Abstract

The efficiency of expression of recombinant proteins in host cells depends on achieving proper protein folding, solubility, and biological activity. Critical environmental factors, including temperature, pH, nutrient availability, and inducer concentration, significantly influence protein yield, stability, and functionality. Optimizing these conditions is essential to minimize protein misfolding and aggregation, which can adversely affect expression efficiency and reduce bioactivity. This study aims to measure the effect of temperature on the growth of Escherichia coli BL21 harboring the recombinant JSU protein, derived from the ENV-SU gene of the Jembrana virus. A controlled laboratory experiment was conducted in which bacterial cultures were incubated at 25°C and 37°C under identical conditions, except for temperature. The results indicate a statistically significant difference in bacterial growth between the two temperature conditions (p = 0.08), suggesting that E. coli BL21 exhibits enhanced growth at 37°C compared to 25°C. The mean bacterial growth rate at 37°C was 1.756, whereas at 25°C, the average growth was 1.425, demonstrating that higher temperatures facilitate bacterial proliferation. However, due to the relatively small sample size, further validation with a larger dataset is required to confirm these findings. These findings highlight the critical role of temperature optimization in enhancing bacterial growth and improving recombinant protein production efficiency, emphasizing the need for further investigation with a larger dataset to validate the results. Abstract

Benzo(a)pyrene (BaP) merupakan senyawa toksik dan karsinogenik yang dapat menyebabkan stres oksidatif, peradangan, dan gangguan fungsi organ, termasuk ginjal. Penelitian ini bertujuan untuk menguji efek nefroprotektif ekstrak metanol daun biwa (Eriobotrya japonica) terhadap kadar kreatinin tikus model kanker payudara yang diinduksi BaP. Penelitian ini menggunakan rancangan acak lengkap dengan lima kelompok perlakuan, yaitu kontrol negatif (tanpa perlakuan), kontrol positif (BaP 50 mg/kg BB), serta tiga kelompok perlakuan dengan pemberian ekstrak daun biwa dosis 200, 300, dan 400 mg/kg BB selama 30 hari. Kadar kreatinin diukur menggunakan metode modifikasi Jaffe. Data hasil penelitian dianalisis menggunakan uji ANOVA dan dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan bahwa kadar kreatinin serum darah tikus pada semua kelompok masih berada dalam rentang normal, dengan nilai antara 0,66 hingga 0,73 mg/dl, dan tidak terdapat perbedaan yang signifikan (p0,05) antara kelompok perlakuan dan kontrol. Hal ini mengindikasikan bahwa paparan BaP tidak secara langsung mengganggu fungsi ginjal tikus model kanker payudara, serta pemberian ekstrak daun biwa tidak menunjukkan efek protektif yang signifikan terhadap kadar kreatinin. Namun, kandungan antioksidan dalam daun biwa, seperti flavonoid dan triterpenoid, tetap berpotensi dalam menjaga stabilitas fungsi ginjal melalui mekanisme antioksidatif dan antiinflamasi

Abstract

Parasites are divided into two based on their habitat, namely endoparasites and ectoparasites. Endoparasites are a group of parasites that live in the host's body to obtain food from the host. The spread of endoparasites in livestock can occur through contaminated feed, water, and livestock equipment. The purpose of this study was to identify the species of worm eggs in the feces of native chickens (Gallus gallus domesticus) at the Rumput Ganas Mini Farm, Loa Janan Ilir, Samarinda. 2g of chicken feces were taken and then 10 ml of NaCl solution was added. Homogenized and filtered using gauze. Poured into a centrifuge tube, then centrifuged for five minutes at a speed of 1500 rpm. The results of the centrifuge solution that settled on the surface were taken using a dropper pipette and then placed on an object glass. Covered using a cover glass, then observed under a microscope starting from the smallest magnification. The results of the study showed that two species of endoparasites were found in the feces of local chickens found in the mini farm loa janan ilir's vicious grass, namely Diphyllobothrium latum and Ascaridia galli.  Keywords: Ascaridia galli, Dyphyllopbothrium latum, Endoparasite, Gallus gallus domesticus, Poultry Farming

Abstract

This study aims to determine the effect of administering Jamblang Stem Bark Ethanol Extract (Syzygium cumini L.) (JSBEE) on the activity and phagocytosis capacity of macrophages in mice (Mus musculus) infected with Staphylococcus aureus bacteria. The research method used a completely randomized design (CRD) with five treatments and five repetitions, conducted in-vitro and in vivo test. The treatments for in vitro involved administering distilled water (T0), Stimuno (T1), and JSBEE at concentrations of 10 ppm (T2), 100 ppm (T3), and 1000 ppm (T4). Subsequently, in vivo treatment was conducted using distilled water (T0), stimuno (T1), JSBEE at 10 mg/kg (T2), 100 mg/kg (T3), and 1000 mg/kg (T4). Initially, mice underwent in vivo administration of JSBEE, administered orally via catheter tip, with a dosage of 1 mL per 10 g of body weight. JSBEE was administered orally for 10 days, followed by infection with S. aureus on the 11th day. The in vitro tests were conducted by isolating macrophage cells from the intraperitoneal fluid, to which S. aureus and JSBEE were added. Intraperitoneal fluid collected from the mice was used to prepare smears using the thin blood smear method and Giemsa staining. Macrophage phagocytosis activity was observed and assessed based on the percentage activity formula, and the phagocytic capacity of macrophages was measured by the number of S. aureus cells phagocytosed. The results indicated that JSBEE significantly affected (P0.05) both the activity and phagocytic capacity of macrophages in vivo and in-vitro. The best concentration of JSBEE for increasing both the activity value and phagocytic capacity of macrophages was 100 ppm (T3).

Abstract

This community service activity addresses the gap between entrepreneurial theory and practical experience among university students, particularly in preparing them to become job creators. The aim of this initiative was to enhance students entrepreneurial competence through a collaborative and experience-based learning model. Conducted as an international collaboration between Universitas Syiah Kuala, Indonesia, and Politeknik Seberang Perai, Malaysia, the activity employed a Service Learning approach involving interactive sessions, group discussions, and business model design. Students were exposed to real-world entrepreneurial practices presented by experts from both institutions. The results showed a significant improvement in students understanding and ability to develop viable business ideas using the Business Model Canvas. Participants demonstrated increased confidence and motivation to pursue entrepreneurship, and the activity contributed to developing practical skills aligned with sustainable development goals. This program model presents a replicable approach for enhancing entrepreneurial education through cross-border academic collaboration and experiential learning.
