Abstract

The hyphomycete, Veronaeopsis simplex, belongs to the group of so-called dark septate endophytes (DSE) colonize root plant intra and intercellulary without causing harm for the plant host. Knowledge about fungus interaction with the host plant is little. This paper describes the influence of different cultural conditions on protoplast isolation and regeneration from V. simplex fungus. As protoplast is interesting structure that can be used on developing an engineered DSE fungus by mutagenesis or genetic transformation, this research will provide the foundation for further study in order to reveal the unknown mechanism of the fungal role in the plant host. The mycelia age, osmotic stabilizer and lytic enzyme were the main factor that affected the protoplast formation.  The maximum yield of protoplast was 3.2 x 106/g fresh mycelia was obtained from 1 week old mycelia digested with the mixed of 1.5% cellulose, 1 % macerozyme and 1.5 % lysozyme for 3 h incubation at 37oC in the dark. Morphology and structure of protoplast released were observed using microscope. It was found that the regeneration of protoplast was possible after being cultivated on both CMMY and PDA medium supplemented with 0.6 M sucrose.Keywords: DSE, isolation, regeneration, protoplast, Veronaeopsis simplex

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui pengaruh penerapan model pembelajaran berbasis proyek berorientasi bioentrepreneurship terhadap kreativitas dan nilai-nilai kewirausahaan siswa kelas XII SMA pada sub topik bioteknologi konvensional. Penelitian ini menggunakan metode kuasi eksperimen dengan desain penelitian The posttest only control group design. Sampel penelitian yaitu kelas XII IPA 2 sebagai kelas eksperimen dan kelas XII IPA 1 sebagai kelas kontrol yang dipilih secara random sampling. Pengumpulan data menggunakan instrumen tes uraian kemampuan berpikir kreatif, rubrik penilaian produk kreatif, lembar observasi dan angket nilai-nilai kewirausahaan. Hasil analisis hipotesis menggunakan uji-t didapatkan signifikansi kreativitas dari produk kreatif sebesar 0,000 < 0,05 dan dari kemampuan berpikir kreatif sebesar 0,001 < 0,05, sehingga dapat disimpulkan bahwa model pembelajaran berbasis proyek berorientasi bioentrepreneurship berpengaruh terhadap kreativitas siswa. Signifikansi nilai-nilai kewirausahaan dari lembar observasi sebesar 0,000 < 0,05 dan dari angket sebesar 0,019 < 0,05, sehingga dapat disimpulkan bahwa model pembelajaran berbasis proyek berorientasi bioentrepreneurship berpengaruh pada nilai-nilai kewirausahaan. Hasil analisis korelasi disimpulkan bahwa terdapat hubungan yang sangat rendah diantara keduanya, dengan nilai koefisien korelasi sebesar 0,195. Hasil uji regresi linier menunjukan bahwa kreativitas memberikan kontribusi sebesar 3% terhadap nilai-nilai kewirausahan siswa.ABSTRACTEntrepreneurship have long been associated with creativity and need to be developed among students. A quasi experimental method with the posttest only control group design was conducted to determining the influence of project-based learning model oriented to bioentrepreneurship for XII grade students creativity and entrepreneurial values on conventional biotechnology sub topic. The sample was XII science 2 as experimental class and XII science 1 as control class chosen by random sampling. The data collected by using creative thinking ability test, creative product valuation rubric, observation sheet and entrepreneurial values questionnaire. The hypothesis analyzed by using t-test which obtained the significance of creative products was 0,000 < 0,05 and the creative thinking ability was 0,001 < 0,05, so it can be concluded that project-based learning model oriented to bio entrepreneurship effected on student creativity. As for the significance of entrepreneurial values from the observation sheet was 0,000 < 0,05 and the questionnaire was 0,019 < 0,05, so it would be concluded that project-based learning model with bioentrepreneurship oriented effected on students entrepreneurial values. Correlation analysis concluded that there was a correlation between them, with the value of correlation coefficient was 0,195. The result of linear regression test showed that creativity contributed about 3% to students entrepreneurial values.

Abstract

Inhabitants of the Kendeng mountain region depend on the biodiversity of the forest resources for their livelihood. Mushrooms are important resource that provide benefits to mankind. The aim on this research was to document traditional knowledge in bracket fungi utilization to treat health problems in Baduy tribe community. The results described in this paper were obtained through the exploration and identification of bracket fungi as well as the interview to determine their cultural significance for Baduy people. Quantitative approaches were used to determine the Use Values (UV), Informant Consensus Factor (ICF) and Fidelity level (FL) values. A total of 6 species that were identified as bracket fungi were included to Basidiomycota. The Baduy community considers the main use of bracket fungi as a medicine for several ailments. The highest UV, ICF and FL value were obtained by Ganoderma lucidum. This study provides the information regarding bracket fungi species with the high use values that could be employed in pharmacological research and further biotechnological approaches in order to achieve an adequate revenue.

Abstract

The existence of contextual learning resources has been known to be able to improve the meaningful learning, particularly biology learning. This research provides the information about the medicinal plant utilized by the Baduy tribe as well as its endeavour in developing the results obtained as a contextual-based module. This research and development was designed using Thiagarajans’ model, which was limited in three phases (define, design, and development). The data was collected using interview guideline, validation sheets, and students' response questionnaires. The research findings inform that there were 67 species utilized by the Baduy tribe as the medicinal plants. These species belong to Spermatophyte and divided into 31 families. Furthermore, the validation results showed that the media developed was valid; in which the best component of module, based on students' response, was its presentation (the mean value was 89.5%). It can be concluded that the media developed is feasible to use as a learning resource.

Abstract

AbstrakCendawan endofit akar bersepta gelap,Veronaeopsis simplex (CBS 588.66) pertama kali diisolasi dari serasah daun Acacia karoo di Afrika Selatan. Dua isolat Jepang (Y34 dan IBA K45) dewasa ini juga telah berhasil diisolasi dari pulau Yakushima dan Ibaraki. Hasil identifikasi menunjukkan kedua isolat ini memiliki posisi taksonomi yang sama dengan V. simplex. Hasil analisis filogenetik secara molekular menggunakan fragmen ITS 1, 5.8S rDNA dan ITS 2 gene region menunjukkan hubungan kekerabatan yang sangat dekat. Karakter morfologi dan faktor lingkungan yang mempengaruhi pertumbuhan cendawan seperti temperature dan pH berhasil diamati. Dua isolat Jepang dapat tumbuh pada suhu 37°C, namun isolat Afrika Selatan tidak dapat tumbuh pada suhu tersebut. Ketiga isolat dapat tumbuh dengan baik tanpa adanya perbedaan pada perlakuan nitrat dan asam amino Phe, Val, Leu. Cendawan DSE tersebar luas pada rizosfer dan rizoplain tanaman dan salah satu manfaat dari kolonisasi DSE ialah penguraian secara enzimatik sumber nutrisi organik di tanah yang akan ditransfer ke tanaman meningkatkan pertumbuhan tanaman tersebut. Dari ketiga isolat yang diuji, tidak satupun isolat yang menunjukkan kemampuan mendegradasi selulose, lignin, atau pectin, sedangkan lipase, gelatin, amylase, asam tanat dan asam galik menunjukkan respon yang positif

Abstract

The rhizosphere is a part of the soil located around the plant roots. The community of microorganisms in the rhizosphere is more diverse than the non-rhizosphere soil. Fungi are the most common microorganisms found in the rhizosphere and could influence soil fertility and increase plant resistance against pathogen and environmental stress. This research aimed to study the community of fungi in the rhizosphere of local rice varieties from Banten. Samples were collected from the rhizosphere of eight local rice varieties of Banten, i.e. padi Kewal Bulu Hideung, padi Limar, padi Guling, padi Siang, padi Cokrom, ketan Langgasari, ketan Ruyung dan ketan Cae. Fungi were isolated using the serial dilution method and plated into Potato Dextrose Agar medium. The fungal isolates recovered from the medium were purified using the same medium. Fungal isolates were identified based on their morphological characters. In total, 35 isolates were recovered belonging to seven genera, namely Aspergillus, Chaetomium, Fusarium, Gongronella, Mucor, Penicillium, and Trichoderma. The most abundant fungal community was found in the rhizosphere of padi Guling and padi Ruyung. This research gives insight into the diversity of fungal rhizosphere of local rice from Banten, which might have a role in their resistance against pathogens. Testing these isolates against the particular pathogen of rice could be done in the future to see the potentials for biological control used in rice.

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui konsentrasi limbah cair pencucian ikan yang tepat sebagai pupuk organik bagi pertumbuhan tanaman cabai rawit. Penelitian ini merupakan penelitian eksperimen dengan 6 taraf konsentrasi limbah cair dan 3 ulangan dengan menggunakan rancangan acak lengkap (RAL). Taraf konsentrasi pemupukan yang digunakan yaitu 0% (kontrol), 3,5%, 4%, 4,5%, 5%, 5,5% dan 6%. Parameter pertumbuhan cabai rawit yang diukur adalah tinggi batang, jumlah helai daun dan jumlah bunga. Data hasil penelitian dianalisis menggunakan Analisis of Varians (ANOVA) dengan uji lanjutan menggunakan Duncan Multipe Range Test (DMRT). Hasil penelitian menunjukkan bahwa pemberian pupuk organik limbah pencucian ikan berpengaruh nyata terhadap pertumbuhan tanaman cabai rawit dengan konsentrasi 5%. ABSTRACTThe objective of this study was to determine the fish processing waste water used as organic fertilizer on the growth of pepper plant. This study was an experimental study with 6 levels concentration of waste water and 3 replications using a completely randomized design. The fertilization concentrations levels used in were 0% (control), 3%, 4%, 4,5%, 5%, 5,5%, and 6%. The growth of papper plant was observed through steam height, number of leaves and number of flowers. The data from the study were analyzed using Analysis of Variance (ANOVA), and followed by Duncan’s Multipe Range Test (DMRT). The results showed that organic fertilizer from the fish processing waste water significantly affected the growth of pepper plants with a concentration of 5%.

Abstract

Plant Growth Promoting Bacteria Rhizobacteria (PGPR) is one of the potential bacteria to enhance of Capsicum annuum through inhabitation the growth of pathogenic fungi. This study aimed to characterize PGPR in chili plants (Capsicum annuum). PGPR was isolated from the soil habitat of the red chili plant in Cilegon, Indonesia. Screening was then carried out with the dual culture method on Petri dishes and tested through in vivo method on the red chili plant. The selected bacteria were characterized morphologically, biochemically, and physiologically. The results revealed that there were 14 single isolates of bacteria from the roots of the red chili plants. The five single bacterial isolates, namely Azostobacter, Azospirillum, Pseudomonas, Serratia, and Beijerinckia have good potential as PGPR based on multiple culture screening by producing clear zones and positively effect the growth of chili plants.

Abstract

Research results in the form of products and processes can be used as learning resources that can develop appropriate scientific abilities according to student needs. This study aims to analyze the results of research on the effect of heavy metal cadmium on the growth of tiger shrimp which is used as a source of learning biology. The research was conducted at the Department of Biology Education, FKIP Untirta and several state high schools in Banten Provice: SMAN 1 Anyer, SMAN 1 Cinangka, SMAN 4 Cilegon, and SMAN 5 Cilegon. The analysis process is carried out through stages: product identification and research processes, selection and modification of research results, as well as the application and development of research results as a source of biology learning contained in the Lesson Plan (RPP). The results of the study on the effect of heavy metal Cd on the growth of tiger prawns were in accordance with the sub-concept of environmental pollution contained in KD 3.11 and 4.11 in class X SMA. Environmental pollution that occurs can inhibit the growth of biota so that it can damage and disrupt the natural balance caused by contamination from heavy metal cadmium which is the subject of the learning process.
