Abstract

ABSTRAKKelapa Genjah kopyor asal Pati, Jawa Tengah merupakankekayaaan hayati asli Indonesia dengan nilai ekonomi tinggi. Informasikeragaman genetik kelapa kopyor masih terbatas. Data keragamanmorfologi dan genetik diperlukan dalam program pemuliaan kelapakopyor. Penelitian ini mempelajari keragaman tiga varietas kelapa genjahkopyor asal Pati yang telah dilepas berdasarkan karakter morfologi,kuantitas endosperma, dan keragaman alel marka SSR. Penelitiandilakukan di Pati dan di Laboratorium Plant Molecular Biology,Departemen Agronomi dan Hortikultura, IPB. Evaluasi dilakukan terhadaptiga populasi kelapa Genjah kopyor (hijau, coklat, dan kuning) dengan 30tanaman sampel untuk setiap populasi. Rataan data morfologi digunakanuntuk menyusun dendogram. Kuantitas endosperma diamati pada satubuah kelapa kopyor per tanaman yang dievaluasi. Karakteristikendosperma dikelompokkan sesuai kategori yang telah ditetapkan. Untuksetiap populasi, analisis marka dengan lima pasang primer SSR dilakukanpada 10 tanaman sampel. Data yang didapat digunakan untuk menentukankeragaman genetik kelapa Genjah kopyor asal Pati. Hasil pengamatanmenunjukkan keragaman morfologis dan alel SSR antar tanaman dalamvarietasnya (keragaman intra-varietas) rendah. Sebaliknya, keragamanmorfologis dan alel SSR antar varietasnya tinggi. Kuantitas endospermakelapa Genjah kopyor asal Pati bervariasi antara skor 1–6. Keragamangenetik yang rendah dalam varietas dan tinggi antar ketiga varietas (coklat,hijau, dan kuning) memperkuat pelepasan ketiganya sebagai varietas lokal.Selain itu, keragaman genetik antar tanaman dalam varietas yang rendahmendukung penggunaan ketiga varietas lokal sebagai tetua dalam programperakitan varietas kelapa kopyor unggul baru. Tetua yang dipilih dapatdiseleksi intra-varietas berdasarkan persentase buah kopyor per tandandan skor kuantitas endosperma yang tinggi.Kata kunci: Keragaman morfologis, keragaman intra dan antar varietas,kuantitas endospermaABSTRACTKopyor dwarf coconuts are mutants from Pati, Central Java havinghigh economic values. However, morphological and genetic diversities ofthis coconut were still limited. Morphological and genetic diversity dataare needed for breeding program. The research objectives were to evaluateintra and inter-specific diversity based on morphology, endospermquantity, and SSR alleles. Field evaluations were conducted in Pati whilelaboratory activities were at Plant Molecular Biology Laboratory,Department of Agronomy and Horticulture, IPB. Three populations ofkopyor dwarf varieties (brown, green, and yellow) were evaluated. Thirtytrees were sampled for each population. The average of morphologicaldata were used to construct cluster analysis. Endosperm quantity wasscored (0 – 9) based on a single nut sample. Ten palms were analyzedusing five SSR loci for each population and used to determine geneticdiversity of populations. Results of observations indicated intra-varietymorphological and SSR allele variations among kopyor dwarf was low.However, inter-variety variations were high. The endosperm quantityscores among kopyor dwarf coconut ranged from 1–6. The low intra-variety and high inter-variety variations among the three kopyor dwarfcoconut supported their release as different local varieties. Moreover, thelow intra-variety phenotypic and genotypic diversities among kopyorbrown, green, and yellow dwarf coconut support their use as parents fornew and superior kopyor coconut variety development in the future. Forsuch purpose, however, it is necessary to conduct intra-variety selection toidentify desirable parents based on high kopyor fruit percentage per bunchand for high kopyor endosperm quantity.Key words: Morphological diversity, intra and inter variety diversities,quantity, endosperm

Abstract

Induksi poliploidi pada bibit Phalaenopsis amabilis telah dilakukan menggunakan kolkisin secara in vivo. Induksi poliploidi dilakukan dengan meneteskan kolkisin pada pucuk bibit P. amabilis.  Tujuan penelitian ini adalah untuk mendapatkan konsentrasi kolkisin yang efektif untuk induksi poliploidi bibit P. amabilis dan  menghasilkan bibit P. amabilis poliploid. Percobaan disusun dalam rancangan kelompok lengkap teracak dengan satu faktor, yaitu konsentrasi kolkisin. Pucuk bibit P. amabilis ditetesi 0,01 ml kolkisin (0, 1000, 2000, 3000, 4000, dan 5000 mg L-1). Hasil percobaan menunjukkan bahwa peningkatan konsentrasi kolkisin dari 1000 sampai 5000 mg L-1 tidak berpengaruh nyata terhadap persentase hidup dan pertumbuhan bibit pada 24 minggu setelah perlakuan (24 MSP). Bibit P. amabilis poliploid dapat dihasilkan pada penetesan kolkisin 1000, 3000, 4000, dan 5000 mg L-1 dengan konsentrasi kolkisin paling efektif adalah 5000 mg L-1. Bibit poliploid memiliki ukuran stomata lebih besar dari bibit diploid sebaliknya kerapatan stomatanya lebih rendah.

Abstract

Transcription factors have an important function in regulating gene expression and plant responses to stresses. The ERF, bZIP, WRKY, MYB, and NAC are stress inducible transcription factors. The OsNAC6 is a member of the NAC transcription factor family in rice and its expression is induced by abiotic stresses, wounding and blast disease. Characterization of OsNAC6 gene sequences would give a better undersanding on how OsNAC gene functions biologically. The objectives of this research are to isolate the OsNAC6 cDNA from Nipponbare, Batutegi, and Rojolele cultivars, to characterize their DNA sequences, and to compare their sequences to other NAC genes from other plants available in GenBank DNA databases. Isolated cDNA and sequencing of the fragments resulted in a 912 bp DNA sequences. Translation of the sequences yielded a protein consisted of 303 amino acid residue. Blast analysis of amino acid sequences indicated identity of isolated cDNA from three Indonesian rice cultivars are the OsNAC6 gene. Deduced amino acid residues from amplified cDNAs of Nipponbare, Batutegi, and Rojolele cultivars shared 100% sequence identities to rice OsNAC6 (Acc. # BAA89800), 71-100% sequence identity to a number of OsNAC protein from Oryza sativa and 63-83% sequence identity to NAC protein from other plants.

Abstract

In Vitro Multiplication of  Black Orchid (Coelogyne pandurata Lindl.) Using the Combination of NAA and BAPABSTRACTBlack orchid is an indigenous plant from Kalimantan, Indonesia. It becomes endangered because of forest over-exploitation and its low natural reproduction rate. Tissue culture is considered to offer a solution to conserve and propagate this species. The aim of this research was to evaluate the effect of Naphtalene Acetic Acid (NAA) and 6-Benzile Amino Purine (BAP) on shoots multiplication of black orchid. The basic medium used was a half of Murashige & Skoog (MS) composition supplemented with 150 mLL-1 coconut water. Initial explants used were 6-month-old shoots of germinating seeds. The shoot cultures were incubated for 23 weeks. Results showed that the best combination for shoot multiplication was NAA 0.0 mgL-1 with BAP 0.2 mgL-1. Shoot grew better on medium with BAP and without NAA while roots growth was better on medium without the two plant growth regulators. The addition of BAP up to 0.3 mgL-1 increased the leaf number, which however decreased at higher BAP concentration.Keywords: BAP, black orchid, Coelogyne pandurata, multiplication, NAA ABSTRAKAnggrek hitam merupakan flora langka asli Kalimantan, Indonesia. Keberadaa anggrek ini di alam semakin langka akibat eksploitasi berlebihan dan sulitnya perbanyakan secara alami. Kultur jaringan merupakan metode untuk mengatasi kelangkaan anggrek ini. Penelitian ini bertujuan untuk mengetahui pengaruh kombinasi NAA dan BAP terhadap multiplikasi anggrek hitam. Media dasar yang digunakan adalah ½ MS dengan penambahan air kelapa 150 mLL-1. Eksplan yang digunakan adalah tunas hasil semai biji umur 6 bulan. Kultur tunas diinkubasi selama 23 minggu. Hasil penelitian menunjukkan bahwa kombinasi terbaik untuk multiplikasi tunas adalah NAA 0 mgL-1 dengan BAP 0,2 mgL-1. Tunas tumbuh lebih baik dalam media dengan penambahan BAP tanpa NAA, sedangkan akar pada media tanpa NAA dan BAP. Abstract

A study was conducted to evaluate the effects of the growth retardant Cycocel application in the growth and development of poinsettia (Euphorbia pulcherrima) ‘Freedom Red’.  Cycocel was applied by spraying plant shoots at the concentrations of 1000, 1500, 2000, 2500 ppm. The increase in cycocel concentrations reduced plant height, leaf size, internodal length, plant spread and the number of flowering plants. All cycocel concentrations resulted in an ideal potted plant height. The optimum spray concentration of Cycocel used to obtain compact and uniformly flowering plants under tropical West Java environment was 1000 ppm.

Abstract

The aims of this study were to evaluate the physical and chemical change of pummelo fruit during storage with waxing and wrapping, and to determine the effect of waxing and wrapping and their  its interaction in affecting shelf life of pummelo fruits. This research was arranged in a randomized complete block design with two factors and three replications. The first factor was waxing consisted of without waxing, 10% beeswax, 5% beeswax, and chitosan. The second factor was wrapping consisted of without wrapping and wrapping using either transparent yellow or white plastic wrap. Pummelo fruits experience weight loss, reduced total soluble solid (TSS), decrease in firmness and total acidity during 48 days of storage in room temperature. Waxing using beeswax or wrapping using plastic wraps significantly suppressed weight loss and maintained fruit firmness compared to those without waxing and wrapping. However, waxing and wrapping did not significantly affect the quality of pummelo fruits. Based on the fruit’s weight loss, firmness, and panelist preferences, 5% or 10% bees wax can be used as waxing. Both transparent yellow and white plastic wraps can be used to maintain fruit weight, firmness, and appearances during storage.

Abstract

Phalaenopsis amabilis is an Indonesia native orchid species having large, white flowers with yellow labellum coloration. This studies aimed to develop Phal. amabilis micropropagation methods and evaluate the regenerated plantlet fidelity. Media supplemented with Thidiazuron (TDZ) and Polyvinylpyrrolidone (PVP) and medium pH adjustment effects to induce protocorm-like bodies (PLBs) from leaf explants and proliferate secondary PLBs were investigated. Clonal fidelity among regenerated plantlets was evaluated using eighteen SNAP marker loci. The results showed that the ½ MS medium supplemented with 3 mg L-1 TDZ and 0.5 g L-1 PVP was the best for PLB induction while the ½ MS medium supplemented with 0.5 mg L-1 TDZ was the best for PLB proliferation. For PLB induction, the media pH was adjusted into pH=7 for efficient PLB regeneration. Based on the assessment using 18 SNAP marker loci, four variant alleles in three loci (11.8%) out of a total 34 plantlets were detected. The mutation frequency at the evaluated SNAP marker loci was 2.5 x 103 (0.25%). Changes in SNP alleles may not always result in phenotype changes and allele variant occurrences may not affect phenotype fidelity of micro-propagated Phal. amabilis plantlets. Therefore, further studies about the phenotype fidelity among plantlets are necessary.

Abstract

Torbangun (Plectranthus amboinicus) merupakan tanaman obat dari keluarga Lamiaceae. Pada masyarakat Batak, daun torbangun digunakan untuk meningkatkan produksi air susu. Keragaman kandungan fitokimia dapat ditingkatkan salah satunya dengan teknik iradiasi sinar gamma untuk program pemuliaan tanaman. Pada penelitian ini, metode metabolomik yang tidak tertarget digunakan untuk mengevaluasi profil senyawa pada bagian daun tanaman mutan dari iradiasi sinar gamma dan kontrol. Kandungan senyawa planlet mutan dan kontrol dianalisis dengan GC-MS. Data GC-MS dianalisis dengan kemometrik dengan menggunakan hierarchical cluster analysis (HCA). Keragaman kandungan kimia ditunjukkan oleh planlet mutan dan kontrol daun torbangun. Hasil analisa planlet kontrol torbangun menunjukkan terdapat lima senyawa utama diantaranya hydroxymethylfurfurole (35,15%), brevifolin (7,69%), 3-Dihydro-3,5-Dihydroxy-6-Methyl-4H-Pyran-4-one (DDMP) (13,13%), stigmasterol (6,51%) dan ferruginol (8.63%). Hasil analisa mutan torbangun memiliki kandungan senyawa DDMP (19,57%), neophytadiene (8,47%), linolenic acid (9,10%), ferruginol (7,61%), stigmasterol (14,14%) dan gamma.-sitosterol (7,08%). HCA menunjukkan tiga komponen senyawa yang berbeda antara planlet kontrol dan mutan yaitu kenaikan kandungan senyawa DDMP dan stigmasterol, serta penurunan kandungan hydroxymethylfurfurole pada mutan torbangun.

Abstract

Pengembangan kultivar pisang tahan penyakit secara konvensional menghadapi kendala utama, yaitu lamanya waktu yang diperlukan untuk seleksi dan evaluasi tanaman hasil persilangan. Oleh karena itu dilakukan pendekatan bioteknologi melalui pengembangan marka molekuler untuk mempercepat capaian program pemuliaan. Tujuan penelitian ialah mengembangkan marka SNAP untuk seleksi ketahanan tanaman pisang terhadap penyakit berdasarkan substitusi basa nukleotida atau single nucleotide polymorphism (SNP) pada fragmen resistance gene analogue (RGA) asal tanaman pisang (MNBS). Kegiatan penelitian dilaksanakan di Laboratorium Biologi Molekular Tanaman, Fakultas Pertanian, Institut Pertanian Bogor, mulai Bulan Juli sampai November 2012. Penelitian menggunakan sembilan sekuen MNBS yang terdeposit pada bank data NCBI untuk mendisain primer SNAP yang diperlukan. Dari 30 posisi SNP yang ada pada fragmen MNBS, terdapat 19 posisi SNP yang menyebabkan perubahan asam amino yang disandikan. Delapan posisi SNP di antaranya dapat digunakan untuk pengembangan marka SNAP. Dengan menggunakan perangkat lunak WebSNAPER, primer SNAP berhasil didisain dari delapan posisi SNP. Namun satu posisi SNP menghasilkan seperangkat primer yang tidak layak pakai. Dari 64 alternatif primer SNAP yang dihasilkan berhasil dipilih 14 pasang primer SNAP. Sepuluh dari 14 pasang primer terpilih, dihasilkan dari posisi SNP1, SNP2, SNP4, SNP5, dan SNP6. Sepuluh pasang primer SNAP terpilih juga telah diuji dan berhasil mengamplifikasi marka SNAP menggunakan DNA genom pisang cv. Klutuk Wulung dan Barangan. Sepuluh pasang primer SNAP tersebut berpotensi untuk digunakan sebagai marka ketahanan terhadap penyakit padatanaman pisang secara umum.
