FFBA is a fraction that contains flavonoids and the andrographolide compounds has been removed. The antioxidant activity of FFBA was tested using the method of reducing free radicals from DPPH (1,1-diphenyl-2-pikrilhidrazil) and using quercetin as a standard. The results showed that FFBA has antioxidant activity with strong activity with IC50 value of 88.98 μg / mL while quercetin has a very strong activity with IC50 value of 3.42 μg / mL.Key Words : antioxidant, Andrographis paniculata, DPPH, flavonoidABSTRAKPenelitian ini bertujuan untuk mengetahui aktivitas antioksidan pada fraksi flavonoid bebas andrografolid (FFBA) dari herba sambiloto (Andrographis paniculata). FFBA merupakan fraksi yang mengandung flavonoid dan sudah dihilangkan kandungan senyawa andrografolid. Aktivitas antioksidan FFBA diuji dengan menggunakan metode peredaman radikal bebas dari DPPH (1,1-diphenyl-2-pikrilhidrazil) dengan baku pembanding kuersetin. Hasil penelitian menunjukkan bahwa FFBA memiliki aktivitas antioksidan dengan aktivitas yang kuat yaitu dengan nilai IC50sebesar88.98 μg/mL sedangkan kuersetin memiliki aktivitas yang sangat kuat yaitu dengan nilai sebesar 3,42 μg/mL.Kata kunci :antioksidan, Andrographis paniculata, DPPH, flavonoid

Abstract

Acetaminophen/parasetamol adalah obat analgetik-antipiretik yang populer dan banyak digunakan. Sediaan acetaminophen dalam bentuk tablet, selain dengan nama generik juga tersedia dengan nama dagang. Tujuan penelitian ini adalah untuk mengetahui ekivalensi in vitro terhadap mutu produk dagang dan generik berlogo tablet acetaminophen yang beredar di pasaran. Pada penelitian ini dilakukan uji disolusi terbanding acetaminophen produk obat generik berlogo dan produk bermerek dalam media disolusi berupa larutan dapar phosphat pH 5,8. Penentuan laju disolusi acetaminophen dilakukan menggunakan metode paddle (dayung) dengan kecepatan pengadukan 50 rpm, pada suhu 370C ± 0,50C. Sampel cuplikan diambil pada menit ke 5, 10, 15, 20, 30 dan 45. Parameter uji yang diamati adalah kadar obat terlarut pada saat t=30 menit (C30) dengan parameter standar baku C30 menunjukkan hasil tidak boleh kurang dari 80% kadar obat. Hasil penelitian didapatkan bahwa disolusi tablet acetaminophen masing-masing produk baik obat generik berlogo maupun produk bermerek menunjukkan gambaran profil disolusi yang berbeda. Dari 8 sampel diperoleh 5 produk paten dan 3 produk generik semuanya menunjukkan hasil disolusi dengan nilai C30 yang memenuhi syarat (>80)

Abstract

The acute toxicity of Jatropa curcas leaves on Balb/C male mice was studied in rats. This research aimed to determine acute toxicity, evaluate spectrum of toxic effect and mechanism that caused the death of animal test after administration of ethanolic extract of J. curcas leaves, single dosage orally on 24 hours observation. The research used male mice, which are divided into 5 groups. Group I was negative control with CMC-Na. Group II, III, IV, and V were given extract with dose of 1400 mg/kgBW, 2240 mg/kgBW, 3584 mg/kgBW and 5734 mg/kgBW, respectively. Evaluation of the toxic symptoms and death of animal test was done for 24 hours. If the animal test was died before 24 hours then it underwent surgery to take the heart, liver, lung, and kidney. In the end of the evaluation, all mice were killed to take the vital organs for histopathologic examination. No mortality was observed during study. The test resulted LD50 of ethanolic extract from J. curcas leaves using Balb/C male mice was 5734 mg/kg of BW. It was categorized as practically not toxic. Administration of the extract did not cause alterations of animal behaviours. Histopathology examination shows inflammation in lung, liver, and kidney after administration of the extract.

Abstract

In previous studies, Zingiber officinale, Piper retrofractum, and the combination showed cytotoxic activity, induced apoptosis, and p53 expression of HeLa, T47D, and MCF-7 cell lines. This study was conducted to investigate the cytotoxic and apoptotic activity of Zingiber officinale (ZO), Piper retrofractum (PR), and the combination as well as their effect to p53 expression on Myeloma and WiDr cells. The powder of ZO, PR, and ZO + PR combination (1:1) were macerated with 96% ethanol for 3 x 24 hours. MTT cytotoxic assay was performed on Myeloma and WiDr cell lines. Apoptotic cells were stained with ethidium bromide and acridine orange. Imunohistochemical expression of p53 was examined on Myeloma and WiDr cell lines. Doxorubicin was used as positive control in all assays. Results showed that ZO, PR, and ZO + PR combination had cytotoxic activity on Myeloma cells with IC50 of 28, 36, and 55 mg/ml respectively and WiDr cell lines with IC50 of 74, 158, and 64 mg/ml respectively, induced apoptotic activity, and increased p53 expression on Myeloma and WiDr cells. These results suggest that ZO, PR, and their combination induced Myeloma and WiDr cells in apoptosis through p53 expression.

Abstract

Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan herba Andrographispaniculata (sambiloto). Aktivitas antioksidan ekstrak air herba sambiloto dan fraksi-fraksinya diujidengan menggunakan metode peredaman radikal bebas DPPH (Dipheny-1-picrylhydrazyl). Dari hasilpenelitian diketahui bahwa IC50 ekstrak air, fraksi kloroform, fraksi etil asetat dan residu ekstrakherba sambiloto berturut-turut adalah 1036,76l; 820,15; 395,01; dan 1080,60 μg/ml. Hasil penapisanfitokimia menunjukkan adanya senyawa golongan flavonoid.

Abstract

BACKGROUNDJatropha curcas leaves have been proven to be anti-inflammatory andantioxidant. In this study we examined the antiarthritic effects of ethanolicextract of J. curcas leaves using adjuvant induced arthritis (AIA) in rats.METHODSMale Wistar rats were divided into 6 groups (n=8), consisting of normal group (0.9% NaCl), control group (complete Freund’s adjuvant/CFA 1 mg/ml), sodium diclofenac group at a dose of 6.75 mg/kg (p.o), ethanolic extract of J.curcas groups at doses of 150 mg/kg (p.o), 300 mg/kg (p.o) and 600 mg/kg (p.o). Each group was induced by 0.2 ml CFA on day 1 and a booster injection on day.5. Extracts of J. curcas were administered on days 14-28. Arthritic scores weredetermined, then analyzed using Kruskal Wallis followed by Mann Whitneytests. Mobility scores were analyzed using one way analysis of variance, followed by least significant difference multiple comparison test. Arthritic jointhistopathology was observed on day 29.RESULTSThe results showed that the ethanolic extract of J. curcas leaves at doses of150 mg/kg, 300 mg/kg and 600 mg/kg significantly reduced arthritis scores(p<0.05) compared to control group (CFA). The J. curcas leaf extract at doses of 150 and 300 mg/kg BW decreased mobility scores. Histopathology studies showed that the J. curcas extract reduced edema and cartilage destruction in arthritic joints.CONCLUSIONSThe J. curcas leaf extract had anti-arthritic effects by reducing arthritis scoresand mobility scores. The extract should be further examined as a potentialcandidate for anti-arthritic therapies.

Abstract

Ethanolic extract of mango’s leaves variety of bapang (Mangifera indica L. ‘Bapang’) has been known to contains mangiferin which has antidiabetic activity in patients with diabetes mellitus. This study aimed to obtain a formulation of tablets of ethanolic extract mango leaves with variety of bapang with good physical characteristic and know the effect of variations concentration of guar gum on the properties of mucoadhesive. Tablets were prepared using wet granulation method varying levels of guar gum as a bioadhesive matrix. Tablets were tested physical characteristics and mucoadhesive strength in vitro by using a rotating cylinder and observed within 8 hours. The results showed that the tablets of ethanolic extract mango leaves with variety of bapang have good physical characteristics. The result of mucoadhesive in vitro test showed that all formulas have good mucoadhesive strength. Mucoadhesive strength increased with the increase in the concentration of guar gum. The result of this research showed that the tablets of formula 4 with the highest concentration of guar gum showed the best mucoadhesive strength where they could be attach of 7 ± 1 hour.

Abstract

Konsumsi minuman tradisional, secara empirik maupun ilmiah bermanfaat untuk kecantikan, kesehatan dan perawatan badan.Masyarakat modern saat ini mulai bijak dan cenderung kembali ke alam dalam memilih makanan atau minuman yang dikonsumsi. Kegiatan ini bermitra dengan produsen minuman kesehatan tradisional yaitu IKM Giri Slamet Herbal. Tujuan kegiatan ini untuk (1)meningkatkan pengetahuan mengenai teknik pasca panen bahan baku dan proses produksi agar produk yang dihasilkan berkualitas berdasar kandungan komponen bioaktif (2) memberi informasi teknik menentukan masa kadaluarsa (3) inovasi produk baru sediaan sirup minuman rempah. Metode pelaksanaan adalah dengan melakukan sosialisasi dan workshop  tentang metode pasca panen dan metode proses produksi, membuat sediaan sirup rempah yang berkualitas serta penentuan masa kadaluarsa. Pendampingan program ini dilakukan untuk memastikan para perajin dapat melaksanakan informasi ilmu pengetahuan, teknologi dan seni (ipteks) yang diberikan oleh tim pengusul dan untuk menjamin keberlanjutan program. Hasil dari pelaksanaan kegiatan ini adalah perajin mengadopsi ipteks yang diberikan oleh tim pengusul. Perajin mengetahui metode pasca panen, metode proses produksi produk baru yaitu sirup rempah sebagai minuman tradisional dan dapat menentukan masa kadaluarsa.

Abstract

Breast cancer is one of the main causes of death in women. Cancer treatment with surgery, chemotherapy, and radiology often cause undesirable side effects. Therefore, alternative cancer treatment by using plants as traditional medicine was expected to reduce side effects. Nigella sativa is one of the plants used as anticancer empirically. This study conducted to examine the cytotoxic activity of Nigella sativa seeds and identify its components on T47D breast cancer cells. Petroleum ether, chloroform, ethyl acetate, and ethanol were used to extract N. sativa seeds. The extracts were tested their cytotoxic activity on T47D cell line using MTT method. The active compound was separated using column chromatography. Cytotoxic test on T47D cell line was perform for extracts of each separation stage. Data were analyzed by probit analysis to obtain IC50 values. Components identification was performed using GC-MS analysis. The results showed that chloroform extract has cytotoxic activity better than other extracts with IC50 of 124.206 µg/mL. The third fraction has cytotoxic activity better than other fractions with IC50 of 68.568 µg/mL. The GC-MS analysis showed that in the third fraction of the chloroform extract contain linoleat acid, the major compound and tryptamine.
