Abstract

Transforming growth factor-β (TGF-β) is a two-chain polypeptide with molecular weight of 25 kDa which takes significant role in the steroidogenesis process. In the ovarian oocyte in particular, TGF-β has an important role in regulating reproductive function. TGF-β represents a key intrafollicular protein that regulates follicle development and aromatization process. The purpose of this research was to characterize and identify a protein fraction of TGF-β from the bovine isolated oocytes, which is synthesized during in vitro oocyte maturation process. Oocytes were collected from follicles with diameter of 3-8 mm. Oocytes were then matured in TCM 199 media supplemented with 5 μg/mg LH, 3% BSA, and 50 μg/ml gentamicin sulfate, and cultured in CO2 incubator (5%, 38.5 oC) for 20 hours. TGF-β receptors were identified immunohistochemically. Characteristics of the TGF-β protein were determined using SDS PAGE and TGF-β specification was tested using Western Blotting. The results showed that TGF-β receptors were identified and found in cumulus oocyte complexes (COCs). TGF-β protein was isolated from bovine oocytes with molecular weight 25 kDa and it was identified by Western blotting methods in the same molecular weight.

Abstract

Background: Intelligence management can produce superior human resources with integrated brain. Mozart music stimulus during pregnancy has been shown to increase the number of neuron of the fetal brain. The study of pop and religious music needs to be improve because they are more popular in Indonesia. Objective: To analyze the differences of number of Neuron in the Cerebrum Rattus norvegicusoffspring that exposed to Mozart, Indonesian pop music and Indonesian religious music during pregnancy. Methods: An experimental study with a post-test only control group design. Groups divide into treatment music groups: Mozart, pop and religious. Treatment in a soundproof room for 1 hour, starting the 10th-day of pregnancy, intensity of 65 dB with a distance of 25 cm from the cage. The number of neuron was counted from HE brain preparations of the head Rattus norvegicusoffspring and analyzed using appropriate statistics test. Results: There were significant differences in the number of neuron of Rattus norvegicusoffspring in cerebrum between groups with p = 0,000 (mean Mozart music group 28.14 ± 3.02, Indonesian pop music 19.71 ± 1.80, Indonesian religious music 24.14 ± 2.91) and Mozart gave a higher number of neuron than Indonesia religious music and Indonesian pop music. Conclusion: Mozart music gave a higher number of neuron in the Cerebrum than Indonesian religious music and Indonesian pop music. Keywords: neuron; cerebrum; Mozart music; Indonesian music ABSTRAK Latar belakang: Pengelolaan kecerdasan otak yang terintegrasi akan menghasilkan SDM yang unggul. Stimulus musik Mozart selama kehamilan terbukti meningkatkan jumlah sel neuron di otak janin. Musik pop dan religi perlu dilakukan penelitian karena lebih populer di Indonesia. Tujuan: Menganalisis perbedaan jumlah sel neuron di Cerebrum Rattus norvegicusbaru lahir antara yang mendapat paparan musik Mozart, musik pop Indonesia dan musik religi Indonesia selama kebuntingan. Metode: Studi eksperimental dengan desain post test only control group. Kelompok perlakuan dibagi menjadi kelompok musik Mozart, musik pop Indonesia dan musik religi Indonesia. Perlakuan di ruang kedap suara selama 1 jam pada malam hari mulai hari ke-10 kebuntingan, intensitas 65 dB dengan jarak 25 cm antara kandang dan speaker. Jumlah sel neuron dihitung dari preparat pewarnaan Hematoxylin-Eosin otak anak Rattus norvegicusdan dianalisis dengan statistik yang sesuai. Hasil: Terdapat perbedaan yang signifikan pada jumlah sel neuron cerebrum Rattus norvegicus baru lahir antar kelompok dengan nilai p=0,000 (rerata kelompok musik Mozart 28,14±3,02, musik pop Indonesia 19,71±1,80 dan musik religi Indonesia 24,14±2,91) dan musik Mozart memiliki jumlah sel neuron lebih tinggi daripada musik religi Indonesia dan musik pop Indonesia. Kesimpulan: Kelompok musik Mozart memiliki jumlah sel neuron di cerebrum yang lebih tinggi dibandingkan kelompok musik Indonesia. Kata kunci: neuron; cerebrum; musik Mozart; musik Indonesia

Abstract

Background: Brain growth requires genetic potential for a conducive environment, low stress levels, stimulation and nutrition. Brain development during the fetal period in the two years of life firstly requires proper stimulation to increase intelligence. Classical music stimulation has been proven to be able to optimally improve brain function and human intellect. Other music that Indonesian people like is pop and religion. Maybe it is possible to pop and religious music can affect the development and growth of the fetal brain. Objective: Analyze the differences of the index of apoptosis Cerebrum and Cerebellum Rattus norvegicus newborn between those stimulated by Mozart, pop, religious music and not stimulated by music during pregnancy. Method: This study was post test only control group design. groups were randomly divided according to the treatment as stimulation of Mozart, pop, religious music and no music was given from the 10th day-gestation with an intensity of 65 dB in a soundproof box for one hour. Abstract

Moringa pterygosperma is one of the plants in Indonesia that is often used in herbal medicine. Moringa leaves contain many phytochemical compounds, one of the compounds is flavonoids which function as antioxidants. Flavonoid compounds in vitro can inhibit lipid peroxidation by breaking the chain of peroxyl radicals. Moringa leaf benefits in the field of herbal medicine are formed with a variety of preparations. One of the preparations is in the form of extracts with the aim of attracting chemical components to natural materials The extraction process has a disadvantage because the level of solubility in water is still low. The nanoparticle technique is a strategy to increase the bioavalbicibility of herbal active compounds. This research was experimental in order to determine the total flavonoid levels contained in Moringa leaf extract. In addition, it was also to find out the antioxidant activity and particle size of the moringa leaf extract nanoparticles. Determination of flavonoid levels using spectrophotometric methods. The antioxidant activity of moringa leaf extract nanoparticles was tested using the DPPH method (2,2-diphenyl-picrylhdrazyl) and to determine the particle size used by using the Particle Size Analyzer (PSA). The results of the study showed total flavonoid levels in moringa leaf extract (1.97 ± 1.07)%. Antioxidant activity of 451.8 ppm with a reduction of 2.8% and particle size in the moringa leaf extract nanoparticles of ± 134.78 nm. Moringa leaf extract nanoparticles include low antioxidants. Abstract

Classical music stimulation is a proven method that affect intelligence as well as brain function. Mozart’s classical music is classical music that could stimulate the growth of synaptic networks. To analyze the difference of synapsin expression of the brain of Rattus norvegicus  offspring on Mozart’s, pop, religious and without music stimulation during pregnancy. Experimental. Treatments were subjected to Rattus norvegicusas a subject organism, initiated on the 10th day of pregnancy, in 1 hour duration in a soundproof chamber, intensity of 65 dB. On the 19th day of pregnancy the subjects were sacrificed and three offsprings were chosen. The head of the chosen Rattus norvegicusoffsprings were decapitated and the brain subsequently sectioned to calculate synapsin expression by immunohistochemistry method with 400 magnification strength microscope on the 5 field of view. Significant difference of synapsin expression of the brain of Rattus norvegicus offspring was observed between Mozart’s and control group. Significant differences of synapsin expressions were observed in the cerebellum of the newly born Rattus novergicus between Mozart and pop music groups (p = 0,007), Mozart and religious music groups (p = 0,008), Mozart and without music groups (p = 0,004).Synapsin expression of Rattus norvegicus brain offsprings were higher in Mozart music stimulation compared with pop and religious music stimulation

Abstract

Background: Endometriosis is a gynaecological disorder characterised by the presence of endometrial tissue outside the uterine cavity. Apoptosis, regulated by the balance of Bcl-2/Bax, plays an important role in the endometrial improvement. Metformin, an insulin sensitizer that is known to have beneficial effect in the endometriosis treatment, is expected to lower Bcl-2/Bax expression ratio and reduce endometrial implants.Objective: To explore the effect of metformin on the Bcl-2/Bax expression ratio and endometrial implant area of endometriosis-induced mice.Methods: This experimental study used 3-month old 33 BALB/c mice of endometriosis that were randomly and equally divided into three groups (P0, P1, and P2). On the 15th day, the P0 group was first terminated for Bcl-2/Bax examination and the size of endometrial implants. The P1 group was given aquabidest, whereas the P2 group was given metformin 4 mg/day for 14 days. The immunohistochemistry of Bcl-2/Bax expression were performed from cavum abdomen and pelvis peritoneal tissues of the mice and measured by the Remmele Scale Index, whereas the extracted mice’ endometrial implants were analysed with computer tracing method. All data normality test was calculated with Shapiro-Wilk test. The mean difference test of all groups was analysed using the one-way ANOVA test, whereas the mean difference test between groups was completed using the Unpaired T-test (LSD/Least Significance Difference).Results: The Bcl-2/Bax expression ratio and endometrial implant area in the P2 group were significantly lower compared to P0 and P1 (p<0.001). There were no significant differences in the Bcl-2/Bax expression ratio or endometrial implant area in P0 and P1 (p>0.05)Conclusion: Metformin may be a potential effective drug treatment for endometriosis by decreasing Bcl-2/Bax expression ratio and endometrial implants.

Abstract

Ikan nila merah Jatimbulan dan ikan nila Larasati merupakan hasil rekayasa genetik dari dua lokasi yang berbeda. Ikan yang berasal dari lokasi yang berbeda mempunyai karakter yang berbeda karena adanya pengaruh interaksi antara genetik dan lingkungannya. Lingkungan merupakan salah satu faktor pembatas dari tiap populasi ikan dari segi morfometrik ataupun genetik. Perbedaan suhu, cahaya, nutrisi dan faktor lingkungan lain memiliki dampak perbedaan kromosom pada setiap strain ikan. Dalam upaya mendapatkan informasi mengenai hal tersebut, maka penelitian ini akan membahas bagaimana gambaran kromosom dari dua strain ikan nila merah yang dibudidayakan di lokasi yang berbeda. Penelitian ini bertujuan untuk mengetahui gambaran kromosom (jumlah kromosom, bentuk kromosom dan susunan kromosom) ikan nila merah Jatimbulan dan ikan nila Larasati. Hasil dari penelitian ini menunjukkan jumlah kromosom ikan nila merah Jatimbulan dan ikan Larasati adalah 44 pasang (2n=44). Bentuk dan susunan kromosom (karyotipe) ikan nila merah Jatimbulan adalah 4 pasang sub metasentrik, 9 pasang sub telosentrik dan 9 pasang telosentrik (4SM-9ST-9T), sedangkan kromosom dari ikan nila Larasati adalah 3 pasang sub metasentrik, 14 pasang sub telosentrik dan 5 pasang telosentrik (3SM- 14ST-5T).

Abstract

This study aims to evaluate the expression of BAX, BCL-2, and BAX/BCL-2 ratio in maturation media of cow oocytes which supplemented with Urea in vitro because BAX and BCL-2 are the main regulators of apoptosis. A total of 263 oocytes from follicle aspirations originating from ovaries taken from slaughterhouses and were saturated with 3 addition of urea which was divided into three groups. The control group (P0) was control group without the addition of urea, P1 group was added with urea 20 mg/dL, while P2 group was added with urea 40 mg/dL. The results of in vitro oocyte maturation were continued with identification using immunocytochemical staining with the addition of BAX and BCL-2 antibodies. Positive results showed a brownish color on the oocyte and its cumulus. The results of this study indicated that there were significant differences (P0.05) in BAX and BCL-2 expression, although both curves were equally increase. The increase in BCL-2 was more significant than BAX, while the BAX BCL-2 ratio did not show a significant difference (P0.05) in whichthe curve of BAX/BCL-2 ratio was decreased. It can be concluded that the addition of urea does not affect the level of apoptosis.

Abstract

Tujuan dari penelitian ini adalah mengidentifikasi suatu protein dengan berat molekul 46 kDa yang diduga sebagai epidermal growth factor (EGF) yang diisolasi dari oosit sapi yang telah dimaturasi secara in vitro dengan metode elektroforesis. Ovarium sapi yang berasal dari rumah potong hewan, diaspirasi pada folikel dengan diameter permukaan ≤5 mm menggunakan spuit dan jarum. Oosit dimaturasi dalam tissue culture medium (TCM) 199 selama 22 jam pada suhu 38,5° C di dalam inkubator CO2. Preparasi protein dengan berat molekul 46 kDa menggunakan sodium dodecyl sulphate polyacrilamide gel elektroforesis (SDS PAGE). Berdasarkan perhitungan jumlah regresi dari protein marker, didapatkan 12 fraksi protein yaitu BM 172,7; 153,09; 118,24; 102,43; 89,27; 59,75; 46,41; 43,82; 40,2; 36,06; 23,45; dan 18,42 kDa. Protein dengan berat molekul 46,41 kDa yang tampak pada pita protein dapat diidentifikasi sebagai protein yang diduga EGF yang berperan dalam proses maturasi oosit.
