Abstract

BACKGROUND Guava is an herbal with proven antioxidant and anti-inflammatory properties. The aim of this study was to investigate the anti-arthritic activity of the ethanol extract of Psidium gujava leaves (EEPG) against complete Freund’s adjuvant (CFA) induced arthritis in rats. METHODSAn experimental study was conducted on 40 male Wistar Sprague Dawley rats, which were divided into 5 groups. Each group was induced with 0.2 mL CFA (1 mg/mL) on day 1 and 0.1 CFA mL booster injection on day 5. Group I served as an arthritic control, group II received dexamethasone (6.75 mg.kg-1 orally), group III, IV and V received EEPG at oral doses of 250, 500, and 750 mg/kg BW, respectively, on days 14 to 28. Anti-arthritic activity was observed from the arthritis score, the paw circumference was measured on days 0, 1, 4, 8, 12, 16, 20, 24, and 28, the mobility score was determined on days 12 and 28, and the histolopathology of the knee joint was examined on day 29. RESULTSEthanol extract of Psidium guajava leaves significantly suppressed the swelling of the paws in chronic phase based on increasing of edema (%), while starting on day 20. EEPG at 250 mg/kg was most effective in significantly reducing arthritis scores (p<0.05). Histopathological examination showed repair of the knee joint synovial membrane and cartilage. CONCLUSIONSPsidium guajava leaf extract is effective in decreasing the inflammatory response and arthritic symptoms in rats with adjuvant-induced arthritis. Psidium guajava leaves can be developed into an alternative anti-arthritis treatment.

Abstract

Cocor bebek (Bryophyllum pinnatum) has been proven as analgesics for relief of acute pain. Flavonoid is one of the compounds in Bryophyllum pinnatum which is potentially developed as analgesics for the treatment of chronic inflammatory pain. This study was conducted to determine the effect of ethanolic leaf extract of Bryophyllum pinnatum as attenuation of thermal hyperalgesia in rats with chronic inflammatory pain. This study used a completely randomized design with complete Freund’s Adjuvant/CFAinduced chronic inflammatory pain in wistar rats, at the first day. Thirty rats were divided into 5 (five) groups (n = 6), consisting of a positive control (ketamine 10 mg/kg bw), negative control (CMC Na 0.5%), and three treatment groups of ethanolic leaf extract of Bryophyllum pinnatum with doses of 200, 400 and 600 mg/kg bw. The extract was given twice a week for four weeks,at day-3 and day-6. The effect of the treatment on the attenuation of thermal hyperalgesia was investigated by using hot-plate at day-7, day-14, day-21 and day-28 to measure latency and subsequently, the percentage of the increase in latency was calculated. Analysis of Variance (ANOVA) was employed to analyze the data and continued by Least Significance Different test (LSD). The ethanolic leaf extracts of Bryophyllum pinnatum at the doses of 200, 400, and 600 mg/kg bwhad effect of the attenuation of thermal hyperalgesia by increasing latency by 3.18, 4.33 and 6.12 seconds, respectively. The optimum dosage as antihiperalgesia was 600 mg/kg bw, with latency was increased to twice than those of negative control group.

Abstract

Permasalahan kurangnya pengetahuan tentang penggunaan obat pada ibu hamil dan menyusui masih ditemui di  masyarakat. Edukasi perlu dilakukan untuk meningkatkan pengetahuan kader posyandu tentang penggunaan obat pada ibu hamil dan menyusui. Edukasi dilakukan dengan metode modul, ceramah dan diskusi. Instrumen penelitian berupa kuesioner yang dibagikan kepada responden sebelum dan sesudah edukasi untuk mengevaluasi tingkat pengetahuan peserta. Kuesioner berisi 10 item pertanyaan tertutup terkait pengetahuan penggunaan obat pada ibu hamil dan menyusui. Data dianalisis dengan uji statistik paired t-test. Hasil menunjukkan bahwa rata-rata nilai pengetahuan kader meningkat 25,73% dari rata-rata nilai pengetahuan awal setelah dilakukan edukasi. Setelah dilakukan edukasi ada peningkatan tingkat pengetahuan yang signifikan. Hal ini menunjukkan bahwa edukasi dengan metode modul, ceramah dan diskusi mampu meningkatkan pengetahuan kader Posyandu.

Abstract

Fungi endofit berpotensi sebagai alternatif pengobatan karena menghasilkan senyawa yang sama dengan tumbuhan inangnya. Allium cepa memiliki manfaat sebagai antifungi dan antikolesterol. Tujuan penelitian ini adalah isolasi, identifikasi, serta melakukan uji antifungi dan antikolesterol. Isolat yang diperoleh diidentifikasi secara makroskopik dan mikroskopik. Metode uji antifungi terhadap Candida albicans adalah metode difusi agar dan metode uji antikolesterol menggunakan rancangan acak lengkap (pre and posttest control design). Hasil isolasi fungi endofit diperoleh isolat FEAC 1 dan FEAC 2. Hasil penelitian menunjukkan bahwa FEAC 2 memiliki zona hambat sebesar 7,389 mm dan menurunkan kolesterol total secara signifikan serta meningkatkan kolesterol HDL.

Abstract

The acute toxicity of Jatropa curcas leaves on Balb/C male mice was studied in rats. This research aimed to determine acute toxicity, evaluate spectrum of toxic effect and mechanism that caused the death of animal test after administration of ethanolic extract of J. curcas leaves, single dosage orally on 24 hours observation. The research used male mice, which are divided into 5 groups. Group I was negative control with CMC-Na. Group II, III, IV, and V were given extract with dose of 1400 mg/kgBW, 2240 mg/kgBW, 3584 mg/kgBW and 5734 mg/kgBW, respectively. Evaluation of the toxic symptoms and death of animal test was done for 24 hours. If the animal test was died before 24 hours then it underwent surgery to take the heart, liver, lung, and kidney. In the end of the evaluation, all mice were killed to take the vital organs for histopathologic examination. No mortality was observed during study. The test resulted LD50 of ethanolic extract from J. curcas leaves using Balb/C male mice was 5734 mg/kg of BW. It was categorized as practically not toxic. Administration of the extract did not cause alterations of animal behaviours. Histopathology examination shows inflammation in lung, liver, and kidney after administration of the extract.

Abstract

Free radical in the body could cause degenerative diseases such as diabetes mellitus and hyperlipidemia, which could be prevented by the supplementation of antioxidant whether it is synthetic or natural. Flavanoids is a phenolic compound, was extracted from natural sources is known for its antioxidative potential. This research aimed to investigate the active compounds in brotowali (Tinospora crispa) stem to find new antioxidative potential using its ability to bind 2, 2’-diphenyl-1-picrylhydrazyl (DPPH). Brotowali stem was extracted using ethanol and fractioned using n-hexane, ethyl acetate, and water. Water fraction was hydrolyzed by chloric acid. The antioxidant activity of ethanol extract, water fraction, and hydrolized water fraction (subfraction I and subfraction II) were determined using spectrophotometry (DPPH methods). The compound of flavonoid was identified using KLT with AlCl3 reagent spray. The results showed that ethanol extract and water fraction of brotowali stem contained flavonoid glycoside of flavonol group. Antioxidant activity, quantified using IC50, of ethanol extract, water fraction, subfraction I and subfractiont II were 49.92μg/mL, 38.25 μg/mL, 36.12 μg/mL, and 16.18 μg/mL, respectively. In addition, this research was found that hydrolysis of water fraction using chloric acid like in subfraction I and subfraction II was able to improve its antioxidant activity.

Abstract

Studies have been conducted to determine the potential antiproliferative effect of Nigella sativa seed(NSS) extract on 7,12- dimetilbenza[a]antrasena (DMBA)-induced female rats. The chloroform NSS extracteffect was observed on rat gatric cells. Sprague Dawley  female rats were divided into five groups, eachgroup consisted of 12 individuals. A control group was DMBA, Groups B, C and D were the groups treatedwith NSS extract with ranked dose 250 mg / kg, 500 mg / kg and 750 mg / kg and group E is the corn oilcontrol group. Histopathological observation of cells using H&E staining and cell proliferation activity wasobserved with the mean AgNOR (mAgNOR). mAgNOR values   were analyzed using Kolmogorov-Smirnovtest, than followed with ANOVA one way, and Tukey test HSD. The results showed H&E staining and  thechloroform exctract showed mAgNOR N.sativa semen were able to reduced gastric cell damage and lowergastric cell proliferation in DMBA- induced rat. The results showed that N.sativa potential to develop asa chemopreventive agent in gastric cancer.
