Abstract

Aquaculture production is vulnerable to adverse impacts to disease and environmental conditions. One of the pathogenic bacteria that infection cultured is Edwardsiella tarda. The aim of this study was to find out the sensitivity of R.apiculata leaf extract in inhibiting the growth of E.tarda. The research method used is the experimental method, with the Kirby-Bauer disc method. The doses used were 100% (10000 ppm), 90% (9000 ppm), 80% (8000 ppm), 70% (7000 ppm), 60% (6000 ppm), 50% (5000 ppm), 40% (4000 ppm), 30% (3000 ppm), 20% (2000 ppm), 10% (1000 ppm) and control (Oxytetracycline), blank discs used were 6 mm in size. The results showed that the ethanolic extract of the leaves of R. apiculata at a dose of 1000-10000 ppm gave various inhibitory diameters ranging from 6.27-9.87 mm and presenting inhibition zone diameters in the medium category

Abstract

One of the plants that have the potential as antibacterial ingredients is Rhizophora apiculata leaf because it contains secondary metabolites, such as tannins, saponins, flavonoids, steroids, and terpenoids. This study aims to find out the inhibition of hot water extract of R. apiculata leaves in inhibiting the growth of Edwardsiella tarda. The research method used is an experimental method which includes sample preparation, extraction, antibacterial activity test using the Kirby-Bauer disc method and applying a Completely Randomized Design (CRD). The doses used were 100% (10000 ppm), 90% (9000 ppm), 80% (8000 ppm), 70% (7000 ppm), 60% (6000 ppm), 50% (5000 ppm), 40% (4000 ppm), 30% (3000 ppm), 20% (2000 ppm), 10% (1000 ppm) and control (Oxytetracycline). Abstract

The use of herbal supplements is an alternative to increase food security, because it can stimulate fish appetite and increase fish immunity against disease. The aims of this research to know the total leukocytes, leukocyte differentiation and survival rate of striped catfish fed with dosage fermented herbal supplements, and to obtain the best dosage. The implementation of research was from June until August 2019 at the Parasites and Fish  Diseases Laboratory, Faculty of Fisheries and Marine, Riau University. The method used is a completely randomized design (CRD), with four treatments and three replications, namely P0 (without the addition of herbal supplements), P1 (herbal supplement dose 100 mL / kg of feed), P2 (200 mL / kg of feed), and P3 (300 mL / kg of feed). Maintenance of fish is carried out for 60 days, with a stocking density of 50 fish / m. The results showed that the herbal supplementation at a dose of 200 mL / kg of feed (P2) was the optimal dosage in stimulating total leukocytes and differentiation of catfish leukocytes. P2 treatment resulted in total leukocytes of 11.26 x 104 cells/mm3, further differentiation of leukocytes such as lymphocytes, monocytes, neutrophils and platelets of 81.67%, 6.33%, 5.67% and 6.33%. Abstract

Peningkatan keamanan pangan menggunakan bahan alami, seperti kunyit, kencur, dan temulawak sebagai suplemen pada pakan disebabkan karena bahan tersebut memiliki sifat yang ramah lingkungan, biodegradable, dan mudah ditemukan di alam. Penelitian ini bertujuan untuk mendapatkan dosis suplemen herbal dan padat tebar yang efektif dalam meningkatkan status kesehatan ikan ditinjau dari profil hematologis. Rancangan yang digunakan adalah Rancangan Acak Lengkap pola Faktorial. Perlakuan yang diberikan adalah dosis suplemen herbal (D1= 100 mL/kg pakan, D2= 200 mL/kg pakan, dan D3 = 300 mL/kg pakan) dan padat tebar ikan P1 = 50 ekor/ m3 dan P2 = 75 ekor/ m3. Ikan patin (Pangasius hypophthalmus) yang digunakan berukuran 8-12 cm, pemeliharaan dilakukan selama 60 hari di keramba jaring apung berukuran 1,5x1x1m. Pengambilan sampel darah dilakukan sebanyak tiga kali, yakni sebelum perlakuan, kedua pada hari ke-30 dan hari ke-60 pemeliharaan. Hasil penelitian menunjukkan bahwa penambahan dosis suplemen herbal 200 mL/kg pakan dan padat tebar 75 ekor/m3 yang dipelihara selama 60 hari memberikan hasil tertinggi terhadap profil hematologis, yaitu total eritrosit sebesar 2,81 x 106 sel/mm3, nilai hematokrit 39,00%, kadar hemoglobin 10,73 g/dL, total leukosit 11,41 x 104 sel/mm3 dan kelulusanhidup 100%. Hasil penelitian dapat disimpulkan bahwa kombinasi padat tebar 75 ekor/m3 dan dosis suplemen herbal 200 mL/kg pakan adalah yang terbaik untuk peningkatan kesehatan ikan patin.

Abstract

This study aimed to observe the inhibition capability of Rhizophora sp. leaf extract towards the Streptococcus agalactiae and Edwardsiella tarda bacteria. Rhizophora sp. leaf was extracted using ethanol. Inhibition action of Rhizophora sp. leaf extract towards Streptococcus agalactiae and Edwardsiella tarda was tested on TSA solid media. The concentration of Rhizophora sp. leaf extract used were 1000, 2000, 3000, 4000, 5000, 6000, 7000, 8000, 9000, 10000 ppm, and novobiocin antibiotic was used as a control. Each treatment was conducted three times. MIC (minimum inhibition concentration) was performed to determine the minimum dose that could inhibit the bacterial growth. The results showed that the inhibition value of Rhizophora sp. leaf extract at 2000 to 10000 ppm towards Streptococcus agalactiae was 8.60-16.30 mm, and 6.97-12.27 mm towards Edwardsiella tarda, whereas the inhibition value of novobiocin for both bacteria was 18.00-20.45 mm. The results of MIC value at dose of 2.000 ppm of Rhizophora sp. leaf extract was towards Streptococcus agalactiae with bacterial density of 165x108 CFU/mL, and towards Edwardsiella tarda 75x108 CFU/mL, respectively. In conclusion, Rhizophora sp. leaf extract had more bacteriostatic activity against Gram-positive bacteria (Streptococcus agalactiae) rather than the Gram-negative bacteria (Edwardsiella tarda).

Abstract

Striped catfish Pangasianodon hypophthalmus (Sauvage, 1878) has a high economic value and relatively fast growth, so that optimum stocking density and supplement of natural ingredients, such as Curcuma domestica Val., Kaempferia galanga Linn., and Curcuma xanthorriza Robx. can increase growth and immunity. This study aimed to obtain the optimum stocking density and herbal supplement dosage in feed to increase the growth and survival rates of striped catfish. An experimental method by applying factorial design was used, i.e. the dose and stocking density factors. The dosage of herbal supplements used were 100 mL kg-1 feed (D1), 200 mL kg-1 (D2) and 300 mL kg-1 (D3). Stocking density used was 50 m-3 (P1) and 75 m-3 (P2). The fish used were 8.5±0.5 cm long and weighed 7.5±0.3 g, kept in floating cages of 1 m x 1.5 m x 1 m. Fish maintenance was carried out for 60 days with feeding frequency of three times a day, as much as 10% of body weight. The results showed that the treatment of 200 mL kg-1 dosage and 75 m-3 stocking densities was the best treatment. This treatment showed that specific growth rate, absolute weight, absolute length and feed efficiency were 4.86%, 119.07 g, 15.45 cm, 82.93%, respectively. A supplement dose of 200 mL kg-1 in feed and stocking densities of 75 m-3 could increase growth rates, survival, and feed utilization efficiency of striped catfish. Abstrak Ikan jambal siam Pangasianodon hypophthalmus (Sauvage, 1878) memiliki nilai ekonomis tinggi dan pertumbuhan yang relatif cepat, sehingga dibutuhkan padat penebaran yang optimum dan suplemen pakan berbahan alami, seperti kunyit (Curcuma domestica Val), kencur (Kaempferia galanga, Linn) dan temulawak (Curcuma xanthorriza Robx) yang mampu meningkatkan pertumbuhan dan imunitas. Penelitian ini bertujuan untuk mendapatkan padat tebar dan dosis suplemen herbal yang optimum dalam pakan terhadap peningkatkan laju pertumbuhan dan sintasan ikan jambal siam. Metode yang digunakan adalah metode eksperimen dengan menerapkan Rancangan Faktorial, yakni faktor dosis dan padat tebar. Dosis suplemen herbal yang digunakan, yaitu 100 mL kg-1 pakan (D1), 200 mL kg-1 pakan (D2) dan 300 mL kg-1 pakan (D3). Padat tebar yang digunakan adalah 50 ekor m-3 (P1) dan 75 ekor m-3 (P2). Ikan jambal siam yang digunakan berukuran panjang 8,5±0,5 cm dan bobot 7,5±0,3 g, dipelihara di dalam keramba jaring apung berukuran 1 m x 1,5 m x 1 m. Pemeliharaan ikan dilakukan selama 60 hari, frekuensi pemberian pakan sebanyak tiga kali sehari, sebanyak 10% dari bobot tubuh. Hasil penelitian menunjukkan bahwa padat tebar dan dosis suplemen herbal yang optimum ditunjukkan oleh perlakuan dosis 200 mL kg-1 dan padat tebar 75 ekor m-3 menghasilkan laju pertumbuhan spesifik, bobot mutlak, dan panjang mutlak sebesar 4,86%, 119,07 g, dan 15,45 cm, serta dapat meningkatkan efisiensi pakan 82,93%. Disimpulkan bahwa dosis suplemen 200 mL kg-1 pada pakan dan padat tebar 75 ekor m-3 dapat meningkatkan laju pertumbuhan, sintasan, dan efisiensi pemanfaatan pakan.

Abstract

Tanaman mangrove Rhizophora apiculatamemiliki potensi yang sangat besar digunakan dalam bidang fitofarmakasebagai bahanantibakteri. Penelitian ini bertujuan untuk melihat sensitivitas daun R. apiculata yang diekstraksi menggunakan 4 jenis bahan pelarut berbeda, yaitu etanol, air panas, etil asetat, dan n-heksana terhadap Aeromonas hydrophila. Metode yang digunakan pertama pembuatan ekstrakdaun R. apiculata dengan pelarut etanol 96%, yang selanjutnya dipartisi menggunakan pelarut air panas, etil asetat, dan n-heksana. Kemudian dilakukan uji sensitivitas (zona hambat)  ekstrak daunR. apiculata terhadap  A. hydrophila. Konsentrasi ekstrakdaun R. apiculata dari setiap jenis pelarut yang digunakan adalah 1000, 2000, 3000, 4000, 5000, 6000, 7000, 8000, 9000, 10000ppm, sebagai kontrol digunakan antibiotik oxytetracyclin, disc  blank yang digunakan berukuran 6mm. Setiap perlakuan dilakukan tiga kali ulangan. Hasil penelitian menunjukkan bahwa ekstrak daun R. apiculata yang diekstraksi menggunakan pelarut berbeda pada dosis 1000 sampai 10000 ppm mampu menghambat pertumbuhan bakteri A. hydrophila, yakni berkisar antara 6,15-9,08 mm dengan pelarut etanol, air panas berkisar antara 6,20-8,50 mm, etil asetat berkisar antara 7,55-12,03 mm, dan n-heksana berkisar antara 6,15-8,45. Simpulan yang diperoleh bahwa ekstrak etil asetat lebih sensitif dibandingkan pelarut lainnya terhadap pertumbuhan A. hydrophila. Kata kunci :Rhizophora apiculata, Aeromonas hydrophila, Bahan Pelarut, dan                   Sensitivitas

Abstract

Penggunaan tumbuhan sebagai alternatif suplemen pakan memiliki kelebihan, salah satunya bersifat bio-degradable dan ramah lingkungan, serta mudah ditemukan di alam, seperti kunyit, temulawak, dan kencur. Tumbuhan ini bersifat antibakteri dan antimikroba, serta meningkatkan laju pertumbuhan dan sistem imun ikan. Tujuan penelitian adalah untuk mengetahui efektivitas dari penambahan suplemen herbal pada pakan buatan dalam meningkatkan laju pertumbuhan dan kelulushidupan ikan patin. Penelitian ini dilaksanakan pada bulan Juni s/d Agustus 2019, di Waduk Fakultas Perikanan dan Kelautan Universitas Riau, Pekanbaru. Metode yang digunakan adalah metode eksperimen dengan menerapkan rancangan acak lengkap (RAL) dengan empat perlakuan, yaitu P0 (kontrol), P1 (dosis suplemen 100 mL/kg pakan), P2 (200 mL/kg), P3 (300 mL/kg). Ikan dipelihara dengan padat tebar 75 ekor/m3, panjang 7 cm dan bobot 6 g, serta dipelihara selama 60 hari. Hasil penelitian menunjukkan bahwa pemberian pakan buatan dengan penambahan suplemen herbal mampu meningkatkan pertumbuhan dan tingkat kelulushidupan ikan patin. Dosis pemberian suplemen herbal sebesar 200 mL/kg pakan menghasilkan laju pertumbuhan dan tingkat kelulushidupan ikan patin terbaik, menghasilkan pertumbuhan bobot mutlak sebesar 119,08 g, laju pertumbuhan spesifik 4,86%/hari, panjang mutlak 15,45 cm, konversi pakan 1,21, efisiensi pakan 82,93% dan tingkat kelulushidupan 100%. Kualitas air selama penelitian berada dalam kisaran yang mendukung untuk pertumbuhan ikan patin, yaitu suhu berkisar antara 26,9-30,2oC, oksigen terlarut berkisar antara 4,3-6,1 mg/L, dan pH berkisar antara 6,1-6.8.

Abstract

Chaetomorpha sp. is a green algae that has anti-bacterial compounds and at certain times found very much. The purpose of this study was to see the inhibition zone produced from the ethanol extract of Chaetomorpha sp. against A. hydrophilla and Vibrio sp. This research was done in March 2022. Extraction of Chaetomorpha sp., antibacterial effectiveness testing was done at the Biotechnology Laboratory, Faculty of Fisheries and Marine, Riau University. The method used is descriptive method. Sensitivity test was performed using the Kirby Bauer disc method. To reduce the error rate, it was repeated three times. The doses of macroalgae extract used were, doses of 100% (10,000 ppm), 90% (9,000 ppm), 80% (8,000 ppm), 70% (7000 ppm), 60% (6000 ppm), 50% (5000 ppm), 40 % (4000 ppm), 30% (3000 ppm), 20% (2000 ppm), 10% (1000 ppm) and control (Oxytetracycline). Based on the observations, it was found that the use of ethanol extract of Chaetomorpha sp. The result is able to inhibit the growth of bacteria A. hydrophilla and Vibrio sp. at the highest dose (10000 ppm) in an range of 13.5 -14.5 mm which was classified as strong category, the lowest dose was at 5000 ppm as moderate category.
