Abstract

The research about the effect of lime (Citrus aurantifolia (chrism.) Swing) to levels of  triglycerides and body weight of male rats (Rattus norvegicus)  wistar strain at a doses of 1.5, 3 and 4.5 ml/kg body weight has done. This study conducted on five groups consisted of norml control , negative control (-),  dose of 1.5; 3 and 4 ml/kg body weight,  each group consisted of  5 rats , The normal control group  were given regular feed, control (-) was given 2 ml cow's brain and regular feed then the test group were given lime each with a dose of 1.5, 3 and 4.5 ml / kg body weight was added with 2 ml of cow's brain and regular feed. The method of measurement used is Enzymatic Colorimetric Test GPO (Glycerol-3-Phosphate Oxidase) at a wavelength of 546 nm. The results showed that administration of  lime doses of 3 and 4.5 ml/kg body weight have a significant difference (p <0,05) to the control (-). However, at a dose of 4.5 ml/kg body weight more effectively and more quickly lowering triglyceride levels because the first measurement time of the second measurement time already showed a significant difference (p<0,05) .

Abstract

Betel nut (Areca catechu L.) is a natural material that has antibacterial activity in treating diseased on teeth, halitosis and can inhibit of dental caries. The objective of this research is type of bacteria based on morphology, the results of bacterial identification tests on the tongue and to determine the effect of the inhibitory power of betel nut ethanol extract (Areca catechu L.) to bacteria of the tongue. Antibacterial activity testing using disc diffusion method with various concentrations of 10%, 20% and 30%. Secondary metabolite contents are alkaloid, terpenoid and flavonoid.The results obtained from the identification test based on the color of bacteria Branhamella catarrhalis, Staphylococcus epidermidis and Straphylococcus aureus. Ethanol extract of areca nut (Areca catechu L.) is better to inhibiting the growth of Staphylococcus epidermidis and Staphylococcus aureus compared to Branhamella catarrhalis. Based on Two Way Anova statistical data on inhibitory zone diameters, there were differences between the concentration groups of 10%, 20% and 30% and there were significant differences in the inhibitory activity of Branhamella catarrhalis bacteria compared to Staphylococcus epidermidis and Straphylococcus aureus bacteria but not significantly different between Staphylococcus epidermidis and Straphylococcus aureus bacteria

Abstract

The present investigation aims to evaluate the antihypercholesterolemic potential of Uncaria cordata (Lour.) Merr. as an ethanolic extract in diet-induced hypercholesterolemic male white mice model. In this study, white mice were segregated into 6 groups; all the groups except the normal control group were given a high-fat diet to induce hypercholesterolemia. After induction of cholesterolemia, normal and negative control groups were treated with NaCMC, the positive control group was treated with atorvastatin, and the remaining three groups received ethanolic extract Uncaria cordata (Lour.) Merr. in three doses (100, 200, and 400 mg/Kg BW) for a treatment period of 29 days. Measurement of cholesterol levels was performed on days 0, 15, 22, and 29 using EasyTouch® GCU digital devices. The results were analyzed by one way ANOVA test and ANOVA Repeated test. The results showed that the ethanol extract of the Uncaria cordata (Lour.) Merr. root plant dose 100, 200, and 400 mg/Kg BW effect in lowering total cholesterol in male white mice significantly (P <0,05). Ethanol extract from Uncaria cordata (Lour.) Merr. a dose of 200 mg/Kg BW showed a better decrease in cholesterol levels on day 29 compared to day 22.

Abstract

BACKGROUND: Adipose tissue expansion in obesity leads to abnormal adipocyte function, chronic low grade inflammation, primary reticulum stress, and mitochondrial stress. This induces mitochondrial unfolded protein response (UPRmt) that selectively upregulates mitochondrial chaperone protein. Heat shock Protein 60 (HSP60) is the primary chaperone in mitochondrial matrix. Inflammatory stress promotes HSP60 released from adipocytes and induces insulin resistance. In this study we attempted to investigate the correlation between exHSP60 and HOMA-IR in men with different ranges of waist circumference (WC).METHODS: This study was an observational cross sectional study carried out on 141 non diabetic men, aged 30-55 years old, who were divided into three groups based on WC; WC ≤90 cm, 90 cm < WC ≤ 100 cm and WC > 100 cm. Fasting plasma glucose, triglyceride, HDL-C, hsCRP, HSP60 serum and anti-HSP60 antibody, serum IL-1β serum, insulin, were examined. For statistical analysis, Kolmogorov-Smirnov and Spearman’s correlation analysis were conducted.RESULTS: There were a significant correlation between exHSP60 and HOMA-IR (r=0.281; p=0.041) in WC ≤ 90 cm group; and a negative significant correlation between exHSP60 and HOMA-IR (r=-0.508; p=0.007) in WC > 100 cm group.CONCLUSION: This study showed that there was a dynamic correlation between exHSP60 and HOMA-IR in WC ≤ 90 cm group compared with WC > 100 cm group. We also found inverse correlation patterns between exHSP60 and HOMA-IR, and between anti-HSP60 antibody and HOMA-IR in non diabetic subjects.KEYWORDS: obesity, insulin resistance, mitochondrial stress, exHSP60, hsCRP, HOMA-IR

Abstract

BACKGROUND: Several researches reported that inflammatory and immunological mechanism such as autoantibody to β2-glycoprotein I (anti β2GPI) appear as related factors in initiation and progress of atherosclerosis lesion in patient with autoimmune disease. Antibody to β2GPI titers are correlated with atherosclerosis and in vitro studies showed that they enhance oxidized low density lipoprotein (ox-LDL) uptake by macrophages. Immunization with auto-antigen β2GPI elicits an immune response to influence lesion progression that mostly happens in autoimmune subjects. The metabolic syndrome (MetS) is combination of several metabolic disorders such as obesity, dyslipidemia, Diabetes Mellitus (DM) and conditions due to inflammation and stress oxidative. The Correlation between inflammatory markers such as High sensitivity C-Reactive Protein (hsCRP) and anti-β2GPI antibody in MetS needs to be further investigated.METHODS: This was an observational study with cross sectional design on subject with MetS as determined by the International Diabetes Federation (IDF) 2005’s criteria.RESULTS:There was a positive and significant correlation between hsCRP and anti-β2GPI antibody in MetS group (r=0.406; p≤0.05) as compared to non-MetS group. We found that there was elevated level of anti-β2GPI antibody in hsCRP of 3-10 mg/L.CONCLUSIONS: Anti-β2GPI antibody may be elevated in subjects with MetS who have low grade of inflammation as shown by hsCRP.KEYWORDS: metabolic syndrome, inflammation, autoantigen, atherosclerosis, obesity

Abstract

Uncaria cordata is a plant that is used to treat diabetes and has antioxidant properties. Research has been carried out on Acute and Delayed Toxicity Tests of Butanol Fraction of Leaves of Kaik-Kaik Root Plant (Uncaria cordata (Lour) Merr.) on Male White Mice (Mus musculus L. This study aims to determine the effect of acute toxicity of the butanol fraction of the leaves of kaik-kaik root by determining the LD50 value. Observations made in this test were the number of dead animals, toxic symptoms, and observations of the organs of the test animals. Observations were made for 24 hours to determine the LD50 value to 14 days after administration of the test preparation. In the observation of toxic symptoms, there were no tremors, seizures, salivation, diarrhea, lethargy, weakness, sleep, and coma in each experimental animal. In the relative weight parameters of organs consisting of the heart, liver and kidneys, there was no effect of this fraction, and there was no change when compared to the control group. The results showed LD50 values > 5000 mg/kgBB are considered relatively harmless and potentially as standardized herbal medicine

Abstract

Areca nut is traditionally used by the community to strengthen teeth. Areca nuts contain alkaloid, flavonoid, and terpenoid as antibacterials. This study aims to determine the types of dental bacteria resulting from isolates and determine the effect of the administration of ethanol extract of areca nut (Areca catechu L.) on the results of isolates of dental bacteria. Antibacterial activity testing using disc diffusion method with the concentration of areca nut ethanol extract 10%, 20% and 30%. The results showed that bacteria of the tooth are Lactobacillus casei, Branhamella catarrhalis, Streptococcus sp. Ethanol extract of areca nut (Areca catechu L.) is better to inhibit the growth of Lactobacillus casei compared to Branhamella catarrhalis and Streptococcus sp in the medium to weak category. Two Way ANOVA test results on inhibitory zone diameter stated that there were differences between the concentration groups of 10%, 20%, 30%, positive control and negative controls and there were differences in the inhibitory activity of Lactobacillus casei compared to Branhamella catarrhalis and Streptococcus sp but there were no significant differences on the inhibitory activity of Branhamella catarrhalis with Streptococcus sp.

Abstract

Chalcones are compounds derived from nature, it is show the anticancer activity. The aim of thisstudy is to determine the binding free energy (affi nity) and interaction of chalcone derivatives as inhibitors for A549 lung cancer cells using 4HFZ protein as receptors. This study used 10 chalcone derivatives which were selected with three lowest IC50 values, fi ve medium IC50 values and two with higher IC50 values (i.e. more than 100 μg/mL) and 4HFZ protein was used as receptor. The structure of the chalcone derivative is transformed in 3D structure, then the docking process of chalcone compounds with amino acids on the MDM2 receptor (4HFZ) is carried out. The simulation results shown that data in the form of binding free energy value (kcal / mol) shown the stability of ligand interaction of chalcone compounds on amino acids on MDM2 receptors. The docking results shown that, compound 2 is able to bind strongly with MDM2 receptor (4HFZ) and it is also stable because it has a low binding-free energy value of -7.2 kcal / mol, with a RMSD value of 0,000. Construction of some hydrogen bonds is the same as active receptor (gefi tinib), they are Leu54 on the NH2.

Abstract

Hiperkolesterolemia merupakan peningkatan kadar kolesterol dalam darah. Kelainan kadar lemak bukanlah suatu penyakit, tetapimerupakan faktor risiko bagi penyakit lainnya terutama penyakit jantung dan pembuluh darah. Berat badan adalah ukuran yang lazim atausering dipakai untuk menilai keadaan suatu gizi manusia. Penelitian ini bertujuan untuk mengetahui apakah ada penurunan kadar kolesteroldarah dan berat badan pada mencit putih jantan setelah pemberian sari buah jeruk nipis. Metode yang digunakan adalah cholesterol metertest. Dari hasil penelitianmenunjukkan hasil kadar kolesterol yang signifikan (p<0,05). Pada Post Hoc Testterdapat perbedaan signifikan<0,05 antara T0 dengan T1, antara T2 dengan T3 dan T4 tetapi tidak terdapat perbedaan antara T0 dengan T3 dan T4 dan juga tidak adaperbedaan yang signifikan antara T3 dan T4. Pada hasil berat badan menunjukkan ada perbedaan yang signifikan antara T0 dengan T1 daT2, tidak terdapat perbedaan antara T3 dengan T4. Hasil dari analisis data dapat disimpulkan bahwa sari buah jeruk nipis dapat menurunkankadar kolesterol dan berat badan secara signifikan pada mencit putih jantan, tetapi secara signifikan tidak terjadi perbedaan yang bermaknaantara sari buah jeruk nipis 1,7 ml dan 2 ml.
