mengembalikan denyut nadi dan suhu tubuh kemabali ke kondisi denyut nadi dan suhu tubuh sebelum melakukan aktivitas olahraga. Kenyamanan tubuh diukur dengan menggunakan kuisioner Kenyamanan Pakaian Olahraga Compression Base Layer. Data dianalisis dengan uji T-paired dengan taraf signifikan p<0,05. Hasil: Hasil analisis menunjukkan bahwa terdapat perbedaan yang bermakna (p<0,05) antara penggunaan pakaian olahraga compression base layer dengan pakaian olahraga berbahan katun. Compression base layer terbukti mampu untuk: mempercepat waktu pemulihan denyut nadi dari 826,30 detik menjadi 648,70 detik, mempercepat waktu pemulihan suhu tubuh dari 494 detik menjadi 400 detik, dan meningkatan skor kenyamanan tubuh dari 42,27 menjadi 49,83. Simpulan: Disimpulkan penggunaan pakaian olahraga compression base layer mempercepat waktu pemulihan denyut nadi dan suhu tubuh serta meningkatan kenyamanan tubuh saat berolahraga.
