Claims (15)
- 시료용액을 R1이 R2및 R3뿐만아니라 측정하려는 물질과 특이적으로 결합할 수 있고, R2가 고체상으로의 결합을 중개하고, 또 R3이 표식을 갖는 최소한 3가지의 수용체 R1, R2및 R3와 인큐베이션시키고, 결합된 표식과 결합되지 않은 표식을 분리하고, 두 상중 한쪽에 있는 표식을 측정하므로써 특이적으로 결합 가능한 물질을 측정하는 방법에 있어, 수용체 R1, 으로서 측정하려는 물질의 에피토프와 특이적인 결합을 하는 결합부위를 최소한 두개 갖는 수용체를 사용하고, R2로서 특이적으로 결합하는 쌍의 성분 P1과 측정하려는 물질 또는 이것의 유도체에 상응하며 측정하려고 하는 물질의 에피토프를 최소한 하나가지는 물질 S와의 접합체를 사용하고 그러므로서 성분 P1은 고체상에 결합되거나 또는 고정화되고, R3으로서 최소한 하나의 물질 S와 표식을 함유하는 복합체를 사용하는 것을 특징으로 하는, 특이적으로 결합할 수 있는 물질의 측정방법.
- 제1항에 있어서, 수용체 R1으로서, 측정하려는 물질이나 이것의 RFab2단편과 특이적으로 결합할 수 있는 항체가 사용되는 방법.
- 제1항에 있어서, 수용체 R2로서, 물질 S가 성분 P1에 의해 결합되어 있는 고체상이 사용되는 방법.
- 제1항에 있어서, 수용체 R2로서 물질 S와 특이적 결합쌍의 성분 P1과의 접합체가 사용되고 고체상으로서 P1에 상보적인 여러특이적 결합쌍의 성분 P2가 결합되어 있는 매트릭스가 사용되는 방법.
- 제1항에 있어서, 수용체 R2로서 물질 S와 성분 P1과의 접합체가 사용되고 고체상으로서 다수의 성분 P1이 결합되어 있는 매트릭스가 사용되며 P1에 대해 특이적 결합부위를 최소한 두개 갖는 성분 P2를 부가시키므로서 인큐베이션 시킨후에 고정화가 일어나는 방법.
- 제5항에 있어서, 성분 P1으로서 비오틴이 사용되고 성분 P2로서 스트렙타비딘, 아비딘 또는 비오틴에 대한 항체가 사용되는 방법.
- 제1항에 있어서, 수용체 R3로서 물질 S와 표식과의 접합체가 사용되는 방법.
- 제7항에 있어서, 표식으로서 방사성 물질, 화학발광성물질 또는 형광성 물질이나 효소가 사용되는 방법.
- 제1항에 있어서, 수용체 R3로서 물질 S와 특이적 결합쌍의 성분 P1과의 접합체 및 P1에 상보적인 특이적 결합쌍의 성분 P2와 표식과의 접합체가 사용되는 방법.
- 제1항에 있어서, 결합쌍 P1-P2로서 비오틴-스트렙타비딘 또는 아비딘, 비오틴-비오틴 항체, 항원-항체, 합텐-결합 단백질 또는 올리고펩티드-항체가 사용되는 방법.
- 제4항에 있어서, 고체상으로서, 합성 수지로 구성되며 다수의 결합 성분 P2가 직접 결합되어 있거나 스페이서를 거쳐 결합되어 있는 매트릭스가 사용되는 방법.
- 제1항에 있어서, 성분 P1으로서 비오틴이 사용되고 성분 P2로서 스트렙타비딘 도는 아비딘이 사용되는 방법.
- 측정하려는 물질에 대해 최소한 2개의 특이적 결합부위를 갖는 수용체 R1, 특이적으로 결합하는 쌍의 성분 P1과 측정하려는 물질에 상응하거나, 또는 이의 유도체이고 측정하려는 물질의 에피토프를 최소한 하나 가지는 물질 S와의 접합체인 수용체 R2, 및 표식과 물질 S를 함유하는 복합체인 수용체 R3, 및 이것으로 부터 물리적으로 분리된, P1이 고정화되어 있는 고체상을 함유하는, 제1항 내지 제12항의 어느 한항의 방법에 의해 특이적으로 결합가능한 물질을 측정하기 위한 시약.
- 측정하려는 물질에 대해 최소한 2개의 특이적 결합부위를 갖는 수용체 R1, 특이적으로 결합하는 쌍의 성분 P1과 측정하려는 물질에 상응하거나 또는 이의 유도체이며 측정하려는 물질의 에피토프를 최소한 하나가지는 물질 S와의 접합체인 수용체 R2, 및 최소한 하나의 표식과 성분 P2를 함유하는 복합체인 수용체 R3' 및 이것으로 부터 물리적으로 분리된, P2가 고정화 되어있는 고체상을 함유하는, 제1항 내지 제12항의 어느 한 항의 방법에 의해 특이적으로 결합가능한 물질을 측정하기 위한 시약.
- 특이적 결합쌍의 여러개의 성분 P1이 결합되어있는 고체상뿐 아니라 이것과 물리적으로 분리된 성분 P2및 수용체 R1, R2및 R3또는 R3'를 함유하는, 특이적으로 결합 가능한 물질을 측정하기 위한 시약.
