Production Pattern of Ajmalicine in Catharanthus roseus (L.) G. Don. Cell Aggregates Culture in the Airlift Bioreactor RIZKITA RACHMI ESYANTI; AIDA MUSPIAH

Abstract

A research has been conducted to optimize the rate of aeration and initial weight of cell aggregates in the production of ajmalicine in Catharanthus roseus cell culture in airlift bioreactor. Catharanthus roseus culture were grown in Zenk medium with the addition of 2.50 x 10-6 M naphthalene acetic acid (NAA) and 10-5 M benzyl amino purine (BAP). Cell aggregates were sub-cultured two times before transferring 20 and 30 g/fw of cell aggregates into bioreactor, respectively, and aerated with the rate of 0.25 l min-1 and 0.34 l min-1, respectively. The pattern of ajmalicine production in bioreactor were observed in every three days within 24 days. Qualitative and quantitative analysis were conducted using HPLC connected to Cromatopac CL-7A Plus. The results showed that the cell aggregates and medium contain ajmalicine. The highest concentration was obtained in combination of 30 g/fw and 0.34 l min-1 aeration compare to 20 g/fw - 0.25 l min-1, 20 g/fw - 0.34 l min-1, as well as 30 g/fw – 0.25 l min-1/sup>. The highest ajmalicine content in cell aggregates was obtained on the 12 days (79.23 µg g-) whilst in medium was obtained in the 18thsup> days (981.15 µg l-). Abstract

Abstract

Pengolahan kedelai menjadi tahu menghasilkan limbah padat dan cair. Limbah cair tahu secara kimia merupakan bahan yang mempunyai komposisi nutrisi yang baik sehingga dapat didaur ulang untuk menghasilkan produk lain yang bermanfaat. Salah satu teknologi pemanfaatan limbah cair tahu menjadi produk yang lebih menghasilkan dan sekaligus bermanfaat untuk mengurangi tingkat cemaran terhadap lingkungan perairan, yaitu pembuatan nata de soya yang berbahan dasar limbah cair tahu (whey tofu) sebagai medium tumbuh bagi bakteri asam asetat pembentuk nata Gluconacetobacter xylinus. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk memberikan pemahaman kepada kelompok pengrajin tahu di Kelurahan Kekalik Jaya Kota Mataram mengenai dampak pembuangan sampah organik bagi lingkungan perairan. Selanjutnya kegiatan pengabdian ini juga bertujuan memberikan pemahaman dan keterampilan kepada pengrajin tahu mengenai teknologi pembuatan produk nata de soya dengan memanfaatkan limbah cair tahu (whey) sebagai medium dasar. Kegiatan ini terdiri dari 3 tahapan yaitu : penyuluhan mengenai dampak limbah pengolahan tahu terhadap lingkungan perairan dan kesehatan masyarakat, penyuluhan mengenai cara memanfaatkan limbah cair tahu untuk pembuatan nata de soya dan praktek pembuatan nata de soya dari limbah cair tahu. Sasaran kegiatan ini adalah para pengrajin tahu yang ada di Kelurahan Kekalik Jaya Kota Mataram. Kegiatan pengabdian ini telah terlaksana dengan baik dengan dihadiri oleh 25 orang perajin tahu sebagai peserta, selain dihadiri oleh lurah dan PPL yang membina kelompok perajin tahu di Kelurahan Kekalik Jaya. Peserta antusias mengikuti kegiatan sampai selesai dan menghendaki kegiatan ini ada tindak lanjutnya agar perserta betul-betul menguasai cara memproduksi nata de soya dari limbah cair pengolahan tahu.

Abstract

Gelangsar Village, West Lombok is one of the villages located in a hilly area which resulted in several points in the village of Gelangsar experiencing landslides. The topography which is hilly and steep, triggers landslides in the village of Gelangsar. In addition, the relatively high rainfall in the last few months has made the soil conditions more flexible. Areas that have the potential to cause landslides can be overcome by planting plants that can bind the soil tightly so that it can overcome the potential for landslides. Therefore, this service aims to socialize the use of vetiver grass as a bioengineering method in managing landslides in Gelangsar Village, Gunungsari, West Lombok. This Community Service activity uses the counseling method and the creation of a pilot project. Counseling was conducted to introduce vetiver grass plants and their cultivation techniques. Making a pilot project (pilot project) have been done in the homes of residents who are partners. The Pilot Project was created as a media for socialization and as a strategy for efficiency and effectiveness for target activities. The community of partners are willing to implement and develop vetiver grass as a biotechnology agent to prevent erosion and also as an economic opportunity. This programme has been able to provide knowledge in mitigation landslide disaster management activities independently. Planting vetiver grass is expected to be able to reduce the risk of erosion at landslide-prone points in the village of Gelangsar, improve the physical condition of the land and increase land productivity.

Abstract

Desa Aik Prapa Kecamatan Aikmel. Lombok Timur memiliki potensi alami yang mendukung pengembangan budidaya lebah madu, yaitu ketersediaan koloni lebah Tetragonula sp. di alam dan kelimpahan pakan lebah dari hutan di sekitar desa. Keterbatasan pengetahuan dan keterampilan menyebabkan masyarakat Desa Aik Prapa belum memanfaatkan potensi alam tersebut sebagai peluang usaha. Kegiatan ini bertujuan untuk memberikan pengetahuan dan keterampilan kepada masyarakat dalam budidaya lebah madu Tetragonula sp. Metode yang digunakan dalam kegiatan ini adalah ceramah dan diskusi, penyebaran leflet, dan pembentukan plot percontohan. Sosialisasi dilakukan untuk memperkenalkan potensi lebah madu Tetragonula sp. pakan alami dan teknik budidayanya. Leaflet yang dibagikan kepada masyarakat berisikan panduan praktis teknik budidaya lebah madu. Pada tahap akhir kegiatan dilakukan demonstrasi dan pembentukan plot percontohan. Plot percontohan dibuat di rumah salah satu peserta. Plot percontohan yang berhasil dapat mendorong masyarakat untuk menerapkan inovasi dalam budidaya lebah madu. Hasil kegiatan menunjukkan bahwa masyarakat desa Aik Prapa memiliki ketertarikan dalam budidaya lebah madu. Penerapan budidaya lebah madu di Desa Aik Prapa memerlukan pendampingan yang intensif agar dapat dapat memberikan manfaat bagi perekonomian masyarakat. Kemiskinan, keterbatasan waktu, keterbatasan jaringan, kurang adaptif, karakter dan cara berfikir yang pesimis, rendahnya semangat berusaha, inkonsisten, rendahnya tingkat kesehatan dan gizi masyarakat merupakan faktor yang menghambat berkembangnya kewirausahaan di pedesaan.

Abstract

The Meninting Sub-watershed area is one of the areas prone to landslides. Most of the Meninting Watershed Area is included in the Gunungsar District Area. Two villages in Gunungsari District, namely Kekait Village and Bukit Tinggi Village, are included in the most widespread level of landslide vulnerability. The aim of this community service activity is to provide information and data regarding landslide-prone areas, increase the knowledge of the community regarding the risk of landslides, and mitigation actions that can be taken. The method used is the focus group discussion (FGD) method. The FGD was carried out in two stages, namely socialization material and discussion. The socialization was carried out to convey data and information regarding villages in Gunungsari District which are included in the Medium to High landslide prone category. The discussion was carried out to encourage the active participation of the Gunungsari community in designing landslide disaster mitigation efforts. The results of the service showed that there was a positive response from the people of Gunungsari District as indicated by an agreement on a follow-up plan to utilize the results of recommendations for landslide disaster mitigation and adaptation efforts. Mitigation efforts are carried out using physical and non-physical efforts. Physical efforts that can be carried out include spatial planning in the Meninting Watershed Area, application of bioengineering techniques using vetiver grass on cliff edges. Non-physical efforts that can be made are increasing understanding and knowledge about landslide disasters. Adaptation efforts are carried out by recognizing early signs of landslides, preparing disaster emergency equipment, and avoiding settlements in areas near rivers or steep slopes

Abstract

Giri Madia Village, Lingsar District, West Lombok Regency is one of the villages prone to landslides. The results of the study showed that around 49.87 Ha of land is classified as a high landslide-prone area. Therefore, mitigation efforts were made to reduce the risk of landslides by utilizing vetiver grass (Chrysopogon zizanioides). This community service activity was carried out through collaboration between the community service team, the village government, and the Giri Madia Village community, from December 2023 to February 2024. The program implementation started from preparation (observation, finding partners, and licensing), implementation (provision of seeds and planting), and monitoring. Coordination with the village government and the local community was carried out through open discussions. The seedling planting activity was carried out by the community service team and the community in three predetermined locations. The monitoring results showed that almost all of the vetiver grass that was planted was able to survive even though it was not watered regularly by the community. Community activities, namely cutting down trees around the vetiver grass planting location, caused more than a third of the total seedlings to die from being hit. In addition, the obstacles that arise are the lack of community participation in grass care such as watering and fertilizing. This can be caused by the lack of public awareness to help care for the fragrant root grass seeds. The community feels that they do not have time to monitor because of their daily activities of earning a living.
