Rapid Purification and Partial Characterization of an Extracellular Enantioselective Lipase from Aspergillus niger Malle, Dominggus; Garingan, Gina; Peralta, Milagros; Revilleza, Maria Jamela

Abstract

Lipases  [EC.  3.1.1 .3]  are  capable  of hydrolyzing  ester  bonds  of triglycerides  and  have  received  much attention  from  protein  researchers  after  these  enzymes  have  shown  potential  applications  such  us,  in  the manufacture of commercially important chiral drugs due to their enantioselectivity properties. A novel  extracellular lipase  produced  by Aspergillus  niger was  purified  and  partially  characterized.  Using  the  culture medium  as crude extract, the Lipase was purified  to 25 .7  folds after Ultrafiltration and DEAE ion exchange chromatography. The enzyme was characterized to have optimum activity at neutral pH and at 30-35°C, while its kinetic parameters, Vmax and Km, were determined  to  be  L80 U/mL/min  and 0.2 mL , respectively. SDS-PAGE analysis  revealed  the presence of at  least two  bands  (±87  and ±72  kDa), while  native  PAGE  showed  a  single  band.  The  two  bands could represent subunits  in  a complex or isoforms of the enzyme. The use of  the enzyme  in  th  hydrolysis of a racemic  mixture  of 2-arylpropionic  butyl  ester  analyzed  after HPLC demonstrated  an  apparent predominant enantioselectivity for the S (+) enantiomer. Thus, this lipase is a promising enzyme for the chiral drug preparation of  single enantiomer of 2-arylpropionic acid (ibuprofen) .Keywords: Iipase, Aspergillus niger, purification, enantioselectivity, chiral  drug, ibuprofen

Abstract

Hot springs are a common source of thermophyles which produce thermostable enzymes. The objective of this study was to isolate and identify thermostable amylase-producing bacteria from a local geothermal spring. An amylase–producing bacterium strain was isolated from this hot spring which excreted amylase after being grown on starch agar screening plates at 37 °C. It was identified as Bacillus megaterium using the method of 16S ribosomal DNA. The organism is a rod-shape and is a spore-forming  bacterium.  Maximum amylase production was achieved after incubation in the production media for 72 hours. Preliminary analysis of the secreted amylase showed that the enzyme could bind to DEAE-Sepharose matrix and was discharged by eluting with 0.5 M NaCl. The partially purified enzyme was stable up to 75 °C, showing that this enzyme might have potential application in the starch-processing industry.

Abstract

This study was aims to optimize bioethanol production from Borassus palmyrah and to determine content of yield optimization bioethanol fermentation. Fermentation process conducted by Borassus palmyrah optimize found ammonium sulfate addition in fermentation bottled depth as nitrogen resources. Fermentation process done by 10 g of yeast, 15 g of yeast, and 20g of yeast. This bioethanol will be used as a renewable energy source that uses Borassus palmyrah as a raw material. Qualitative test was conducted by a graph method and HPLC method. The result of the research showed that ethanol content with the addition of ammonium sulfate based on the graphic method was 77% - 85%, and the results test using HPLC was obtained an ethanol content of 98.41% - 99.72%. Thus, it can be concluded that the optimal bioethanol production process is more efficient by adding ammonium sulfate and yeast as a biocatalyst. Keywords: bioethanol, Borassus. Abstract

This research aims to determine the relationship between feed consumption and growth and feed conversion of broilers in the PT Mitra Sinar Jaya partnership. The research material is the broiler strain CP-707 which is kept in open house cages with a population of 6,500 in the 4 cages and commercial feed S10, S11, and S12. This panel research was carried out for 35 days and reared broilers at PT Mitra Sinar Jaya partnership breeders in Seri Village, Nusaniwe District, Ambon City. The method used in this research is survey and observation (direct observation in the field) to collect data related to the performance of the CP-707 broiler strain. The variables observed in this research were feed consumption, body weight gain, and feed conversion ratio (FCR). The data obtained was analyzed using a simple linear regression method. The research results showed a relationship between feed consumption and body weight gain and feed conversion ratio (FCR) with R2 values ​​of 0.9802 and 0.0228 respectively, with an average feed consumption of 514.13 g/head, UN of 311 .04 g/head, and an FCR value of 1.65.

Abstract

Polysaccharide from Gracilaria sp (locally known as “Sayur Karang”) was extracted using hot alkaline solutions (NaOH and KOH). Fractionation using DEAE-Sepharose Fast Flow column showed that the polysaccharide discharged after being eluted with 0.5 – 1.0 M NaCl. Spectroscopic analysis using FTIR showed that the polysaccharide contained hydroxyl (3420 cm-1 (NaOH frcation) dan 3446 cm-1 (KOH frcation)), carbonyl (1642 cm-1 (NaOH frcation) and 1644 cm-1 (KOH frcation)) and sulphate (1271 cm-1 (NaOH frcation) and 1262 cm-1 (KOH frcation)) groups proving that the polysaccharide is a sulphated polysaccharide. A further study is needed to elucidate the structure of the sulphated polysaccharide as well as physiochemical properties of the polysaccharide.

Abstract

Bioethanol can be produced through glucose fermentation using saccharomyces cerevisiae. Was done to make coconut water contains a small amount of carbohydrate. A researcher had taken research was done to make bioethanol from coconut water waste. The result of the research shows that coconut water fermentation with yeast has maximum speed after 70 minutes of incubation. The fermentation solution was then distilled. Bioethanol purity was about 76-80% after HPLC analysis.

Abstract

The papain enzyme was isolated from the latex of papaya and it was purified by ammonium sulfat fractionation and dialysis methods. Determination of papain enzyme activity showed that the highest activity was found in the 80% fraction with a specific activity of 10.5797 x 105 U/g. Papain concentration after Bradford method was 53 mg/mL. Molecular mass of papain was detected to be 21.6 kDa after SDS-PAGE analysis. Characterization of papain showed that the optimum activity of papain was found at pH 8.0 with a specific activity of 14.07031 x 10 U/g, and at temperature 60 °C with a specific activity of 14.39755 x 10 U/g.

Abstract

The research about analysis of cadmium (Cd) and lead (Pb) metal content on sediment inner part of Ambon Bay has been done. Sampling point located at Dermaga Ferry Poka, PLTD Poka, Kate-kate, Waiheru, Lateri III, Passo dan PLTD Galala. The results of analyzed of sediment particle size inside the Ambon Bay showed 3 components were sand, gravel, and sludge, but the sediments was dominated by sludge sand. Analysis of Cd content showed that at 7 stations in Ambon Bay detected Cd metal with the highest concentration at 4th station of Waiheru village was 0,6640 mg/Kg and the lowest concentration at 3th station of Kate-kate village was 0,0980 mg/Kg. Meanwhile, the highest Pb content was 20,075 mg/kg at 4th station and the lowest was 5,755 mg/kg at 2nd station of PLN Poka. The content of elements composition of sediment are SiO2, K2O, P2O5, Fe2, BaO, NiO and Ga2O3 but Cd and Pb metal was not detected.

Abstract

This research was conducted to find bacteria from the genus Acidithiobacillus ferrooxidans which isolated from mine acid water in the copper mining area on Wetar Island, Maluku Province. This bacterium will be utilized in the biohidrometallurgical process (bioleaching) for copper extraction in mining area. The results of this study indicate that bacteria can be isolated from mine acid water samples by enrichment method that was use liquid media from Leathen which is optimized using Trypton Soya Broth (TSB) and bacterial growth on solid media. The presence of Thiobacillussp bacteria was confirmed by the results of gene analysis using a 16S rRNA sequence showing the presence of mixed bacterial colonies but not a single colony.
