- Game Show Host: The title says it all.
- Large Ham: What's Ruff without this?
- Insistent Terminology: As established in the first episode, he doesn't like it when Studio G is called a garage.Ruff: All right, rule number one! We never refer to Studio G as "the garage", okay? It makes me a little insecure.
Scruff Ruffman

- Evil Twin: Is this to Ruff.
- Noodle Incident: We never learn how or why he ended up in prison.
Grandma Ruffman
Ruff's grandmother, who was a game show host in her youth.
- Ambiguously Jewish: Nothing is mentioned about her exact religion, but she has a Yiddish accent and calls Ruff "bubby" (a diminutive of "bubala", a Yiddish term of affection).
Great Uncle McRuffmantosh
Ruff's great-uncle, who is a scholar and historian.
Glen
Ruff and Scruff's nephew, who is a fan boy of the fantasy and superhero genres.
- Ambiguously Related: Downplayed as he is explicitly Ruff and Scruff's nephew, but whether he is Roxy's son or the son of another unidentified Ruffman sibling in Ruff's generation is never revealed.
- Pubescent Braces: Wears braces, making it hard for Ruff to understand what he is saying at times.
Ruffael Ruffmanowitz
Ruff's great-great-great-great-great-great-grandfather, who was an inventor.
- Posthumous Character: Because of the 6 times great grandfather relationship to Ruff, he is only mentioned.
Helga Von Ruffman
Ruff's cousin, who lives in Germany and works as a fashion designer.

A Ruffman family member who is a pirate.

Ruff's parents, who disappeared when he was a puppy. Finding them is the plot of season 5.

Charlene
The poodle living next door to Ruff, whom he has a crush on.
- Satellite Love Interest: We only know that Ruff wants to be with her, and not much else, but it's obvious she won't return Ruff's feelings and isn't interested in him.
Spot Spotnik
Ruff's long time rival, who mainly feuds with him over Charlene.
- The Rival: Ruff's rival who constantly tries to one up him, and get with Charlene.
- Wardrobe Malfunction: In the end of one episode, Spot's pants disintegrate leaving his bottom half exposed in public due to Ruff activating its self-destruct mechanism to get back at him.
Blossom

- Cats Are Snarkers: Or rather, SilentSnarkers: Blossom doesn't speak, but her actions scream sarcastic.
- Hyper-Competent Sidekick: Before her promotion she's a highly skilled assistant, with knowledge of any topic relevant to the episode. She's also a skilled acrobat.
- Not So Stoic: She's normally a Silent Snarker who doesn't express much emotion, but when Marc compares Ruff's girth to that of a beluga whale, she can't stop herself from full-on laughing at the comment. It is incredibly precious.
- Overly Long Name: Her actual name is 'Princess Blossom Pepper Doodle Van Yum Yum.'
- Smart People Wear Glasses: She wears glasses and is shown to be much more intelligent than Ruff or Chet.
- What Happened to the Mouse?: Or rather, cat. We don't see her after The Ruff Ruffman Show.
- Why Did It Have to Be Snakes?: She's afraid of vacuum cleaners.

- Troll: In "Ruff Amuck", she transfers Ruff into her computer and erased his body details except his face. She uses A.I. to type and add body parts to him, she ends up giving him different body parts, such as as an orange, and even rabbit ears, much to Ruff's annoyance.'

- Why Did It Have to Be Snakes?: She shares Ruff's arachnophobia and isn't too thrilled about the challenge in which she and Sterling learn about spiders. Thankfully, she gets over her fear of them and earns 5 bonus points for it.

- Earn Your Happy Ending: Marco was in last place by the last episode and one could be forgiven for thinking that he wouldn't win; he overcame the odds to become the final winner of the series though.

- Graceful Loser: She gave Marco a hug when he won.
- Odd Name Out: Like Anna and Isaac before her, she has a name that starts with a vowel instead of a consonant like her fellow contestants.
