Abstract

In this study, cellulase was produced by endophytic fungus isolated from seagrass. Substrate used for incubating the fungus was the waste of agar industry. The objectives of this study were to optimize cellulase production and to characterize the highest activity of fungal crude cellulase. In our previous study, the EN isolate (isolated from Enhalus sp.) showed the highest cellulolytic index. Therefore, in this research we focused on cellulase activity of the isolate. Cellulase activity was determined based on endoglucanase activity, total cellulase activity, and β-glucosidase activity. The highest activity was then used to determine cellulase activity in enzyme characterization. The fungus was cultured in different concentration of agar extraction algal wastes. The fungal culture was incubated for 3-21 days with 120 rpm orbital shaker. The results showed that endoglucanase activity was 0.019-0.031 U/mL, total cellulase activity was 0.007-0.013 U/mL, and β-glucosidase activity was 0.00012-0.00361 U/mL. The highest endoglucanase and total cellulase activity were obtained from the fungal culture after 9 days incubation, β-glucosidase was obtained from the fungal culture after 15 days incubation with 1.5% of algal waste as substrate. The optimum pH and temperature were determined as 4 and 60 oC, maximum reaction rate (Vmax) and Michaelis-Menten constant (Km) for endoglucanase activity was determined as 0.044 U/mL and 0.103% respectively. Abstract

Cellulolytic fungi are potentially useful in biotechnology. Decomposition of substrates containing cellulose can be applied in bioethanol and enzymes production. Fungi are one group of microorganisms that can decompose cellulase materials.  This study aimed to obtain potential cellulolytic fungi isolated from marine habitat. Screening of the fungi was performed using CBM media with different salinity. Six isolates were tested quantitatively for cellulolytic activity using congo red staining. Cellulolytic index of algicolous and manglicolous fungi were lower than those of EN isolate, Veronea sp. KT19, and sponge associate (SMH). Fungi cultured in freshwater and seawater media possessed higher cellulolytic index in compared to those of cultures in 3% NaCl medium. However, freshwater and seawater media showed no significant effect. Cellulolytic index of EN isolate, Veronaea sp. KT19, and SMH after six days of incubation were 1.357; 1.267, and 1.278, respectively. All six isolates potentially produced cellulase in all medium tested. Abstract

ABSTRAKPenelitian ini bertujuan untuk menentukan waktu optimum produksi enzim protease dan amilase isolat bakteri asal terasi ikan teri Stolephorus sp. Penelitian ini dilakukan dalam dua tahap yaitu pengukuran pertumbuhan isolat bakteri dan penentuan waktu optimum produksi enzim protease dan amilase. Pengujian yang dilakukan meliputi uji aktivitas enzim protease dan amilase dan  kadar protein dari tiap-tiap enzim tersebut. Data hasil pengujian dianalisis secara deskriptif. Ada 4 isolat bakteri, 2 isolat merupakan bakteri penghasil protease yaitu isolat P2 dan P4, dan 2 isolat yang merupakan bakteri penghasil amilase yaitu A2 dan A4. Aktivitas protease optimum terjadi pada jam ke-36 untuk isolat P2 sebesar 0,073 U/mL dengan aktivitas spesifik sebesar 1,632 U/mg dan isolat P4 yaitu 0,057 U/mL dengan aktivitas spesifik 4,91U/mg. Aktivitas amilase optimum terjadi pada jam ke-36 untuk isolat A2 sebesar 0,360 U/mL dengan aktivitas spesifik sebesar 7,73 U/mg dan aktivitas amilase optimum pada isolat A4 sebesar 0,239 U/mL dengan aktivitas spesifik 5,24 U/mg. ABSTRACTThe purpose of this research was to know the optimization of incubation time in the production of protease and amylase by bacterial isolates of anchovy Stolephorus sp. paste origin. This research was conducted in two stages, namely the growth of bacterial isolates measurement and determination the optimum time production of the enzyme protease and amylase. Testing conducted include the test proteases and amylase enzyme activity and protein levels of each of these enzymes. The data will be analyzed test results are descriptive. There are 4 bacterial isolates, where 2 isolates, which is a protease-producing bacteria isolates that is P2 and P4, and 2 isolates, which is the amylase-producing bacteria that is A2 and A4. The activity of the protease optimum occurs at to-36 hours to isolate P2 of 0.073 U/mL with a specific activity of 1.632 U/mg and isolates P4 that is 0.057 U/mL with a specific activity of 4.91 U/mg. Whereas amylase activity, optimum occurs at to-36ohours for A2 isolates of 0.360 U/mL with a specific activity of 7.73 U/mg and activity of amylase optimum on A4 isolates of 0.239 U/mL with a specific activity of  5.224 U/mg.

Abstract

Eeltail catfish (Paraplotosus albilabris) is a type of marine fish that has high nutritional content but is still rarely used further. In this study, we examined the proximate content and types of amino acids found in the flesh and skin of eeltail fish (Paraplotosus albilabris). The method used in this research is measurment of morphometric, proximate content and amino acids contained in flesh and skin of eeltail catfish The result of this study showed flesh contained 16.98% protein, 1.36% ash, 79.95% water, 1.12% fat, and 0.59% carbohydrates, respectively. On the skin, there was 33.49% protein, 1.41% ash, 64.26% water, 0.36% fat, and 0.48% carbohydrates. Meat had an essential amino acid yield of 84,222.14 mg/kg (52.97%) and skin had 76,429 mg/kg (30.45%), The amount of non-essential amino acids was 74,795.7 mg/kg (47.03%), and the amount of skin was 174.669 mg/kg (69.57%).

Abstract

Most fish processing industries only use fish meat as raw material for industry. Whereas fish bones are contain high mineral content needed by the human body and can be utilized as high calcium fishbone meal. Fishbone meal in this reaseach was made from waste from the fishery industries in Tanjungpinang, Riau Islands. Mackerel fishbone meal was made in several steps consist of preparation of raw materials, boiling of fishbones, washing and cleaning of the bones, presto processing of fish bones with the addition of sodium bicarbonate (NaHCO3), drying, siege and sieving. The purpose of this study is to determine the effect of presto duration and concentration of NaHCO3. This research was conducted by the factorial randomized block design method (RAKF) with two factors: duration of presto (2 hours (P1) and 4 hours (P2)) and concentration of NaHCO3 (0% (N1), 1% (N2) and 2% (N3)). The results showed that the duration of presto treatment and the concentration of sodium bicarbonate (NaHCO3) had a significant effect on the 5% level of yield but not significant on calsium content. The best results showed on 2 hours presto with addition NaHCO3 1%. The best treatment was used as a sample to measure the chemical characteristics of mackerel fish bone meal. Fish bone meal obtained 4.86% water content, 69.84% ash content, 3.72% protein content, 17.21% fat content, 12.56% phosphorus content, and 0.46% crude fiber. The chemical content of fishbone meal has been appropriate with Indonesian Nasional Standard.

Abstract

Abstract. The richness of biological diversity is an asset for the development and prosperity of the nation, but human needs and environmental pressure increased, especially marine biological resources, it has resulted in a decline in the population of several marine life. This causes several marine life to be rare and endangered. Conservation efforts need to be carried out covering aspects of preservation, protection, and utilization. Another preventive way that can be done is providing education to the community, especially in the elementary school young generation. Given that education related to the preservation of marine biodiversity is important to learn as early as possible, elementary school young generation are the main target for socialization about protected marine life to create a sense of care, willingness, and knowledge so that marine wealth can be preserved. Introduction of protected marine life in the Riau Islands region with participants of grade VI students at the State/Public Elementary School (SDN) 06 Gunung Kijang, Bintan Regency. The activity is carried out in several stages, namely pre-test knowledge of protected marine life, education of protected marine life in the Riau Islands region (video and pocketbook containing short stories and information on protected marine life in Riau Islands), and post-test to know enhancement of student knowledge of protected marine life. The level of understanding increased from a range of poor-moderate understanding by pre-test correct answers (47% -79%) to a moderate-high range of understanding by post-test correct answers (74% -100%).             Abstrak. Kekayaan keanekaragaman hayati menjadi aset bagi pembangunan dan kemakmuran bangsa, namun dengan meningkatnya kebutuhan manusia dan tekanan terhadap lingkungan khususnya sumberdaya hayati laut, mengakibatkan terjadinya penurunan populasi beberapa biota perairan. Hal ini menyebabkan beberapa biota perairan menjadi langka dan terancam punah. Upaya konservasi perlu dilakukan yang meliputi aspek pelestarian, perlindungan, dan pemanfaatan. Cara preventif lain yang dapat dilakukan adalah pemberian edukasi kepada masyarakat khususnya anak-anak sekolah. Mengingat pendidikan terkait kelestarian keanekaragaman hayati laut penting untuk dipelajari sedini mungkin, maka anak-anak sekolah dasar menjadi sasaran yang tepat untuk dilakukan sosialisasi mengenai biota-biota laut yang dilindungi agar terwujudnya rasa peduli, kemauan, serta ilmu pengetahuan sehingga kekayaan laut dapat terjaga. Pengenalan biota laut dilindungi di wilayah Kepulauan Riau dengan peserta siswa-siswi kelas VI di Sekolah Dasar Negeri (SDN) 006 Gunung Kijang, Kabupaten Bintan. Kegiatan dilaksanakan dalam beberapa tahap, yaitu pre test pengetahuan tentang biota laut yang dilindungi, edukasi biota laut yang dilindungi di wilayah Kepulauan Riau (media video dan buku saku yang berisi cerita pendek serta keterangan biota laut dilindungi yang ada di Kepulauan Riau), dan post test pengetahuan tentang biota laut yang dilindungi. Tingkat pemahaman meningkat dari rentang pemahaman yang kurang sampai sedang dilihat dari persentase jawaban benar pre-test sebesar 47%-79% menjadi rentang pemahaman sedang sampai tinggi dengan hasil jawaban benar post-test sebesar 74%-100%.

Abstract

Konsumen dan industri perikanan membutuhkan ikan segar pada waktu yang tepat, harga yang tepat dan ditempat yang tepat. Pengabdian kepada masyarakat ini dilakukan untuk memberikan rekomendasi kepada pemerintahan terkait, pemahaman kepada aktor distribusi hasil perikanan dan pentingnya menjaga kesegaran selama transportasi ikan. Rantai distribusi ikan yang terdapat di tanjungpinang meliputi Nelayan, Pengumpul, Distributor, Pengolah ikan dan Konsumen. Masing-masing rantai distribusi memiliki peranan dalam mengatur arus barang, infromasi dan keuangan. Hasil pengabdian kepada masyarakat memberikan rekomendasi terkait evaluasi pola distribusi sehingga mampu menjaga harga dan kesegaran ikan. Aktor-aktor distribusi diberikan pemahaman tentang cara menangani ikan yang baik terutama dalam hal waktu dan suhu yang digunakan selama transportasi

Abstract

Sea cucumbers which have the scientific name Holothuroidea are fishery commodities that have high economic value and are in great demand in the global market. Sea cucumbers are the main commodity of Benan Village which is the outermost island of Lingga district. Most of the people in Benan Village work as sea cucumber fishermen. As an aquatic resource owned by the village, sea cucumbers are still constrained in terms of handling so that it reduces the selling value of the dried sea cucumbers produced. Therefore, it is important to empower the people of Benan Village regarding the postharvest handling of sea cucumbers which aims to improve the quality as well as directly increase the selling value of that resources. Community empowerment activities are expected to add insight and improve the ability of the Benan Village community in handling sea cucumbers post-harvest. Empowerment was carried out through three methods, namely the distribution of questionnaires and discussions in the form of questions and answers with the Benan Village community group regarding the problem process in handling post-harvest sea cucumbers, which was carried out after distributing questionnaires to the sea cucumber aquaculture community, then education and discussion on good procedures were carried out in post-harvest handling. To improve the quality of this sea cucumber commodity, then in the third stage, mentoring and practice regarding the correct handling of sea cucumbers is carried out to produce good quality sea cucumbers from washing to packaging. The selection of discussion methods, lectures/education, and direct practice is considered effective in increasing public knowledge about the process of handling sea cucumbers postharvest properly and correctly, considering that the target stakeholders are sea cucumber farmers.

Abstract

The Anambas Islands have many unique traditional products derived from marine products. One of the famous traditional products is smoked tuna. This study aims to determine the chemical characteristics of smoked tuna processed by smoked fish craftsmen on Jemaja Island, Anambas Islands Regency. This research was divided into three stages, identifying the type of raw materials used, sensory analysis, and proximate analysis. The raw material for smoked tuna is known to come from two types of fish, namely Auxis thazard and Thunnus tonggol. The sensory analysis results with the parameters of appearance, aroma, taste, and texture of smoked tuna from the two raw materials were not significantly different for all parameters. These results are the same as the proximate analysis values which consist of testing the water, ash, fat, protein, and carbohydrate content. Based on the overall results of smoked tuna made from Auxis thazard and Thunnus tonggol it is still by the applicable SNI 2725:2013, except moisture content.

Abstract

Resistensi antibiotik merupakan masalah kesehatan yang dialami oleh makhluk hidup, disebabkan oleh penggunaan antibiotik jangka panjang. Thallasia hemprichii memiliki kandungan senyawa metabolit yang berpotensi pada bidang farmasi. Banyak penelitian menunjukkan bahwa lamun memiliki aktivitas senyawa bioaktif alami sebagai antifouling, antibakteri, dan antikapang. Tujuan penelitian ini yaitu untuk mengetahui jumlah isolat, dan karakteristik morfologi kapang endofit serta mengetahui aktivitas antagonis yang dihasilkannya terhadap bakteri uji Staphylococcus aureus dan Escherichia coli. Penelitian dimulai bulan April-September 2021 menggunakan perairan Desa Malang Rapat. Jenis penelitian yang dilakukan yaitu deskriptif. Terdapat 4 tahap penelitian yaitu pengambilan dan preparasi akar serta daun lamun sebagai inang kapang endofit, isolasi kapang endofit lamun dengan metode isolasi tanam langsung, pengamatan morfologi kapang, dan uji antagonis kapang menggunakan metode uji antagonis oleh kapang terhadap bakteri uji. Hasil penelitian mendapatkan 3 isolat dari sampel akar dan 7 isolat asal daun, dengan isolat yang memiliki hifa septum dimiliki oleh D32.1, D32.2, A37, A35, A21, dan D45. Karakteristik pigmentasi putih dimiliki oleh isolat D32.2, D31, D21, D22, D18, dan A37. Uji antagonis terhadap bakteri E. coli dan S. aureus menunjukkan bahwa isolat D18 memiliki diameter zona hambat yang paling besar yaitu 27,7 mm, dan isolat D32.1 menunjukkan diameter zona hambat yang paling kecil yaitu 13,1 mm terhadap bakteri E. coli. Isolat D22 terhadap bakteri S. aureus memperlihatkan diameter zona hambat sebesar 5,9 mm.
