Abstract

Characteristics and Kinetics of Chitinase Enzyme from Entomopathogenic Fungus Beauveria bassianaBeauveria bassiana is one of the entomopathogenic fungi that produces chitinase when infecting the host. Chitinase is widely used as biocontrol agents because it can degrade chitin into an environmentally friendly product. This study aims to characterize and test the kinetics of chitinase from B. bassiana. This characterization includes determination of pH and optimum temperature, enzyme stability and enzyme kinetics test by determining Km and Vmax value with Lineweaver-Burk equations. The result of experiment showed that the chitinase B. bassiana had pH and optimum temperature of 5 and 40ºC respectively. This enzyme was stable until 90 minutes incubation at 40ºC. The Km and Vmax values were 0.181 mg/L and 0.022 mg/L.sec respectively. The Km value is higher than Vmax, which means the affinity of the enzyme to the lower substrate requiring high substrate concentration to increase the reaction rate. It can be concluded that the chitinase activity of B. bassiana is still low.Keywords: Beauveria bassiana, characteristics and kinetics, chitinase enzyme, entomopathogenic, Lineweaver-BurkABSTRAKBeauveria bassiana merupakan salah satu jamur entomopatogen yang memproduksi kitinase saat menginfeksi inangnya. Enzim kitinase saat ini banyak digunakan sebagai agen biokontrol karena dapat mendegradasi kitin menjadi produk yang ramah lingkungan. Penelitian ini bertujuan untuk mengkarakterisasi dan menguji kinetika enzim kitinase asal jamur B. bassiana. Metode yang digunakan dalam karakterisasi ini mencakup penentuan pH dan suhu optimum, kestabilan enzim pada suhu optimumnya, dan uji kinetika enzim yang mencakup penentuan nilai Km dan Vmaks dengan persamaan Lineweaver-Burk. Hasil penelitian karakterisasi menunjukkan bahwa enzim kitinase B. bassiana mempunyai pH dan suhu optimum masing-masing 5 dan 40ºC. Enzim ini stabil sampai pada 90 menit inkubasi pada suhu 40ºC. Nilai Km diperoleh 0,181 mg/L dan Vmaks sebesar 0,022 mg/L.detik. Nilai Km lebih tinggi daripada Vmaks, yang artinya afinitas enzim terhadap substrat rendah sehingga membutuhkan konsentrasi substrat yang tinggi untuk meningkatkan kecepatan reaksi, maka dapat disimpulkan bahwa aktivitas kitinase dari B. bassiana masih tergolong rendah.Kata kunci: Beauveria bassiana, entomopatogen, enzim kitinase, karakteristik dan kinetik, Lineweaver-Burk

Abstract

Coal waste contains few macro-and micronutrients, which makes it less likely to become a growth site for microorganisms. One way to screen coal waste resources is to identify the diversity of microbes in them to study the relationship between these microbial communities in contributing to improving environmental pollution using metagenomics to determine microbial diversity. The purpose of this research is to study the diversity of microorganisms in areas contaminated with heavy metals using a metagenomic method. This study was performed using next-generation sequencing techniques, including DNA extraction, 16S rRNA amplification, and gene sequencing analysis. The results of this research found that the 10 most commonly found species were Baekduia soli, Nocardiodes iriomotensis, Nocardioides mesophilus, Nocardioides pakistanensis, Propionibacterium cyclohexanicum, Solirubrobacter ginsenosidimutans, Gemmatirosa kalamazoonensis, Roseisolibacter agri, Kosakonia saccahri, and Dickeya fangzhongdai. Based on this research, it can be concluded that most of the microbial communities from coal waste are dominated by the phylum Actinobaceria. The results of this study can be used as an adaptive microbial germplasm for industrial waste management strategies.

Abstract

This article explores the development and application of smart aeroponic innovation as an effort to improve efficiency in energy-friendly urban farming. Smart aeroponics, which utilizes sensor technology and automatic control systems, is designed to maximize plant growth while minimizing energy and water use. In this community service project, we implemented a smart aeroponic system in several urban locations and trained local communities in its operation and maintenance. Evaluation of the results showed that this system can increase energy efficiency by up to 70% when compared to hydroponics with the Deep Flow Technique (DFT) method or each can save 29.02 kWh for mustard greens cultivation. And 41.00 kWh for lettuce cultivation. The potential of this technology is an effort to improve the efficiency of urban agriculture, but also provides significant ecological and economic benefits for urban communities, so that it can be a sustainable model for future agricultural development.

Abstract

Sugihmanik Village in Grobogan Regency, Central Java, has many people who work as tofu producers. However, this village is experiencing an environmental management crisis, one of which is the management of sludge resulting from the liquid waste processing process at their wastewater treatment plant (IPAL). One of the proposed solutions is the use of tofu waste sludge as maggot feed, which can provide economic benefits and reduce environmental pollution. The Krida Manik Waste Bank is expected to be a pioneer in managing and utilizing tofu mud to feed maggots which can later be used as animal feed. This study uses a qualitative descriptive method where data is extracted as much as possible to obtain information regarding program management. The results of the activity show that there is potential for managing IPAL sludge using maggots. With the abundant potential of mud, people can use it as a renewable resource. However, capacity development and ongoing assistance need to be carried out to get maximum results.

Abstract

Sugihmanik Village, a major tofu production center, produces substantial amounts of organic waste, particularly wastewater that contaminates local rivers and disrupts the surrounding ecosystem. A community service program has been developed to convert tofu waste sludge into maggot feed, followed by drying using a hybrid solar and gas-powered maggot drying machine. Initial activities involved program outreach and technical training on maggot cultivation and the drying process. A drying trial demonstrated that 200 grams of wet maggots were reduced to 114 grams within seven minutes. Monthly drying operations required only four 3-kg LPG cylinders, resulting in 81 kilograms of dry maggots and a potential monthly income of IDR 4.050.000. Environmental degradation caused by tofu waste has been significantly mitigated, while local economic productivity has improved. The drying machine enhances the commercial value of maggots as durable livestock feed. The initiative strengthens the operational sustainability of the Krida Manik Waste Bank and offers a scalable model for resource-based environmental solutions.

Abstract

Ramadan fasting is a fast that must be done by Muslims during the month of Ramadan for a full month. Diet during Ramadan tends to change, due to differences in meal times compared to other months. The pandemic period coincides with the month of Ramadan which sometimes neglects health and diet during breaking and sahur. It is important to maintain immunity when fasting during a pandemic in order to improve health. This community service activity program is to increase knowledge and understanding of maintaining diet when fasting. The result of this activity was an increase in knowledge about health and continued with medical examinations, namely checking blood pressure, checking blood sugar levels, checking uric acid levels and checking cholesterol levels.

Abstract

Remaja merupakan tahap transisi dari masa kanak-kanak menuju dewasa, yang ditandai dengan pertumbuhan fisik yang cepat, sehingga membutuhkan asupan gizi yang memadai termasuk zat besi. Asupan zat besi yang kurang dapat menimbulkan anemia pada remaja. Tujuan pengabdian masyarakat ini untuk memberikan edukasi dan pemeriksaan hemoglobin serta pengukuran antropometri kepada remaja di SMK Nurul Islami Semarang mengenai anemia dan upaya pencegahan anemia pada remaja. Pengabdian ini dilakukan dengan metode sosialisasi dan Focus Group Discussion (FGD) serta pengecekan kadar Hemoglobin dan antropometri pada siswa-siswi SMK Nurul Islami Semarang. Kegiatan ini diikuti oleh 35 siswa. Hasil pengabdian menujukkan bahwa siswa SMK Nurul Islami Semarang tidak terindikasi anemia, dengan rata-rata siswa memiliki kadar hemoglobin normal sebanyak memiliki rata-rata kadar hemoglobin normal sebanyak 30 orang (85,71%),yang didukung dengan rata-rata IMT siswa berstatus gizi baik (65,71%), dengan ukuran LILA normal sebanyak 27 orang (77,14%) meskipun beberapa memiliki tekanan darah rendah yaitu sebanyak 24 orang (68,57%). Dengan adanya pengecekan kadar hemoglobin dan antropometri pada kegiatan pengabdian ini siswa siswi menjadi lebih memahami pentingnya pencegahan anemia dengan mengonsumsi tablet penambah darah dan makanan yang bergizi dengan didukung aktivitas fisik.

Abstract

Ketersediaan lahan yang terbatas merupakan salah satu masalah dalam memenuhi kebutuhan pangan. Urban farming dapat digunakan sebagai salah satu upaya untuk mengatasi masalah ini karena urban farming merupakan solusi dari terbatasnya lahan untuk bercocok tanam. Saat ini sistem urban farming masih menggunakan metode manual yaitu terkait media yang digunakan untuk memperkenalkan kegiatan urban farming serta memperkenalkan produk hasil panen kepada konsumen. Guna mendukung berkembangnya kegiatan urban farming maka di butuhkan peranan Internet of Things (IoT). Tujuan kegiatan pengabdian masyarakat adalah agar masyarakat dapat memanfaatan lahan pekarangan yang sempit sebagai penghasil sayur-sayuran yang sehat untuk keluarga dengan budidaya tanaman teknik vertikultur berbasis IoT khususnya kelurahan tambakharjo. Pelaksanaan pengabdian ini menggunakan metode ceramah dengan dialog interaktif kepda peserta untuk menyampaikan pendapat, tanggapan maupun pertanyaan serta seluas-luasnya. Serta praktek secara langsung untuk menyiapkan benih, serta pengontrolan menggunakan aplikasi Tuya Smart. Hasil dari kegiatan ini yaitu mampu meningkatkan pengetahuan dan keterampilan warga atau peserta tentang budidaya tanaman di lahan pekarangan. Berdasarkan hal tersebut maka dapat disimpulkan bahwa program pemberdayaan pengembangan Smart Aeroponik ini mampu mengoptimalkan potensi lahan kosong yang berada di pekarangan warga agar menjadi lahan yang produktif dan bernilai guna. Hasil panen dari urban farming dapat dimanfaatkan untuk konsumsi sendiri maupun dijual.
