Abstract

The main compound of Uncaria gambir Roxb. (gambir), catechin and it’s derivates have been believed to be potential as antiviral. Epigallocatechin gallate (EGCG) and epicatechin are catechin derivates which are found to be potential as antiviral against Human Immunodeficiency Virus (HIV). However, gambir extract also contains quercetin that has possibility to be mutagenic. Therefore, a preliminary study towards safety of those compounds within gambir extract, mutagenicity assay using Ames Method has been performed.Sample (gambir extract) was obtained from West Sumatera, Indonesia. The extract was characterized according to Farmakope Herbal Indonesia and WHO methods. Mutagenicity test by Ames method utilized a colorimetric microplate in 6 various concentration (125 mg/mL; 62.5 mg/mL; 31.25 mg/mL; 15.625 mg/mL; 7.81 mg/mL dan 3.91 mg/mL) against mutant bacteria Salmonella typhimurium TA 98, Salmonella typhimurium TA 100 and Escherichia coli WP2 uvrA with and without the addition of S-9 enzyme. Extract of gambir in this study contains 86.60% of catechin, 12.92% moisture content, 22.49% water-soluble extract content, 80.63% ethanol-soluble extract content, 0.81% total ash, 0.32% acid insoluble ash content and 10.38% in dryness level. From the mutagenicity test and calculation, fold increase (over baseline) of the sample in 6 various concentration with and without adding S-9 enzyme are lower than 2. Gambir extract from West Sumatra with catechin contains 86.6% hasn’t showed mutagenic effect due to the fold increase (over baseline) of mutagenicity test lower than 2.

Abstract

Amaranthus tricolor L. was reported as source of carotenoid compounds (tetraterpenoids) which is a natural pigments and has bioactivity as antioxidants. They are beneficial to health. Carotenoids isolate are obtained by maceration method in methanol, saponification with KOH / MeOH 5%, fractionated by vacuum column chromatography, preparative thin layer chromatography (preparative-TLC) repeatedly and each fraction monitored using thin layer chromatography (TLC). Structure analysis of carotenoids based on UV, IR and GCMS spectroscopic data showed similarities as lutein structure.

Abstract

Duku (Lansium domesticum Corr var duku) is a seasonal native plant from South Sumatra and Jambi. Peel of Duku is useful as botanical insecticide. in South Sumatra and Jambi. The research proves that the skin duku can be useful as an insecticide plant. Quality of compounds in plants can be affected by the origin area of plant, body parts of plants, and extract characters. This study aims to characterize the simplicia and duku peel extract from South Sumatra and Jambi Province. The sample were collected from Simpang Agung Village, OKU Selatan District and Rengas Bandung Village, Muaro Jambi District. The peel was extracted using acetone, while the charactherization based on standardization method from Badan POM Indonesia. Non-Specific parameters which measured were water content, water soluble ash and acid insoluble ash. Specific parameters which measured were water soluble extract, ethanol soluble extract, chromatogram, and chemical compound. Simplicia from Simpang Agung Village contain lower water content, water soluble ash, and acid insoluble ash than simplicia from Rengas Bandung Village. Chromatogram pattern of the peel extract from both village were similar and contain terpenoid and fenolic compound. However hexane yield extract from Simpang Agung Village higher than extract from Rengas Bandung Village therefore contain more insecticides material (terpenoid).

Abstract

Star fruit (Averrhoa carambola L.) is potential as raw material for medicine, native in tropic areas, including Indonesia. According to other study report, star fruit leaves containing flavonoids apigenin and quercetin as potential anti-inflammatory and anticancer agents. The raw material for the drug in Indonesia mostly obtained through imports from other countries. In order to support the independence of traditional medicine raw materials, it is important to standardize the quality of traditional medicine raw materials, in this case is star fruit leaves by High Performance Liquid Chromatography (HPLC) method. The sample used is star fruit leaves extract obtained from maceration process using ethanol 70%; water fraction, ethyl acetate and hexane fractions obtained from fractionation process of the ethanolic extract. Physical parameters analyzed in sample include appearance, color, odor, taste, extract yield, water content, loss of drying, total ash content, residual solvent. Chemical parameters analyzed include apigenin and quercetin contents. The results shows that star fruit leaves used in this study meet the standards of Indonesian Herbal Pharmacopoeia with highest apigenin (6.37%) and quercetin (4.49%) content are in ethyl acetate fraction.

Abstract

Java turmeric (Curcuma xanthorrhiza Roxb.) is a native plant of Indonesia which has been proved as anti-inflamation, antioxidant, antimicrobial, antitumor, hepatoprotective and antihyperlipidemic agent. The bitter taste with pungent aroma of java turmeric, improper storage that decreasing essential oil and curcuminoid level can be prevented by coating java turmeric as microcapsule. Recently, many microcapsule formulation is made based on organic solvent which have high risk on toxicity and also flammable. The study aimed to produce microcapsule using water based coating material from mixture of tapioca starch and carboxymethylceIlulose (CMC). There are 3 formulas of coating material that were used in this study, with proportion (w/v) of tapioca starch and CMC in formula I (3% : 1 %), formula II (2% : 2 %), and formula III (1% : 3%). The results showed that microcapsule with coating material made from formula III was the best, regarding to the measurements of physical parameter (recovery, water content, absorption efficiency, flow rate and particle size) and chemical parameter (curcumin content) compared to formula I and II. The higher CMC concentration on coating material compotition, the better microcapsule will be produced.

Abstract

Garcinia tetranda Pierre (Clusiaceae) is a plant that is widely used to treat various diseases and its chemical compounds has not been much studied. Compound isolation using the three methods of chromatography (Vacuum Liquid Chromatography Column (VLCC), preparative Thin Layer Chromatography (pTLC) and TLC) generate novel compound 5,7,4 ', 5'', 7'', 3''', 4 '''-heptahidroksi-2'''-methoxy-flavanon [ 3,8 ]-flavone'' which is different from the compounds that had been found previously in Garcinia tetranda Pierre. Structure identification was conducted using UV, IR and NMR spectroscopic data and by comparing the 13C and 1H-NMR data of the isolate compound and known biflavonoid compounds. Biogenesis pathway analysis of the new compound showing relationship with molecular compounds which have previously been found in Garcinia. Antibacterial test of the novel compound using Kirby-Bauer agar diffusion methode against Gram-negative bacteria Salmonella typhi showed that the compound is not potentially used as antibacterial agent compared to chloramphenicol.
