With the aim of finding methods that could constitute a solid alternative to melissopalynological and physicochemical analyses to determine the botanical origin (floral or honeydew) of honeys, the free amino acid content of 46 honey samples has been determined. The honeys were collected in a small geographic area of ∼2000 km 2 in central Spain. Twenty-seven honey samples were classified as floral and 19 as honeydew according to their palynological and physicochemical analyses. The resulting data have been subjected to different multivariant analysis techniques. One hundred percent of honey samples have been correctly classified into either the floral or the honeydew groups, according to their content in glutamic acid and tryptophan. It is concluded that free amino acids are good indicators of the botanical origin of honeys, saving time compared with more tedious analyses.

The monoclonal antibodies MAC236 and MAC265, raised against a soluble component of pea nodules, were used to elucidate the presence and subcellular localization of glycoprotein epitopes during the development of lupin (Lupinus albus L. cv. Multolupa) nodules, by means of immunocytochemistry and Western blot analysis. These antibodies recognize a single band of 95 kDa in pea, soybean and bean nodules, whilst two different bands of 240 and 135 kDa cross-react with MAC236 and MAC265 respectively in lupin nodules. This fact may indicate that the recognized epitopes can be present in different subcellular compartments and/or play different roles through the development of functional nodules. The results show that MAC265 is mainly associated with Bradyrhizobium infection and with the development of nodule primordium, in the first stages of nodulation. MAC265 is also detected when glycoprotein transport takes place across the cytoplasm and the cell wall, and also in the intercellular spaces of the middle cortex, attached to cell walls. The amount of MAC265 remains constant through nodule development. In contrast the amount of MAC236 increases with nodule age, parallel to the establishment of nitrogenase activity. This antibody is localized in cytoplasmic globules attached to the inner side of cell walls in the middle cortex, and mainly in the matrix filling the intercellular spaces of the middle and inner cortex. This main site of localization of MAC236 may indicate a role in the functioning of the oxygen diffusion barrier.

Immunolocalization of alkaloids in lupin seeds (Lupinus spp.) has been performed by cryofixation and conventional methods. Alkaloids were localized in the protein bodies of the cotyledon ceils. Some immunogold particles in the walls of these cells were also observed. There were no differences in the sites of localization between the two mentioned methods. X-ray microanalysis of elements showed the presence of R Mg, S, and K in the protein bodies of cotyledon cells in lupin seeds. The role of K + in alkaloids transport is discussed.

The effect of boron deficiency on symbiotic nitrogen fixation in pea (Pisum sativum) was examined. The absence of boron in the culture medium resulted in a decrease of the number of nodules and an alteration of nodule development leading to an inhibition of nitrogenase activity. Examination of boron-deficient nodules showed dramatic changes in cell walls and in both peribacteroid and infection thread membranes, suggesting a role for boron in the stability of these structures. These results indicate that boron is a requirement for normal nodule development and functionality.

La presente revisión es un resumen de algunos trabajos originales realizados en nuestro Departamento para el estudio de diferentes aspectos de esta peculiar simbiosis, siempre integrando estructura y función. La primera parte está dedicada a un estudio estructural llevado a cabo con el fin de conocer los mecanismos de infección, que difieren de la mayoría de las leguminosas estudiadas. Para este fin se aplicaron técnicas de immunofluorescencia con la green fluorescent protein (GPT), técnicas de inmunocitoquímica y de hibridación in situ. Mediante estos estudios se ha observado que la infección se realiza por vía intercelular. Junto al estudio estructural se describen algunos componentes enzimáticos del nódulo, MAPKinasas (Mitogen-activated protein kinase) y aldehído oxidasa, presentes en estadios tempranos del reconocimiento bacteria-planta. Se muestran los resultados de producción de aceituna y rendimiento de aceite de 3 campañas consecutivas (2011/12-2013/14) en un olivar en secano de la variedad Cornicabra en la Comunidad de Madrid, con el empleo de 3 cubiertas vegetales (cebada, esparceta y Brahcypodium) y un control. También se presentan los resultados de calidad de aceite (polifenoles totales, pigmentos y color) de la última campaña para cada uno de los tratamientos. En ninguna de las campañas estudiadas la producción de aceituna o el rendimiento de aceite ha sido diferente entre tratamientos. Sí se han encontrado diferencias estadísticamente significativas en los aceites elaborados en la última campaña, con un mayor contenido en polifenoles y pigmentos totales para el tratamiento Brachypodium, estando los aceites de la cubierta cebada en el lado opuesto.
