Abstract

Pseudomonas aeruginosa is an opportunistic bacterium that causes infections in human. The wall cell of its bacteria contains lipopolysaccharide as virulency factors to protect it from human immunity. Lipopolysaccharide can inhibit phagocytosis in the body. Polysaccharide krestin (PSK) from Coriolus versicolor extract contains ?-glucan that can increase phagocytic activity and capacity. This research aimed to identify the effect on polysaccharide krestin from C. versicolor extract on phagocytic activity and capacity of mice exposed by P. aeruginosa. The design of this research was experimental design. There were six treatment groups. The phagocytic activity and capacity were counted on slide smears of mice peritoneal fluid. The data was analyzed by using one way ANOVA. The results of the phagocytic activity and capacity showed that PSK was added before exposure (P1) or after exposure (P2) or before-after exposure (P3) had potential to increase phagocytic activity and capacity. The conclusion of the research was that adding polysaccharide krestin either before exposure or after exposure or both of them could increase phagocytic activity and capacity. Abstract

Mycobacterium tuberculosis is a major infection agent of tuberculosis that is controlled by the response of cell-mediated immunity. It is macrophages and cytolytic T lymphocytes. Activated macrophages will produce free radicals. Excessive free radicals cause tissue damage. Polysaccharide krestin contains ?-glucan. It is a scavenger of free radicals. This research aimed to identify the influence of polysaccharide krestin from C. versicolor on nitrite and malondialdehyde concentrations of mice serum exposed by M. tuberculosis. Nitrite concentration was determined by nitrite assay. Malondialdehyde concentration was determined by TBARS assay. The result showed that adding polysaccharide krestin before exposure (P1) and adding polysaccharide krestin before-after exposure (P3) had the best potential to decrease nitrite concentration. Nitrite concentrations of P1 and P3 were 1.364 0.523 M and 1.456 0.712 M respectively. Meanwhile, P1 group and adding polysaccharide krestin after exposure (P2) had the best potential to decrease malondialdehyde concentration. Malondialdehyde concentrations of P1 and P2 were 1125.86 97.96 M and 953.86 328.16 M respectively. Their nitrite and malondialdehyde concentrations decreased, compared to K and K- groups. Abstract

Okra (Abelmoschus esculentus) is reported to have various biological functions such as antioxidant, anticancer and anti-inflammation. However, only few studies have been reported immunomodulatory activities of okra to prevent disease caused by bacteria infection. In this study, the immunomodulatory activities of polysaccharides from okra pods were investigated further through the spleen weight and B-lymphocytes proliferation in mice infected by Staphylococcus aureus. Okra polysaccharides were obtained by water extraction and ethanol precipitation. Okra polysaccharides with doses of 25, 50, 75, and 100 mg/kg BW were orally administrated to mice with or without Staphylococcus aureus infection. Spleen weight was evaluated in both treatment group and control group. B-lymphocytes proliferation was evaluated by MTT assay using LPS induction. Results showed that okra polysaccharide at the doses of 50, 75, and 100 mg/kg increased spleen weight (p < 0.05) significantly. While at the dose of 75 and 100 mg/kg, it increased the B-lymphocytes proliferation (p < 0.05) significantly. There is positive correlation between the spleen weight and B-lymphocytes proliferation by 73.3%. These result reveal that okra polysaccharide could improve the immune response and be utilized as a novel candidate of neutraceutical.

Abstract

The purpose of this research was to determine the effect of polysaccharide krestin (PSK) on the testicular protein profiles and testosterone levels of Mus musculus with variety of dosages. This research used a completely randomized design. It were devide into four treatment group i.e. control group, PSK treatment at a dose of  15, 30, 60 mg/kgBW. Each group had six replications. Testicular proteins were isolated by flushing technique and analized by SDS-PAGE. Testosterone levels were analized using ELISA technique at wavelength 450 nm. Protein bands analysis showed that there were no diversification between four treatments. Molecular weight of protein bands were 87, 63, 57, 35, and 30 kDa. The results of research showed that the testosterone levels at dosage 60 mg/kgBW had significanly different with control, PSK treatment of 15 and 30 mg/kgBW. PSK treatment of  60 mg/kgBW had lowest level at dosage, i.e. 25946.8 ρg/mL. It can be concluded that giving variety of dosages of polysaccharide krestin did not affect to testicular protein profiles but giving effect to testosterone levels of Mus musculus.

Abstract

Sodium nitrite (NaNO2) found in vegetables, drinking water, and cured meats, can damage tissue because it is an oxidant. Plant phytochemicals such as quercetin are antioxidants. This study aimed to determine the potential of red okra pods ethanol extract (ROE) to repair kidney damage in mice (Mus musculus) induced by NaNO2. The red okra pods were extracted three times with saturated ethanol. The experiment used 36 male BALB/c mice aged 6-8 weeks and body weight of about 28 g. There are six research groups, namely, normal control, negative control (exposure to NaNO2 50 mg/kg BW), treatment of exposure to NaNO2 and administration of ROE at doses of 25, 50, 75, and 100 mg/kg BW. Sodium nitrite and ROE were given daily for 23 days by gavage. On day 24, the serum was isolated. Blood urea nitrogen (BUN) and creatinine (Cre) levels are measured to assess kidney function, as well as measuring the oxidant malondialdehyde (MDA) and the antioxidant enzyme of superoxide dismutase (SOD). The kidneys were made histological preparations and analyzed on the proximal convoluted tubule (PCT).  All data were statistically analyzed (α=0.05). This study indicated that the administration of ROE at a 100 mg/kg BW dose is the most optimal in repairing damage to the PCT with increased normal cells and reduced necrosis. Besides, it degraded BUN, Cre, and MDA levels in the serum of mice exposed to NaNO2 compared to the other treatments. All doses of ROE promoted the SOD level. ROE restore kidney tissue, especially on PCT to normal. Kidney damage due to exposure to NaNO2 preservatives can be reduced by administering ROE. ROE prevents kidney damage through an increase in antioxidant enzymes. ROE can be used as a food ingredient as a source of antioxidants, thereby reducing the impact of oxidant compounds.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh pemberian polisakarida krestin ekstrak Coriolus veriscolor pada toksisitas kronis terhadap histologi ginjal dan kadar kreatinin serum Mus musculus. Dua puluh lima mencit betina, umur 8-10 minggu, berat 25-30 g dibagi menjadi 5 kelompok perlakuan dengan 5 ulangan. Kelompok perlakuan adalah kontrol (K) yang diberi akuades 0,2 mL selama 4 bulan,  perlakuan (P1, P2, P3 dan P4) diberi polisakarida krestin dari ekstrak jamur C. veriscolor dengan dosis yang berbeda yaitu: 0,5; 1; 2; dan 4 mg/kg BB selama 4 bulan.  Akhir dari perlakuan, ginjal diambil untuk dilihat histologinya dan isolasi serum darah untuk diukur kadar kreatinin. Data sel ginjal normal, sel yang bengkak, dan kadar kreatinin dianalisis dengan Anova  dan Duncan. Data nekrosis dianalisis dengan Brown-Forsythe dan Games Howell. Polisakarida krestin diberikan secara oral. Histologi ginjal (degenerasi dan nekrosis tubuli ginjal) diidentifikasi pada slide ukuran 4 µm.  Kadar kreatinin diukur dengan metode Jaffe reaction. Hasil penelitian menunjukkan bahwa perlakuan P1 sudah tampak adanya pembengkakan 32,80%  dan nekrosis sel 8,88%. Perlakuan P2 mempunyai jumlah pembengkakan sel 38,28% dan nekrosis sel 12,08%. Perlakuan P3 dengan jumlah pembengkakan sel 62,24% dan nekrosis sel 13,08%. Perlakuan P4 mempunyai jumlah pembengkakan sel 53,28% dan nekrosis sel 21,40%. Jumlah sel ginjal yang normal semakin turun seiring dengan kenaikan dosis PSK. Kadar kreatinin pada K, P1, P2, P3 dan P4 menunjukkan kadar kreatinin yang normal pada masing-masing perlakuan, yaitu 0,40; 0,64; 0,72; 0,80 dan 0,80 mg/dL. Pemberian polisakarida krestin (PSK) dari ekstrak C. veriscolor pada toksisitas kronis menyebabkan pembengkakan, nekrosis sel, tetapi tidak meningkatkan kadar kreatinin serum mencit secara signifikan. Kata Kunci:    polisakarida krestin, Coriolus veriscolor, toksisitas kronis, histologi ginjal, kadar kreatinin

Abstract

Coriolus versicolor is a mushroom that has polysaccharopeptide krestin (PSK) and polysaccharopeptide (PSP). Many reports showed that polysaccharide krestin as a compound which could reduce mutagen induction, radiation, and development of cancer spontaneusly. However, all substances entering in body could change become toxic depending on dosage. Therefore, this research was aimed to know the effects of PSK on hepatocyte damages, SGPT and SGOT enzymes. Polysaccharide krestin was given by intraperitoneal injection once with treatment groups as follow: K0, was given only saline; P1, was given 80 mg/kg BB PSK dosage; P2, was given 120 mg/ kg BB PSK dosage; P3, was given 160 mg/ kg BB PSK dosage; P4, was given 200 mg/ kg BB PSK dosage; dan P5, was given 240 mg/ kg BB PSK dosage. The results showed that PSK caused hepatic lesions, such as parenchymatous (hydrophic) degeneration of hepatocytes in groups P2 and P3, mild to moderate necrosis in group P4, and moderate to severe necrosis in group P5. Level of SGPT enzyme was not increased, butlevel of SGOT enzyme was increased in mice given 160 mg/ kg BB dosage with value 151,62 ± 26,62 IU/ L.

Abstract

The objectives of this study are to examine the impact of consumers' perceptions regarding the ethics of online retailers (CPEOR) on loyalty and to understand the moderating influence of individual characteristics in consumers' perceptions regarding the ethics of online retailers on loyalty. The CPEOR scale includes security, privacy, non-deception and fulfillment/reliability while the individual characteristics comprise vertical individualism characteristcs and horizontal individualism characteristics. The sample was collected by accidental sampling technic with the total of 200 respondents whom experiencing shopping online within the last three months. The research methods that this study uses are the simple linear regression which examines the impact of consumers' perceptions regarding the ethics of online retailers on loyalty and moderated regression analysis (MRA) which examines the moderating influence of individual characteristics in consumers' perceptions regarding the ethics of online retailers on loyalty. Also, this study has got two hypotheses and the result indicates that consumers' perceptions regarding the ethics of online retailers significantly affects loyalty. Study also found that individual characteristics does moderating consumers' perceptions regarding the ethics of online retailers on loyalty.

Abstract

Brand community has been extensively built and developed by either the companies or the customers voluntarily. These two gorups with different perspective hold different purposes. For the companies, brand community is considered an effective marketing strategy which is expected to achieve customer brand engagement. Anyhow, often companies focus more on the purposes of the brand community, but do not be quite aware of the policies, rules, or traditions firstly created as norms in order to lead the way the community members perceive and act. Often norms become pressures for the brand community members causing them reluctant to engage with he community. Referring to the theory of relationship marketing, this study develops a model of normative community pressure that is affected by both brand community identification and inter-member relationship quality, that are previously affected by brand relationship quality. The empirical analysis derives primary data of young entrepreneurship community created by one of the big five banks in Indonesia quantitatively analyzed using Structural Equation Modeling (Warp-PLS). The result helps explain the antecedents causing normative community pressure of the brand community. The implication of this study is that communitfy setter should regularly identify and analyze rules or practices within the community that might be potential barrier for the members in building engagement with the community.

Abstract

This study aimed to determine the activity of polysaccharide krestin (PSK) from Coriolus versicolor extract to the dura-tion of estrous cycle and estradiol levels. PSK was administrated for 24 days in female Mus musculus, strain Balb/C, 10 weeks old, weight about 25-30 grams. Polysaccharide krestin given in sub-chronic dose. There were 4 groups, i.e K0 (0 mg/kg BW), P1 (15 mg/kg BW), P2 (30 mg/kg BW), and P3 (60 mg/kg BW). Each treatment group contained 7 replica-tions. Data were analyzed using One Way ANOVA and Duncan test at α = 5%. The duration of estrous cycle was de-termined by vaginal smear methods. The estradiol levels were measured using ELISA kit. The results show that the administration of PSK decreased the duration of estrous cycle with an average of 4.4 days. Polysaccharide krestin dose of 15 and 60 mg/kg BW did not affect the estradiol levels. Decrease of estrous cycle duration was still in the range of normal estrous time between 4-6 days. PSK dose of 15 and 60 mg/kg BW can be used as a therapeutic dose.
