Abstract

The objective of this study was to isolate actinomycetes from soil of tea plantations, Jamus ngawi as a microbiology lab manual chapter isolation and characterization of microbes. The medium used is the medium of MHA and MHB. Soil samples taken to a depth of 10-15 cm from the surface with capture location is about 07o 4153,07 S and 111  37 24.64 E. The isolates were obtained observed morphologic characters include colony shape, colony color, surface characteristics of colonies, colony growth, colony elevation, and the edge of the colony. Isolatesof actinomycetes were successfully isolated are Streptomyces, Nocardia and Nocardiopsis. Actinomycetes isolates have been found that can be used as a collection of the microbiologylaboratory.Keywords : actinomycetes, soil of tea plantation, microbiology lab manual

Abstract

Aspergillusniger is one type of mold that has a high ability to produce cellulases. Cellulases in the manufacturing process required the right conditions in order to obtain optimum quality and quantity. This study aims to determine the effect of different concentrations of inoculum and incubation time on cellulases activity of Aspergillusniger.The method used was a quantitative experiment with pattern completely randomized design(CRD) two factorial. Treatment research includes different of inoculum (K) which are 10%(K1), 15% (K2), 20% (K3) and incubation times (T) are 5 days (T1), 7 days (T2), 9days (T3). The datawere taken from the treatment is glucose levels of cellulases activity using theNelson Somogyi method. Analysis data using ANOVA two lanes with 5% of a significance level and continued to test Honestly Significant Difference (HSD). The results showed that: Fhit>Ftabso that there are significant differences in inoculum concentrations and incubation time of cellulolytic activity from Aspergillusniger.The highest glucose levels obtained in K3T1 treatment (20% of concentration and 5days of incubation time) with 0.072%.Keywords: Aspergillusniger, glucose, cellulases activity

Abstract

Pengembangan kacang gude sebagai sumber fitoestrogen merupakan salah satu upaya pelestarian keragaman hayati. Tujuan penelitian ini untuk menganalisis metabolisme fitoestrogen kacang gude dalam tubuh dan retensinya dalam jaringan. Hewan coba menggunakan sembilan ekor tikus putih betina galur Sprague Dawley umur 6-7 bulan, dan dikelompokkan menjadi 3 kelompok.  Kelompok I kontrol (P1), kelompok II diberi larutan kacang gude perbandingan 24g : 24ml (P2) dan kelompok III diberi larutan kacang gude perbandingan 8g : 24ml (P3). Pemberian larutan kacang gude menggunakan sonde, dimasukkan dalam lambung. Darah, urin, dan feses dikoleksi pada jam ke-8, ke-16, dan ke-24 (fraksi I, fraksi II, dan fraksi III) setelah pemberian kacang gude. Analisis fitoestrogen daidzein dalam darah, urin, dan feses dilakukan menggunakan high performance liquid chromatography (HPLC). Hasil analisis menunjukkan kadar daidzein kacang gude di darah lebih tinggi daripada di urin dan feses. Kadar daidzein meningkat pada fraksi II dan menurun pada fraksi III. Daidzein mengalami absorbsi, distribusi, ekskresi, dan retensi di dalam jaringan. Retensi fitoestrogen daidzein dalam jaringan sebesar 95,472%. The purpose of this research to analyze the metabolism of phytoestrogens pigeon pea in the body and its retention in the tissues. Animals used nine female Sprague-Dawley rats 6-7 months. The rats were grouped into 3 treatment groups. The first group was the control (P1), the second group contained those given a solution of pigeon pea seeds under the ratio of 24g : 24ml (P2) and the third group was given a solution of pigeon pea seeds under 8g : 24ml ratio (P3). The provision of pigeon pea solution by gavage using a sonde into the stomach. Blood, urine, and feces were collected on the hour all 8 hours of the 16th, and the 24th hour (fractions I, II fractions and fractions III). Analysis of phytoestrogen daidzein in the blood, urine, and feces was performed using high performance liquid chromatography (HPLC). The analysis showed that the levels of daidzein pigeon pea in the blood is higher than the levels in the urine and feces. Daidzein levels increased in the second fraction and decreases in fraction III. Daidzein experiencing absorption, distribution, excretion and retention in the tissues. Retention phytoestrogen daidzein in tissue 95.472%

Abstract

POTENTIAL OF MIND MAPS THROUGH COMMUNITY LEARNING TO IMPROVE LEARNING ACTIVITY ON SKELETAL SYSTEM CONCEPT                                 AbstractThis research described the use of mind map with discussion during learning activity that potentially increased learning community to deliver skeletal system concept. The mind map was applied in four meeting with 27 second-grade senior high school students. Students were put in groups and each groups was assigned to design a mind map of skeletal system concept. The data of learning community was obtained from: 1) The mind map results from all groups, 2) The discussion along every group to arrange the mind map, and 3) The students’ scores of skeletal system concept test.  The instruments used were rubric of mind map assessment, observation sheet, and essay test. Qualitative descriptive was used to analyse data. The results showed that 80% of the mind map achieved the assessment rubric while the activity of students was 64.4%. The last, the highest grade of students’ test was gained by group 4, that was 8.84 out of 10. AbstrakPenelitian ini mendeskripsikan potensi penggunaan peta pikiran melalui learning community (LC) sebagai upaya meningkatkan aktifitas belajar siswa pada materi system gerak. Penerapan peta pikiran dilakukan sebanyak empat kali tatap muka untuk satu kompetensi dasar pada siswa kelas XI dengan jumlah siswa sebanyak 27 orang. Seluruh kegiatan dilakukan secara berkelompok dengan mendesain peta pikiran yang dikerjakan sesuai materi. Data aktifitas belajar dengan LC yang dikumpulkan didapatkan dari 1) rubrik peta pikiran kelompok siswa, 2) aktifitas diskusi, dan 3) skor tes untuk materi system gerak. Instrumen penelitian yang dipergunakan adalah lembar penilaian, lembar observasi dan tes essay. Analisis data dilakukan secara deskriptif kualitatif. Hasil penelitian menunjukkan bahwa peta pikiran yang dikerjakan kelompok siswa adalah 80%, keaktifan diskusi memiliki rata-rata 64,64 dan rata-rata skor tes per kelompok dicapai oleh kelompok 5 dengan skor tertinggi  adalah 8,84.

Abstract

Masyarakat sambirejo umumnya tidak produktif dalam hal mengolah dan memanfaatkan air limbah, mereka membuang air limbah tersebut langsung ke selokan yang ada di depan rumah, tanpa diolah terlebih dahulu. Akibatnya, sungai yang menjadi tempat bermuaranya selokan tercemar, warnanya menjadi coklat dan mengeluarkan bau busuk, selain bisa menyebabkan ikan-ikan mati, zat-zat polutan yang terkandung di dalam limbah juga bisa menjadi sumber penyakit, seperti kolera, disentri,dan berbagai penyakit lainya. Oleh karena itu, pemahaman masyarakat akan dampak yang ditimbulkan dari air ini perlu ditingkatkan serta pelatihan ilmu pengolahan limbah air ini perlu diterapakan dalam kehidupan sehari-hari dengan Biofilter ini agar limbah air yang ada di desa Sambirejo dapat berkurang dan untuk juga untuk mengurangi pencemaran lingkungan, serta dapat meningkatkan sumber daya manusia di Sambirejo Madiun. Hasil kegiatan sosialisasi pemberdayaan yang dicapai dan analisis kuisioner yang dibagikan sebelum sosialisasi dalah 50% masyarakat mengetahui dampak dari limbah air rumah tangga khususnya deterjen dan 20 % masyarakat mengetahui alat B-FILMA , dan analisis hasil kuisioner yang dibagikan setelah sosialisasi adalah 75% masyarakat menjadi faham mengenai dampak dari limbah air rumah tangga khusunya deterjen dan 50% masyarakat mengetahui alat B-FILMA.

Abstract

Energy is a difficult problem to deal with. The increase in energy demand is caused by increasing population growth. Therefore, the alternative energy needed is biogas. Biogas is a gas produced by the fermentation process of organic material by anaerobic bacteria. Organic material that can be used for the process of making biogas is bagasse. Biogas production process using bagasse will be pretreatment with cellulolytic molds, namely Aspergillus nidulans. The purpose of this study was to determine the effect of fermentation time and differences in substrate concentrations on the optimization of biogas production. The fermentation time used was 48 hours, 96 hours, 144 hours and 192 hours with mold concentration 0%, 20%, 40%, and 60%. Indicators to be measured were biogas pressure, biogas volume and C / N ratio. The results showed that the fermentation time and concentration of mold affected the biogas pressure and biogas volume. The most optimal biogas pressure was at 96 hours fermentation time with 60% mold concentration and 1,00249 atm while the most optimal biogas volume was 96 hours fermentation time with a concentration of 60% and 0.73 cm3.

Abstract

The use of synthetic hormone preparations, one of the hormones estrogen widely used by people in the care and  treatment of diseases as hormone replacement therapy. Pigeon pea (Cajanus cajan) as local Leguminoceae plant has not been used optimally as estrogenic plants . The purpose of this study was to identify preclinical to analyze the Cajanus cajan in the blood profile and potential of the reproductive organs as phytoestrogens. The research method using 24 Sprague -Dawley female rats 6-7 months , grouped 3 treatment. The control group , the treatment group pigeon pea solution of 1 : 1 and group treatment solution pigeon pea 1 : 3. Blood sampling on the 8th, 16th hour and the 24th hour (fractions I, fractions II and fractions III ) for the analysis of isoflavones daidzein HPLC method . Treatment pigeon pea solution made up to 36 days. Surgery and organ harvesting ovarian and mammary continued hematoxylin eosin staining . The results showed levels of isoflavones daidzein maximum in treatment 1 : 1, fractions I, II , and III respectively 92773.474 pg / l ; 96766.278 pg / l ; 72033.062 pg / l. Tissue structure experiencing development of ovarian follicles and  germinal proliferation, proliferation of mammary tissue experiencing the lobules and ducts interlobularis development. Conclusion of the study is the pigeon pea daidzein levels in the blood increased in the 24th hour and potential as a natural estrogen.Key Word         : daidzein, phytoestrogens, ovarian, mammary

Abstract

Pigeonpea (Cajanus cajan) is a tropical legume grown mainly in Indonesia. Though largely considered an orphan crop, pigeonpea has a huge untapped potential for improvement both in quantity and quality of yield. Pigeonpea seeds contain 65 % starch. Based on its starch level can be use as substrate of solid-state fermentation. Solid-state fermentation using Aspergillus niger because they have amylolytic and cellulolytic activity and not produce micotoxin. This study was carried out to know the change reducing sugar level of fermenting pigeonpea as to know a success of fermentation process. Completely randomized design was used, data result were analyzed with ANOVA, then were continued with Least Significant Difference. Different parameters such as inoculums concentration and incubation time. Inoculum concentration of 2 mL/g and incubation time of 72 hours give the best result of 12,50%.Keywords: solid state fermentation, glucose reduction, proteolitic, pigeon pea
