Abstract

The activity of ?-glucosidase inhibition test of local granular green tea (Camellia sinensis) grade of Arraca Yabukita has been done. Granular green tea obtained by vacuum drying of the concentrate of the diafiltration process using nanofiltration membrane (DF-NF) at 50C for 0-80 hours. Every 8 hours of drying time, the analysis of water content, total polyphenols, and ?-glucosidase inhibitory activity of granular green tea were conducted. The?-glucosidase inhibitory activity test was performed by by Kim Yong-Mu method and compared to quercetin as standard. The results showed that, the drying process increases the ?-glucosidase inhibitory activity of granular green tea. At the optimum drying time 72 hours, the ?-glucosidase inhibitory activity of granular green tea is about 18 times higher than quercetin.

Abstract

Antioxidant and toxicity of Puspa (Schima wallichii) were studied. The plant extracts were prepared for bio-logical screening. Antioxidant activity of extracts was analysed by 1,1-diphenyl-2-picrylhydrazyl (DPPH) as stable free radical, while toxicity of extracts was analysed by Brine Shrimp Lethality Test (BSLT) method. The result showed that ethyl acetate, methanol and butanol extracts were very active as antioxidant, as indicated by IC50 value of 11.44, 17.97 and 12.55 µg/mL for DPPH inhibition, respectively. Whilst n-hexane extract was not active as antioxidant indicated by IC50 value of 257.91 µg/mL, but aquadest extract was less active as antioxi-dant indicated by IC50 value of 171.94 µg/mL. The toxicity test result showed that all extracts were active against Artemia salina Leach with LC50 value of 107.35 ? 902 µg/mL. Furthermore, isolation of individual bio-active compound of ethyl acetate extract that most active as antioxidant was carried out using column chroma-tography followed by preparative Thin Layer Chromatography (TLC). The active isolate was then identified using TLC, Fourier Transform Infra-Red (FTIR) and Liquid Chromatography Mass Spectroscopy (LCMS).. The identification result showed that the major compound contained a putative compound known as theanine with MS 174.26 g/mol. The isolate was very active as antioxidant with IC50 value of 48.04 µg/mL, although it was less active than the extract. The isolate was also toxic with LC50 value of 27.71 µg/mL so that puspa leaf potentially to be developed as a natural antioxidant and anticancer agent for preventing of human diseases.

Abstract

Background: Increased lipase activity can increase the amount of monoglycerides and fatty acids absorbed by the body, this is what affects obesity. If pancreatic lipase activity is inhibited, the production of fatty acids will decrease, consequently the level of fat in the blood will also decrease.Objective: To study anti obesity potential of a single herb or a mixture of green tea and other herbs (Syzygium polyanthum. Lv, Artocarpus communis.Lv, Cinnamomum verum..Lv, Manilkara zapota.Lv, Tectona grandis.Lv) by the in vitro inhibition assay of the lipase enzyme activityMethod: A single (8 herbs) or a mixture of three herbs powder (12 formulas) was put into a tea bag with a total weight of 1.5 g to be brewed in 100 mL bottled mineral water at 70⁰-90⁰C for 10 minutes. In vitro lipase enzyme inhibition assay were conducted to measure the ability of these samples as lipase inhibitor. Orlistat used as positive lipase inhibitor. Results: There was a difference results of lipase inhibition activity between the sample of a single and a mixture of herbs. Interestingly, the results shows that formula 3 (mixture of green tea, Manilkara zapota and cinnamon). has 53.942 % inhibition, formula 9 (a mixture of green tea, Syzygium polyanthum and cinnamon) has 67.322 % inhibition and formula 12 (a mixture of green tea, Tectona grandis and cinnamon) has 56.612 % inhibition which close to lipase inhibitory activity by standard Orlistat.Conclusion: The mixture of green tea and two other herbs has a lipase inhibitory activity similar to Orlistat, when compared to the single herb. The highest lipase inhibitory activity is found in formula 9.

Abstract

Preparation process of 2-aminoethylsulfonic acid (taurine) from ethanolamine, sulfuric acid and sodium sulfite has been studied. The process involves two steps of reactions, the first was esterification of ethanolamine (H2N-CH2-CH2-OH) with sulfuric acid to produce the intermediate product of 2-aminoethyl ester (H2N-CH2-CH2-OSO3H) which then was extended to the second step by sulfonation with sodium sulfite to produce 2-aminoethylsulfonic acid (H2N-CH2-CH2- SO3H). These two process conditions were observed by varying mole ratio of reactants, temperature and time period of reactions. Taurine product was qualitatively analyzed using 1 H-NMR and LC-MS. Physical-chemical analysis were done by observing its melting point and determining its water, chloride and sulfate contents. Its melting point, water content, and sulfate content were 290o C, 0.303%, and 3 ppm, respectively, while its chloride content was undetected. After purification, the yield of process was 25.57%.

Abstract

Diabetes Mellitus (DM) is one of the non communicable diseases (NCDs) with the largest number of deaths in the world. The Macaranga hispida (Blume) Mull. Arg is a source of phenolic compounds. Phenolic is grouped as polyphenols group that widely functioned as antidiabetic. The α-glucosidase enzyme plays a role in the forming of glycoproteins and glycolipids. The α-glucosidase enzyme can be used for antidiabetic activity assay because of its capability on breaking down carbohydrate into glucose in the human small intestine. This study aimed to determine phenolic content, total flavonoid content, and antidiabetic activity of M. hispida leaves extract and its fraction. The extract and fraction were tested for the phenolic levels using Folin-Ciocalteau method, the total flavonoid using alumunium chloride method, and the antidiabetic activity using α-glukosidase method. The yield of methanolic extract of the Macaranga hispida was 10.23%. Phenolic content of the ethyl acetate fraction from the methanolic extract was 8.411 mg, equivalent to gallic acid/100 mg extract. Total flavonoid content was 6.14 ± 0.31b/b μg/mL. Antidiabetic activity assay of the extract and the fractions of Macaranga hispida resulted the highest antidiabetic activity (IC50) was in the ethyl acetate fraction with IC50 value 21.91 μg / mL.
