Abstract

Colorectal cancer is one of the leading causes of death in the world caused by various factors such as genetic mutations, epigenetics, dysregulation of communication signaling pathways and gut microbiota. In the presence of genetic, epigenetic mutations, there is a loss of expression and function of PTEN (Phosphatase and TENsin homologues) resulting in continued activation of the PI3K/Akt intracellular signaling cascade involved in colorectal carcinogenesis. The solution to this problem is to use miRNA-21. which plays an important role by suppressing the biological marker PTEN gene that is useful for diagnosis and treatment in colorectal cancer. The aim of this study was to determine the specifi c binding of miRNA-21 with the PTEN gene that plays a role in colorectal cancer based on the AGhybrid and Logistic Probability values. Complete sequences of PTEN genes were obtained from FASTA NCBI and CDS data were obtained from Refseq. Prediction of the binding of mature miRNA-21 with the PTEN gene was carried out using the StarMir Sfold 2.2 website. The predicted results of miRNA-21 binding with 1 target obtained 296 binding models. Specific binding was seen from AGhybrid -23.600 and logistic probability 0.818, there was a binding between PTEN gene binding and miR-21-5p at seedless CDS position 3627-3653 as evidenced by the lowest AGhybrid value of all bindings that occurred.

Abstract

Hipertensi saat ini masih menjadi masalah kesehatan dunia. Data yang diperoleh peneliti di Puskesmas Lepo-Lepo tahun 2022 diperoleh hasil jumlah kasus yang tercatat menderita hipertensi pada tahun 2020 yang menderita hipertensi lansia di atas 60 tahun sebanyak 256 kasus, pada tahun 2021 tercatat sebanyak 112 kasus dan pada tahun 2022 sebanyak 216 kasus. Penelitian ini bertujuan untuk mengetahui pengaruh promosi kesehatan menggunakan media vidio terhadap perilaku lansia tentang kejadian hipertensi di Wilayah kerja Puskesmas Lepo-Lepo Kendari. Jenis penelitian yang dilakukan adalah kuantitatif dengan desain berupa one group pretest posttest design. Populasi dalam penelitian ini sebanyak 216 lansia dan sampel 69 lansia. Metode analisis dengan menggunakan uji Mc Nemar. Hasil penelitian menunjukkan bahwa ada pengaruh promosi kesehatan menggunakan media video terhadap pengetahuan, sikap dan tindakan lansia tentang kejadian hipertensi dengan nilai p=0,000 (p<0,05). Bagi Puskesmas sebaiknya menyarankan lansia untuk aktif memeriksakan kesehatannya dan aktif mengecek tekanan darah untuk mencegah kejadian hipertensi.

Abstract

In Indonesia, various studies have shown that there is still a significant gap in public understanding of applicable laws and regulations, especially in rural or remote areas such as Bila Village. Therefore, efforts to bridge this gap require socialization of legal compliance and awareness in Bila Village, Sabbangparu District, Wajo Regency as a form of community service aimed at increasing public compliance and awareness. This socialization method is considered very effective in internalizing the law in the community to ensure that the law is not only known, understood and internalized but also respected and obeyed.
